RNA-seq of Marchantia polymorpha gemmae during dormancy release

ABSTRACT: We used RNA-seq to obtain a time course transcriptomics response on the liverwort Marchantia polymorpha as gemmae are released from dormancy and initiate germination. Dormant gemmae were collected from gemmae cups of adult thalli and subsequently subjected to different light treatments. Time point 0 was sampled without imbibition. All other samples were placed directly in 100 µl water for imbibition, keeping them wet throughout the sampling period. Imbibition for 1 to 12 hours was performed in constant light or constant darkness at 20°C. The imbibition process was interrupted by freezing the samples in liquid nitrogen after 1, 4, 8, and 12 hours. RNA-seq libraries were sequenced on single-read mode (101 bp) on an Illumina NovaSeq 6000 platform. There are two libraries per sample.
