Strategic Reduction of Hybrid Insulin Peptide Formation Significantly Delays Diabetes Onset in NOD Mice

Project description:Hybrid insulin peptides (HIPs) form in pancreatic beta cells through the covalent cross-linking of proinsulin peptides with various beta cell peptides, creating unique amino acid sequences not encoded in the genome. HIPs have been confidently identified in both human and mouse islets by mass spectrometry and are targeted by autoreactive T cells in type 1 diabetes (T1D) patients and disease-triggering CD4 T cells in non-obese diabetic (NOD) mice. Previous work identified cathepsin D (CatD) as an enzyme involved in HIP formation through transpeptidation reactions targeting a conserved leucine residue (L26) in the C-peptide region of proinsulin. In this study, we generated NOD mice with a leucine-to-isoleucine substitution at this position in the insulin 2 gene (NOD INS2I/I) to prevent CatD-mediated HIP formation. Mass spectrometry analysis of islet peptidomes revealed that the L26I modification effectively blocked CatD-mediated cleavage at this site and significantly reduced the levels of disease-relevant HIPs, including HIP11 and 6.9HIP. These findings were validated through T cell assays showing reduced antigenicity of islets from modified mice when exposed to HIP-reactive T cell clones. The functional consequences of reduced HIP formation were demonstrated in a year-long monitoring study, where NOD INS2I/I mice exhibited significantly delayed disease onset compared to wild-type NOD mice, with 43% versus 10% remaining disease-free after one year. In vitro experiments with human C-peptide further confirmed that the L26I substitution prevents CatD-mediated HIP formation in human samples as well. Our results establish CatD as the primary enzymatic driver of disease-relevant HIP formation and demonstrate that preventing CatD-mediated processing of proinsulin 2 C-peptide in NOD mice significantly reduces HIP formation and delays autoimmune diabetes progression, identifying a potential therapeutic target for T1D prevention.

Localisation-dependent variations in articular cartilage ECM: implications for tissue engineering and cartilage repair

Project description:Articular cartilage (AC) is a specialised connective tissue covering joint surfaces. It enables smooth movement, distributes mechanical loads, and protects the underlying bone. In re-sponse to loading, AC adapts by modifying both its thickness and composition. AC is or-ganised in different zones, with low cellularity and a high abundance of extracellular ma-trix (ECM). Mechanical overloading or immobilisation can lead to structural changes, po-tentially resulting in osteoarthritis (OA), for which no causal treatment currently exists. However, smaller defects can be treated using chondrocyte/cartilage transplantation or tissue engineering. A better understanding of the molecular composition of AC at different locations is essential to improve such therapeutic approaches. For this purpose, we per-formed a comprehensive analysis of porcine femoral knee cartilage at eight defined ana-tomical sites. Cartilage thickness and proteoglycan (PG) content were analysed histologi-cally, while specific ECM proteins were assessed by proteomics and validated by im-munohistochemistry and Western blot. Significant differences were identified, particularly between medial and lateral compartments, in terms of cartilage thickness, PG abundance, and ECM composition. Some proteins also showed zone-specific localisation patterns. These structural differences likely reflect adaptation to mechanical loading and should be considered to optimise future cartilage repair and tissue engineering strategies.

Periostin induces early articular cartilage degeneration in developmental dysplasia of the hip

Project description:Developmental dysplasia of the hip (DDH) is one of the significant risk factors for hip osteoarthritis. In order to investigate the factors that induce early articular cartilage degeneration of the hip joints that are exposed to reduced dynamic loads arising from hip dislocation , we created rodent models of hip dislocation by swaddling. Notably, expression of periostin (Postn) was increased in the acetabular articular cartilage of the DDH models; Postn was a candidate gene associated with early articular cartilage degeneration. We showed that early articular cartilage degeneration was suppressed in Postn-/- DDH mice. Furthermore, a microgravity environment induced the expression of Postn in chondrocytes through STAT3 signaling. Postn induced catabolic factors, interleukin-6 and matrix metalloproteinase 3, in articular chondrocytes through integrin-nuclear factor κB signaling. Additionally, interleukin-6 stimulated Postn expression through STAT3 signaling. Thus, Postn plays a critical role in early articular cartilage degeneration associated with hip dislocation.

Gene expression analysis of Mycobacterium tuberculosis high-persister (hip) mutants.

Project description:Bacterial persister cells are phenotypic variants of regular cells that are tolerant to antibiotics. High-persister (hip) mutants of Mycobacterium tuberculosis produce 10- to 1,000-fold more persister cells than the wild type strain when challenged with various antibiotics. Comparison of gene expression pattern of the hip mutants may provide clues as to the genetic mechanisms underlying persister formation. Transcriptome analysis will provide information on what differentiates M. tuberculosis hip strains from regular strains, which will be useful in the development of anti-persister therapy for persister cell eradication. Twelve independent M. tuberculosis hip mutants and the wild type strain were grown to stationary phase in duplicate. To avoid analyzing the drug effect, total RNA was extracted from the cultures prior to the addition of antibiotic for hybridization to Affymetrix microarrays. Data analysis was performed by comparing the hip mutants to the wild type control.
