Gene expression profile of miniature pig as animal model for toxicity evaluation by using mRNA expression profiling.

ABSTRACT: Miniature pig is a useful animal model to clarify the vital reaction and the molecular mechanisms. However, physiclogical response of pig model to sodium azide (AZIDE) stress is not reveal. To establishment of large animal model for evaluate toxic stress, whole blood of miniature pig were assessed with genomics. 7 month old clawn miniature pigs were fed AZIDE feeding. AZIDE dose of 300Âµg/kg, one hundredth of LD50 was given orally to the miniature pigs. Whole blood gene expression was measured at 0h, 6h and 24h after feeding.

Characterization of whole blood gene and while blood cell expression profiles of dietary-induced hyperlipidemia in miniature pigs

Project description:Miniature pigs are useful model animal for gene expression studies on dietary-induced hyperlipidemia, because they have similar digestive physiology to human. Two typical dietary components were used for dietary-induced hyperlipidemia miniature pig models. One is a high-fat and high-cholesterol diet (HFCD) containing 15% lard and 2% cholesterol, the other is a high-fat, high-cholesterol and high-sucrose diet (HFCSD) containing 15% lard, 2% cholesterol and 37% sucrose. Whole blood gene expression in HFCD, HFCSD and control male miniature pigs was measured at 10, 19 and 27 weeks of feeding periods.M-cM-^@M-^@White blood cell gene expression in HFCD, HFCSD and control male miniature pigs was measured at 27 weeks of feeding period.

RNA-Sequencing for Additional Molar Germs Transcriptomes at Stages of Early Development in Miniature Pigs

Project description:The molecular mechanisms of the regulatory network of mRNAs and long non-coding RNAs during sequential formation of additional molars remain poorly characterized in diphyodont mammals. The diphyodont miniature pig has proved to be a valuable model for studying human molar morphogenesis. Here, we performed RNA-seq on miniature pigs at three molar developmental stages to examine their differential gene expression profiles and potential regulatory networks during additional molar morphogenesis. Many new unannotated genes plus putative long non-coding RNAs were identified. Our data provide fundamental knowledge and a basis for understanding the molecular mechanisms governing cascade initiation of additional molars.

Genome-wide DNA methylation profile of developing deciduous tooth germ in miniature pigs

Project description:Background: DNA methylation is an important epigenetic modification critical to the regulation of gene expression during development. To date, little is known about the role of DNA methylation in tooth development in large animal models. Thus, we carried out a comparative genomic analysis of genome-wide DNA methylation profiles in E50 and E60 tooth germ from miniature pigs using methylated DNA immunoprecipitation-sequencing (MeDIP-seq).Results: We observed different DNA methylation patterns during the different developmental stages of pig tooth germ. A total of 2,469 differentially methylated genes were identified. Functional analysis identified several signaling pathways and 104 genes that may be potential key regulators of pig tooth development from E50 to E60.Conclusions: The present study provided a comprehensive analysis of the global DNA methylation pattern of tooth germ in miniature pigs and identified candidate genes that potentially regulate tooth development from E50 to E60.
