Collagen-binding C-type natriuretic peptide enhances chondrogenesis and osteogenesis

ABSTRACT: To enhance local drug delivery and therapeutic efficacy, we developed a novel synthetic protein by fusing a collagen-binding domain (CBD) to C-type natriuretic peptide (CNP), termed CBD-CNP. CBD-CNP localized to collagen-rich tissues and promoted bone elongation.

Basal-like breast cancer stimulates MUC1 to support an epithelial/mesenchymal hybrid state and survive radiotherapy

Project description:We employed high-throughput sequencing with spatial resolution in the WAP-T mouse model of basal-like breast cancer (BLBC) to investigate the transcriptomic response to radiation therapy (RT). Our analysis revealed a distinct spatial pattern of gene expression (10X visium platform), with irradiated BLBC tumors exhibiting a strong enrichment of epithelial genes and a hybrid epithelial/mesenchymal (E/M) state. Notably, MUC1 expression was upregulated in response to RT, suggesting its role as a key driver of cell survival. These findings provide new insights into the spatial transcriptomic landscape of BLBC under RT and highlight the potential of MUC1 as a therapeutic target or prognostic marker for optimizing patient response to RT.

Effects of (-)-cannabidiol on human sebocytes

Project description:In the current study, we aimed at exploring the effects of the major non-psychotropic phytocannabinoid of Cannabis sativa, (-)-cannabidiol (CBD), on human sebaceous gland functions. We found that CBD behaved as a highly effective anti-acne agent, targeting all the three key, sebocyte-specific steps of the pathogenesis (i.e. without compromising viability or basal sebaceous lipid production, CBD normalized pro-acne agents-induced excessive lipid synthesis, reduced proliferation and exerted anti-inflammatory actions). The goal of the present microarray analyses was to identify signaling pathways and target genes being involved in mediating the above beneficial effects of CBD. Microarray analyses were performed by using three independent sets of control (vehicle-treated) and CBD-treated (10 M-NM-<M, 24-hr) human, immortalized SZ95 sebocytes. The first two sets of the samples were analyzed by two-color microarrays (M-bM-^@M-^\rep1M-bM-^@M-^] and M-bM-^@M-^\rep2M-bM-^@M-^]). Later on, the RNA from the second sample set was used for the M-bM-^@M-^\rep2-1M-bM-^@M-^] hybridization (techical repeat). Finally, we analyzed the third (biological) sample set by using one-color microarray (M-bM-^@M-^\rep3M-bM-^@M-^]).

IL-18 Inhibition Aggravates Atherosclerosis in Jak2V617F Clonal Hematopoiesis

Project description:Clonal hematopoiesis (CH) driven by Jak2V617F is known to accelerate atherosclerosis through inflammasome activation and release of IL-1β and IL-18, yet the specific contribution of IL-18 has remained unclear. In this study, we demonstrate that antibody inhibition of IL-18 in Jak2V617F CH mice increases plaque collagen but paradoxically promotes both early lesion growth and advanced necrotic core formation. Mechanistically, IL-18 blockade reverses AIM2 inflammasome activation but shifts cell death toward apoptosis, and together with impaired efferocytosis, results in greater necrosis. These events appear to be coordinated by reduced IFN-γ signaling, which enhanced collagen deposition while decreasing expression of efferocytotic genes.Our findings challenge the prevailing notion that IL-18 inhibition stabilizes atherosclerotic plaques in CH and provide new mechanistic insight into the interplay between inflammasome biology, adaptive immunity, and plaque stability.

Posttranslational modifications mediate the structural diversity of tauopathy strains

Project description:Tau aggregation is the defining pathological hallmark of tauopathies, but structural insights are revealing key differences between filaments across this class of disorders. Using cryo-electron microscopy, we identify a 107-residue fragment, K274-E380, which forms the insoluble core of tau filaments in the tauopathy corticobasal degeneration (CBD). This tau conformer is distinct from those identified in Alzheimer’s Disease (AD), Pick’s Disease, and Chronic Traumatic Encephalopathy, pointing towards a “one strain per disease” paradigm. Each disease-specific conformer can pack into multiple fibril subtypes and, despite similarities in secondary structure elements between tau filaments from CBD and AD, mass spectrometry revealed key differences in posttranslational modifications (PTMs). Given the abundance of lysine residues in the insoluble tau filament core in both CBD and AD, ubiquitination and acetylation are identified as key PTMs that compete to modify specific residues, which may ultimately determine filament morphology.
