CHIP knockdown effect on breast cancer cell line

ABSTRACT: Analysis of MCF-7 cell clones lacking the E3 ubiquitin liganse CHIP. We used microarrays to detail the global gene expression profiles effected by CHIP in human brest cancer. Total RNA was extracted from CHIP or LacZ knockdown MCF7 and hybridized on Affymetrix microarrays.

Project description:Genome wide DNA methylation profiling of estrogene receptor postive breast cancer cell line MCF-7, treating atmospheric cold plasma (plasma). The Illumina Infinium Human Methylation 450k Bead chip was used to obtain DNA methylation profiles across approximately 450,000 CpGs. This profiling indicates that plasma induces epigenetic and cellular changes. Genomic DNA obtained from MCF-7 where effected by cold plasma.

Time-course effect of estradiol and estradiol-BSA on early gene expression in MCF-7 cells

Project description:Estrogens have been reported to activate several processes via membrane binding to either classic estrogen receptors (ERs) or GPR30. We have used either estradiol or BSA-conjugated estradiol in order to initiate membrane-initiated actions and ICI 172,780 (ICI) or G15 to explore ER- and GPR30-related transcription. Our results show that the majority of G15-inhibited transcription is depending on ERs, as it is also inhibited by ICI. However, a small number of transcripts, related to specific actions/pathways is either exclusively inhibited by G15, providing evidence about a specific GPR30 signature, or not inhibited by ICI or G15 suggesting the existence of another, yet unidentified estrogen receptor. Cells after a 4h incubation with medium containing 10% charcoal stripped FBS were incubated with or without E2-BSA (10-6M) in the presence or absence of specific antagonists, in DMEM/F12 (1:1) supplemented with 10% charcoal stripped FBS, for 3 hours. Total RNA was isolated using Nucleospin II columns (Macheray-Nagel, Dttren, Germany), according to the manufacturer’s instructions. RNA was labeled and hybridized according to the Affymetrix protocol (Affymetrix Gene-Chip Expression Analysis Technical Manual), using the HGU133A plus 2 chip, analyzing a total of 54675 transcripts. Signals were detected by an Affymetrix microarray chip reader.

Project description:Glioma cells are sensitized to the alkylator temozolomide after exposure to IFN-beta. In glioma-initiating cells (GIC), IFN-beta alone reduces clonogenicity. We investigated differentially expressed genes with or without IFN exposure in either longterm glioma cells or GIC. We used microarrays to investigate differential gene regulation in glioma cells after exposure to either ddH20 or IFN-beta 300 IU/ml at 6 h or 24 h. We used either the longterm cell line LNT-229 or the GIC cell lines GS-2 and GS-9 and exposed them to either IFN-beta 300 IU/ml or ddH20 for 6 h or 24 h.

Project description:The innate immune response to 1st generation adenoviral vectors was ascertained through high level infection of primary cells from a inbred mouse strain, replete with an intact and functioning TLR system. Experiment Overall Design: Quiescent murine C57/Bl6 fibroblasts were Ad (E1-E3-LacZ at MOI=300) or Mock infected. Expression data from mobilized human CD34+ cells

Project description:Bortezomib-based secondary induction therapy and mobilization could represent alternative strategies to reduce tumor burden. We used microarrays to investigate genome-wide expression changes between bortezomib and non-bortzomib mobilizaton strategies and identified distinct genes and pathways that were significantly differentially regulated. CD34+ stem cells were enriched from leukapheresis products from multiple myeloma patients treated with a bortezomib- or non-bortezomib-based mobilization. RNA extraction, amplification and hybridization on Affymetrix microarrays was performed. Post-induction stem cell collection was initiated when patients' ANC reached >1.0M-CM-^W10^9/L.
