Project description:Cellulose is the most abundant component of plant litter, which is critical for terrestrial carbon cycling. Nonetheless, it remains unknown how global warming affects cellulose-decomposing microorganisms. Here, we carried out a 3-year litterbag experiment to examine cellulose decomposition undergoing +3°C warming in a tallgrass prairie. Most cellulose-associated bacteria and fungi in litterbags were also detected in bulk soil, and bacteria in litterbags had higher community-level rrn copy numbers, larger genome sizes, and higher genome guanine-cytosine (GC) contents than those in bulk soil, implying higher growth rates. Warming stimulated soil respiration by 32.3% and accelerated mass loss of cellulose, concurring with the increase in relative abundances of most functional genes associated with carbon decomposition in litterbags. Incorporating cellulose-decomposing genes into an ecosystem model reduced model parameter uncertainty and showed that warming stimulated microbial biomass, activity, and soil carbon decomposition. Collectively, our study supports a trait-centric view since cellulose-decomposing genes or genomic traits are amenable for ecosystem modeling. By characterizing the phylogenetically diverse yet functionally similar cellulose-associated microorganisms and their responses to warming, we take a step toward more precise predictions of soil carbon dynamics under future climate scenarios.

Project description:The nuclear cap-binding complex (CBC) stimulates multiple steps in several RNA maturation pathways, but how it functions in humans is incompletely understood. For small capped RNAs like pre-snRNAs, the CBC recruits PHAX. Here, we identify the CBCAP complex, composed of CBC, Ars2 and PHAX, and show that both CBCAP and CBC-Ars2 complexes can be reconstituted from recombinant proteins. Ars2 stimulates PHAX binding to the CBC as well as snRNAs 3'-end processing, hereby coupling maturation with export. In vivo, CBC and Ars2 bind similar capped non-coding and coding RNAs, and stimulate their 3M-bM-^@M-^Y-end processing. Strongest effects are displayed for cap-proximal polyadenylation sites, thereby favoring premature transcription termination. Ars2 functions in part through the mRNA 3M-bM-^@M-^Y-end cleavage factor CLP1, which binds RNA Polymerase II through PCF11. Ars2 is thus a major CBC effector that stimulates functional and cryptic 3'-end processing sites. Project description:1. The catalytic decomposition of undegraded cellulose in the form of cotton fibres is described with hydrogen peroxide at 0.4-0.04% (w/v) concentration in the presence of ferrous salts at pH3-5. 2. Complete solubilization of 5mg. of cotton fibres occurred in about 7 days in the presence of 0.4% hydrogen peroxide and 0.2mm-ferrous sulphate at the optimum pH4.2-4.3. 3. With 0.4% hydrogen peroxide the most rapid decomposition of cellulose was confined to ferrous sulphate concentrations of approx. 2-0.02mm. If the concentrations of the reagents were decreased in proportion extensive breakdown occurred but much more slowly. 4. In the primary stages of breakdown cotton fibres were disintegrated to very short fibres. These were subsequently solubilized, but there was little accumulation of soluble material. Organic matter was lost from solution as the reaction progressed. 5. Other naturally occurring cellulose-containing materials, such as grass, straw, hay and sawdust, were also disintegrated and solubilized by hydrogen peroxide and ferrous sulphate.

Project description:Global climate carbon-cycle models predict acceleration of soil organic carbon losses to the atmosphere with warming, but the size of this feedback is poorly known. The temperature sensitivity of soil carbon decomposition is commonly determined by measuring changes in the rate of carbon dioxide (CO(2)) production under controlled laboratory conditions. We added measurements of carbon isotopes in respired CO(2) to constrain the age of carbon substrates contributing to the temperature response of decomposition for surface soils from two temperate forest sites with very different overall rates of carbon cycling. Roughly one-third of the carbon respired at any temperature was fixed from the atmosphere more than 10 y ago, and the mean age of respired carbon reflected a mixture of substrates of varying ages. Consistent with global ecosystem model predictions, the temperature sensitivity of the carbon fixed more than a decade ago was the same as the temperature sensitivity for carbon fixed less than 10 y ago. However, we also observed an overall increase in the mean age of carbon respired at higher temperatures, even correcting for potential substrate limitation effects. The combination of several age constraints from carbon isotopes showed that warming had a similar effect on respiration of decades-old and younger (<10 y) carbon but a greater effect on decomposition of substrates of intermediate (between 7 and 13 y) age. Our results highlight the vulnerability of soil carbon to warming that is years-to-decades old, which makes up a large fraction of total soil carbon in forest soils globally.
