ABSTRACT: Skeletal muscle fibers are large multinucleated cells that form specialized subdomains such as the neuromuscular and myotendinous junctions (MTJ), the latter connecting muscle to tendon and transmitting contractile force. While transcriptional heterogeneity among myonuclei is established, the contribution of local translation to subdomain identity has remained unclear. To address this, we developed MTJ-AAV, a viral system that enables selective genetic targeting of MTJ myonuclei. Using MTJ-specific ribosome tagging, we uncovered extensive translational regulation underlying MTJ function and remodeling during exercise. This repository contains RNAseq data from whole myofibers and MTJ.
