The cutaneous response to a mosquito bite at different diurnal time point

ABSTRACT: Examination of the transcriptional changes occurring in the skin upon mosquito bite at different times of day. Approximately 20 A. aegypti mosquitoes were allowed to bite mice on the ear at two timepoints, zeitgeber time (ZT)1 and ZT11, corresponding to dawn and dusk, respectively. Twenty-four hours later, biopsies from the mosquito bite sites were collected and transcriptomic analyses were performed. Biopsies from mice that did not receive a mosquito bite served as controls

A female mosquito salivary protein-driven influx of myeloid cells facilitates flavivirus transmission

Project description:Mosquitoes transmit many flaviviruses of global public health significance. Efficient viral transmission to mammalian hosts requires mosquito salivary factors that modulate local host responses, such as recruitment of virus-permissive myeloid cells to the bite sites. However, the specific salivary components facilitating viral transmission and their mechanisms of action remain largely unknown. Here, we showed that a female mosquito salivary gland-specific protein, named Aedes aegypti Neutrophil Recruitment Protein (AaNRP), acts as a key salivary component to facilitate the transmission of Zika (ZIKV) and dengue (DENV) viruses. AaNRP promotes a rapid influx of neutrophils followed by virus-susceptible myeloid cells toward mosquito bite sites, which facilitate establishment of local infection and systemic dissemination. Mechanistically, AaNRP engages TLR1 and TLR4 of skin resident macrophages and activates MyD88-dependent NF-κB signaling to induce the expression of neutrophil chemoattractants. Inhibition of MyD88-NF-κB with dietary resveratrol, a phytochemical, neutralizes the AaNRP effects, thus reducing flavivirus transmission by mosquitoes. This study offers mechanistic insight into saliva-aided viral transmission and provides a potential prophylactic target.

Expression data from ileum of mice suffered from subchronic and mild social defeat stress

Project description:This study aimed to investigate the effects of depression on transcriptome in ileum using a subchronic and mild social defeat stress (sCSDS) model. In addition to exhibiting social deficit and hyperphagia-like behavior, the sCSDS mice keep much more water in their body than control mice. In order to investigate the effect of social defeat stress on not only central nervous system but also function of gastrointestinal tract, the gene expression in ileum of stressed mice was compared with control mice. We used microarrays to detail the gene expression after 10 days of social defeat stress and identified distinct classes of down-regulated genes during this process. The duration of physical contacts was set at 5 min after the first attack bite at Day 1, and then was reduced 0.5 min per day from Day 2 to Day 10.

Mosaic BRAF fusions are a recurrent cause of congenital melanocytic naevi targetable by MEK inhibition

Project description:Among children with multiple congenital melanocytic naevi (CMN), 25% have no established genetic cause. Here, we study 169 CMN patients, 38 of whom were double wild-type for NRAS/BRAF mutations. Nineteen of these 38 patients had sufficient tissue to undergo RNAseq, which revealed mosaic BRAF fusions in 11/19 patients and mosaic RAF1 fusions in 1/19. These findings offer a genetic diagnosis to an additional group of patients opening the possibility for treatment options.

RNA-seq of mosquito transmitted and blood transmitted Plasmodium chabaudi

Project description:This study is a collaboration with Phil Spence and Jean Langhorne at NIMR, Mill Hill where Phil is studying the differences in aetiology of rodent malaria when mice are infected by mosquito bite versus intraperitoneal injection of blood stage parasites. The aim is to determining transcriptomic differences using RNA-Seq between parasites transmitted by mosquito and those transmitted by injection. This data is part of a pre-publication release. For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/Protocol: Mice were bled out at 6 days post-infection, and RNA was extracted from purified parasite populations using Trizol reagent and DNase treated. Poly A+ mRNA was purified from total RNA using oligo dT dyna bead selection and libraries were created using a modified RNA-seq protocol, where RNA was fragmented using Covaris AFA sonication instead of metal ions. The samples were sequenced on an Illumina HiSeq 2000.

From Local to Systemic: The Journey of Tick Bite Biomarkers in Australian patients.

Project description:Tick bites and tick-related diseases are on the rise. Diagnostic tests that identify well-characterised tick-borne pathogens (TBPs) possess limited capacity to address the causation of symptoms asso-ciated with poorly characterised tick-related illnesses, such as debilitating symptom complexes attributed to ticks (DSCATT) in Australia. Identification of local signals in tick-bitten skin that can be detected systemically in blood would have both clinical (diagnostic or prognostic) and research (mechanistic insight) utility, as a blood sample is more readily obtainable than tissue biopsies. We hypothesised that blood samples may reveal signals which reflect relevant local (tissue) events, and that the time course of these signals may align with local pathophysiology. As a first step to-wards testing this hypothesis, we contrasted molecular signatures in skin biopsies taken from the tick-bite location of human participants along with peripheral blood signatures obtained at the same time. This approach captures differentially expressed molecules across multiple omics da-tasets derived from peripheral blood (including cellular and cell-free transcriptomics, proteomics, metabolomics, and DNA methylation), and skin biopsies (spatial transcriptomics). Our data re-vealed that extracellular matrix organisation and platelet degranulation pathways were upregu-lated in skin within 72 hours of a tick bite. The same signals appeared in blood, where they then remained elevated for three months, displaying longitudinally consistent alterations of biological functions. Despite the limited sample size these data represent proof-of-concept that molecular events in the skin following a tick bite can be detectable systemically. This underscores the poten-tial value of blood samples, akin to liquid biopsy, to capture biomarkers reflecting local tissue processes.

Effect of mosquito saliva from distinct species on human dermal endothelial cell function in vitro and West Nile virus pathogenesis in vivo

Project description:During probing and feeding, an infected mosquito injects both virus and saliva into the host skin. The presence of mosquito saliva in the skin increases arbovirus pathogenesis in the bitten host, however the exact mechanism behind this remains to be determined. It is hypothesized that disease enhancement is dependent on the function of the dermal endothelium, where an increased permeability aids in the influx of virus-susceptible cells to the bite site and therefore more cells for the virus to replicate in. Here, we investigate and compare the effects of saliva from Culex and Aedes species on the human dermal endothelial cell function in vitro. Furthermore, we investigate the effect of Culex saliva on West Nile virus (WNV) pathogenesis in a mouse model. We found that salivary gland extract from anthropophilic mosquito species (Aedes and Cx. pipiens molestus) induce permeability of the human dermal endothelium, while an ornithophilic mosquito species (Cx. pip. pipiens) does not. We identified that this effect is due to the presence of protease(s) in Cx. pipiens molestus saliva. In addition, we show that the presence of Cx. saliva at the WNV inoculation site in vivo leads to more consistent weight loss, increased permeability in the inoculation site, and increased mortality compared to inoculation of WNV alone. Moving forward, identification and characterization of novel salivary proteins from similar but genetically distinct mosquito species will advance the development of intervention methods to combat potential transmission risks and disease severity of emerging mosquito-borne pathogens.
