Gene expression of the human trabecular meshwork (TM) of the eye

ABSTRACT: We investigated the gene expression of the human TM. We isolated TM cells from healthy human donor eyes. Next, we performed RNA isolation, amplification, labeling and hybridization against 44k Agilent microarrays. We performed the microarrays against a common reference sample, namely human RPE/choroid RNA. We performed 2 replicates of human TM samples from 2 different donors.

Project description:The ciliary body (CB) of the human eye consists of the non-pigmented (NPE) and pigmented (PE) neuro-epithelia. We investigated the gene expression of the NPE and PE, to shed light on the molecular mechanisms underlying the most important functions of the CB. Therefore we isolated NPE and PE cells from seven healthy human donor eyes using laser dissection microscopy. Next, we performed RNA isolation, amplification, labeling and hybridization against 44k Agilent microarrays. We performed the microarrays against a common reference sample, namely human RPE/choroid RNA. We performed 7 replicates of PE samples from 7 different donors and 7 NPE replicates from the same 7 different donors.

Comparative analysis of transcriptome profiles of G. hirsutum L. acc. TM-1 and its immature fber mutant during fiber secondary cell wall development stages

Project description:To gain novel insights into the molecular mechanisms of fiber secondary cell wall development, fiber transcriptomes of the immature fiber mutant (im) with defective secondary cell wall development and its near-isogenic line, TM-1 (wild-type) were compared using cDNA microarray technology. The expression profiling was performed at 5 developmental time points: 13, 16, 19, 22, and 25dpa. Secondary cell wall development related genes were identified by differentially expressed gene analysis. And these genes could be used as potential candidate genes for manipulation to improve fiber quality. Cotton plants were grown under field condition. Flowers were tagged and cotton bolls were collected during Fiber development stages. Total RNA was isolated from fiber bearing ovules of wild-type, TM-1 and immature fiber mutant (im) collected at various (13, 16, 19, 22 and 25 dpa) fiber development stages. A total of 15 hybridizations with three biological replicates including a swap-dye experiment for each fiber developmental stage were employed.

Microarray comparison of transcriptomic differences between long-term practitioners of the Transcendental Meditation (TM) technique and group-matched non-practitioner controls

Project description:Research on the Transcendental Meditation (TM) technique suggests it is a powerful intervention for reducing the unwanted effects of stress. Investigations of the TM program as a therapeutic intervention include cardiovascular disease (CVD), post-traumatic stress disorder (PTSD), and anxiety disorders. Healthcare utilization studies show reductions in diseases after regular practice of the program. This study compared long-term practitioners of TM (38 ± 4 years) with non-practitioner controls. We used DNA microarray global transcriptomics technologies in Peripheral Mononuclear Blood Cells (PMBCs) to compare a non-practitioner control group (Group III) and a TM-practitioner group (Group IV). Demographically-matched groups of 6 each were studied. These results are expected to be relevant to stress and stress-related diseases such as CVD, diabetes, and PTSD.
