Project description:Samples of perirenal fat tissue from 8 Assaf breed suckling lambs. These animals were selected from a larger group of 17 Assaf suckling lambs for which carcass traits were measured. The 8 selected lambs were those showing the highest and the lowest values, from the larger group, for the percentage of perirenal and cavitary fat relative to the half carcass weight. Hence, considering the values for this trait, we defined the High-PF group (n = 4; average: 3.23 ± 0,.47) and the Low-PF group (n = 4; 1.65 ± 0,.16), respectively.

Project description:Samples of perirenal fat tissue from 8 Assaf breed suckling lambs. These animals were offspring of dams subjected to a Nutritional Protein Restriction (NPR) at the prepubertal age as described by Mateo et al. (2023) [doi: 10.1016/j.livsci.2023.105173]. These animals were reared in the same batch that other 8 Assaf suckling lambs obtained from Control dams of the NPR experiment for which RNA-Seq samples are already available (see E-MTAB-11839 for RNA-Seq data of Control lambs). These 8 NPR animals were selected from a larger group of 17 NPR suckling lambs for which carcass traits were measured. The 8 selected lambs were those from the 17 initially considered lambs showing the highest and the lowest values , from the larger group, for the percentage of kidney knob and channel fat relative to the right half carcass weight (KKCF). Hence, considering the values for this trait, we defined the High-KKCF group (n = 4; average: 3.26 ± 0.11) and the Low-KKCF group (n = 4; 1.68 ± 0.25), respectively.

Project description:Because of the central role of the liver in numerous physiological processes and its possible relation with quality carcass traits in suckling lambs, and considering that female lambs have higher carcass fatness levels, this experiment has analyzed the liver transcriptome of 12 suckling lambs (slaughter age: +/- 25 days), including six males and six females. RNA was extracted from the liver tissue of the lambs at the time of slaughter following a standard protocol. The RNA integrity value was calculated using the Agilent 2,100 Bioanalyzer equipment (Agilent Technologies, CA, USA). cDNA libraries were sequenced to a minimum depth of 30 million paired-end reads, generating 150 bp stranded paired-end reads using an Illumina Novaseq 6,000 system.

Project description:The aim of this study was to perform a comparative analysis of the perirenal fat transcriptomes of suckling lambs from two breeds with different growth and carcass characteristics. The perirenal fat tissue from 14 suckling lambs (Assaf= 8, Churra= 6) was used for the RNA-Seq analysis. The functional enrichment analysis of the 670 highly expressed genes (>150 fragments per kilobase of exon per million fragments mapped) in the perirenal fat transcriptome of both breeds revealed that the majority of these genes were involved in energy processes. The differential expression analysis performed identified 373 differentially expressed genes (DEGs) between the two compared breeds. In Assaf lambs, DEGs were enriched in Gene Ontology (GO) biological processes related to fatty-acid oxidation, while in Churra lambs, the majority of the significantly enriched GO terms were related to cholesterol synthesis, which suggests that upregulated DEGs in Assaf lambs are implicated in fat burning, while the Churra upregulated DEGs are linked to fat accumulation.

Project description:Lambs that inherit a callipyge allele from their dam have an up-regulation of maternally imprinted transcripts near the callipyge mutation but do not exhibit muscle hypertrophy. It is not clear what effects these maternally expressed transcripts have in the muscle or how the inheritance of a maternal callipyge allele prevents the expression of the callipyge phenotype which is seen paternal heterozygotes only. This experiment utilized bovine gene expression arrays to profile the gene expression of the semimembranosus muscle of 30 day old half-sibling lambs of all four possible genotypes with respect to the callipyge allele (+/+, C/+, +/C, and C/C). Experiment Overall Design: 10 lambs were used for this study. N=3 biological reps for C/C genotype, N=3 biological reps for Cmat/+ genotype, N=2 biological reps for +/Cpat genotype, and N=2 biological reps for +/+ genotype. Pairwise contrasts were performed to analyze the presence of a maternally inherited callipyge allele and the effect on a homozygous animal (+/C vs CC; +/+ vs C/+; and C/+ vs C/C). Experiment Overall Design: These data make up two experiments. Experiment Overall Design: [1] The maternal allele (4 genotype) experiment compares all four possible genotypes of a particular allele at one age and one tissue (30 days semimembranosus). Project description:The callipyge mutation causes postnatal muscle hypertrophy in heterozygous lambs that inherit a paternal callipyge allele (+/CLPG). Our hypothesis was that the up-regulation of one or both of the affected paternally expressed genes (DLK1 or PEG11) initiates changes in biochemical and physiological pathways in skeletal muscle to induce hypertrophy. The goal of this study was to identify changes in gene expression during the onset of muscle hypertrophy in order to identify the pathways that are involved in the expression of the callipyge phenotype. Gene expression was analyzed in longissimus dorsi total RNA from lambs at 10, 20, and 30 days of age using the Affymetrix Bovine Expression Array. Experiment Overall Design: Longissimus dorsi were collected from 10, 20, and 30 day old callipyge and normal lambs with 2 biological replicates for each group. Total RNA was isolated and hybridized to Affymetrix Bovine Expression arrays. Data were analyzed in SAS and all genotype effects were followed up by qPCR in 42 lambs ranging from 10 to 200 days of age.

Project description:The ovine Callipyge mutation causes postnatal muscle hypertrophy localized to the pelvic limbs and torso, as well as body leanness. The mechanism underpinning enhanced muscle mass is unclear, as is the systemic impact of the mutation. Using muscle fibre typing immunohistochemistry we confirmed muscle specific effects and demonstrated that affected muscles had greater prevalence and hypertrophy of type 2X fast twitch glycolytic fibres and decreased representation of types 1, 2C, 2A and/or 2AX fibres. To investigate potential systemic effects of the mutation, proton NMR spectra of plasma taken from lambs at 8 and 12 weeks of age were measured. Multivariate statistical analysis of plasma metabolite profiles demonstrated effects of development and genotype but not gender. Plasma from Callipyge lambs at 12 weeks of age, but not 8 weeks, was characterized by a metabolic profile consistent with contributions from the affected hypertrophic fast twitch glycolytic muscle fibres. Microarray analysis of the perirenal adipose tissue depot did not reveal a transcriptional effect of the mutation in this tissue. We conclude that there is an indirect systemic effect of the Callipyge mutation in skeletal muscle in the form of changes of blood metabolites, which may contribute to secondary phenotypes such as body leanness. Microarrays were used for transcription profiling of kidney fat samples taken from Callipyge (n=4) and wild type (n=4) lambs at 12 weeks of age. Sheep used in this experiment were bred from a research flock of Dorset/Suffolk/Rambouillet cross-bred sheep raised at Utah State University and cared for and euthanased for sample collection in accordance with the animal ethics guidelines of Utah State University (Utah, USA). Kidney fat (KF) samples were taken from lambs at 12 weeks of age; four callipyge (CN) genotype animals and four normal (NN) genotype animals were compared.

Project description:Disordered angiogenesis is implicated in the pulmonary vascular remodeling secondary to congenital heart disease (CHD). However, the underlying genes are not well delineated. We have previously shown that an ovine model of CHD with increased pulmonary blood flow (PBF, Shunt) has an M-bM-^@M-^\angiogenesis burstM-bM-^@M-^] between 1-4 weeks of age. Thus, we hypothesized that the increased pulmonary blood flow elicited a pro-angiogenic gene expression profile prior to the onset of vessel growth. To test this we utilized microarray analysis to identify genes that could be responsible for the angiogenic response. Total RNA was isolated from the lungs of Shunt and Control lambs at 3 days of age and hybridized to Affymetrix GeneChips for microarray analyses (n=8 for each group). A total of 89 angiogenesis-related genes were found to be up-regulated and 26 angiogenesis-related genes down-regulated in Shunt lungs compared to control (cutting at 1.2 fold difference, P<0.05). We then confirmed the up-regulation of pro-angiogenic genes: FGF2, Angiopoietin2 (Angpt2), and Birc5 at both mRNA and protein levels and the up-regulation of ccl2 at mRNA level in the 3-day shunt lungs. Further, we found that pulmonary arterial endothelial cells (PAEC) isolated from juvenile lambs exhibited increased expression of FGF2, Angpt2, Birc5, and ccl2 as well as enhanced angiogenesis when exposed to elevated shear stress (35 dyn/cm2) compared to cells exposed to more physiological level shear stress (20 dyn/cm2). Finally, we demonstrated that blocking FGF2, Angpt2, or Birc 5 signaling, using neutralizing antibodies, significantly decreased the angiogenic response induced by shear stress. In conclusion, we have identified a pro-angiogenic gene expression profile in a lamb model of CHD with increased PBF that precedes the onset of pulmonary vascular remodeling. Further, our data indicate that FGF2, Angiopoietin2, Birc5, and ccl2 may play important roles in the angiogenic response. Total RNA was isolated from the lungs of Shunt and Control lambs at 3 days of age and hybridized to Affymetrix GeneChips for microarray analyses. Eight chips were employed for both control and shunt groups.
