Project description:The experiment was made do to assess the influence of increased salt concentration on cucumber chloroplast transcription. We used 0,4M NaCl water solution and we checked its influence on plants after 0, 4, 8, 24, 48 hours. We have also measured changes in chloroplast gene expression after recovery of plants in water (48 hours of 0,4M NaCl solution treatment followed by 48 hours in clean water). Other growth conditions weren't changed comparing to those of standard plants growth in our experiments (check: versatile chamber growth protocol)
Project description:Experiment evaluating aplicability of PlasTi-microarray for cross-species hybridization studies. PlasTi-microarray is a tiling oligonucleotide microarray originally designed for cucumber plastome analysis.Chloroplast RNA from Arabidopsis, tomato and spinach leaves was extracted, labelled and hybridized to PlasTi-microarray with the cucumber samples labelled with the second dye as a control. For each species, one biological sample and one technical replicate (labelled in a dye-swap orientation) were analyzed, resulting in two microarray hybridizations per species and six microarrays (a-f) in total.
Project description:The expression of chloroplast genes of cucumber was influenced by prolonged low temperature treatment. We tried to asses the short term influence (0, 4, 8, 16 hours) of 4 C degree treatment in cucumber plants kept in darkness. As a control we used pooled RNA from all collected samples.
Project description:We compared organ specific chloroplast gene expression in cucumber between fully developed leaves (12th counting from the bottom of plant) and growing tips, female flowers, young leaves, young fruits (3-5 cm long). We have also characterized chloroplast gene transcription in cucumber etiolated seedlings in comparison to mature cucumber leaves.
