Project description:Mitochondria participate in various cellular processes including energy metabolism, apoptosis, autophagy, ROS production, stress responses, inflammation and immunity. We investigated the effects of immune tissue specific mitochondrial perturbations on immune responses at the organismal level. Drosophila melanogaster model was utilized to knockdown genes from oxidative phosphorylation (OXPHOS) complexes cI-V, by targeting the two main immune tissues of Drosophila, the fat body and the immune cells (hemocytes). While fat body targeted OXPHOS perturbations caused detrimental effects on the viability and immunity, hemocyte specific perturbations led to enhanced immunocompetence of the host. This was accompanied by the formation of melanized hemocyte aggregates (melanotic nodules), a sign of activated cell-mediated innate immunity. Furthermore, hemocyte specific OXPHOS perturbations caused proliferation and immune activation of the hemocytes, resulting to a similar hemocyte profile as seen upon infection, and the animals ultimately had an enhanced immune response when infected with a parasitoid. Our data shows that hemocyte targeted OXPHOS perturbations cause mitochondrial membrane depolarization and upregulation of genes associated with mitochondrial unfolded protein response, including aerobic glycolysis and reactive oxygen species. Overall, we show that while the effects of mitochondrial perturbations on immune responses are highly tissue specific, mild mitochondrial dysfunction can be beneficial in immune-challenged individuals and is causing variation in infection outcomes between individuals.

Exercise Activates Interferon Immune Response of the Liver via Gpld1 to Enhance Antiviral Innate Immunity

Project description:<p>Healthy behavioral patterns could modulate organ functions to enhance the body’s immunity. However, whether exercise regulates antiviral innate immunity remains elusive. Here, we found that exercise promotes type-I IFN (IFN-I) production in the liver and enhances IFN-I immune activity of the body. Despite the possibility that many exercise-induced factors could regulate IFN-I production, we identified Gpld1 as a crucial molecule and the liver as the major organ to promote IFN-I production after exercise. Exercise largely loses the efficiency to induce IFN-I in Gpld1-/- mice. Further studies demonstrated that exercise-produced 3-hydroxybutanoic acid (3-HB) critically induces Gpld1 expression in the liver. Gpld1 blocks the PP2A-IRF3 interaction and therefore enhances IRF3 activation and IFN-I production, and improves the body’s antiviral ability. This study reveals that the exercise behavior improves antiviral innate immunity by linking the liver metabolism to systemic IFN-I activity, and uncovers an unknown function of liver cells in innate immunity.</p>

Project description:The therapeutic success of in vitro-transcribed (IVT) mRNA depends on its stability and efficient translation. However, IVT mRNA is highly susceptible to immune-mediated degradation, limiting its efficacy. Here, we co-transfected IVT mCherry mRNA with immune suppressor M or SOCS1 mRNA in human umbilical vein endothelial cells (HUVECs) for 6h and 24h to explore whether hijacking innate immunity could enhance expression and half-life of IVT mRNA. Subsequently, RNA-seq was performed to investigate the transcriptomic changes in HUVECs after transfecting IVT mRNA. We sought to clarify the potential mechanism underlying degradation of IVT mRNA.

Mitochondrial Hyperactivity and Reactive Oxygen Species Drive Innate Immunity to the Yellow Fever Virus-17D Live-Attenuated Vaccine

Project description:The yellow fever virus 17D (YFV-17D) live attenuated vaccine is considered one of the successful vaccines ever generated associated with high antiviral immunity, yet the signaling mechanisms that drive the response in infected cells are not understood. Here, we provide a molecular understanding of how metabolic stress and innate immune responses are linked to drive type I IFN expression in response to YFV-17D infection. Comparison of YFV-17D replication with its parental virus, YFV-Asibi, and a related dengue virus revealed that IFN expression requires RIG-I-like Receptor signaling through MAVS, as expected. However, YFV-17D uniquely induces mitochondrial respiration and major metabolic perturbations, including hyperactivation of electron transport to fuel ATP synthase. Mitochondrial hyperactivity generates reactive oxygen species (ROS) including peroxynitrite, blocking of which abrogated MAVS oligomerization and IFN expression in non-immune cells without reducing YFV-17D replication. Scavenging ROS in YFV-17D-infected human dendritic cells increased cell viability yet globally prevented expression of IFN signaling pathways. Thus, adaptation of YFV-17D for high growth imparts mitochondrial hyperactivity to meet energy demands, resulting in generation of ROS as the critical messengers that convert a blunted IFN response into maximal activation of innate immunity essential for vaccine effectiveness.
