Project description:Obesity and physical inactivity have a profound impact on skeletal muscle metabolism. In the present work, we have investigated differences in protein expression and energy metabolism in primary human skeletal muscle cells established from lean donors (BMI<25 kg/m2) and individuals with obesity (BMI>30 kg/m2). Furthermore, we have studied the effect of fatty acid pretreatment on energy metabolism in myotubes from these donor groups. Alterations in protein expression were investigated using proteomic analysis, and energy metabolism was studied using radiolabeled substrates. Gene Ontology enrichment analysis showed that glycolytic, apoptotic, and hypoxia pathways were upregulated, whereas the pentose phosphate pathway was downregulated in myotubes from donors with obesity compared to myotubes from lean donors. Moreover, fatty acid, glucose, and amino acid uptake were increased in myotubes from individuals with obesity. However, fatty acid oxidation was reduced, glucose oxidation was increased in myotubes from subjects with obesity compared to cells from lean. Pretreatment of myotubes with palmitic acid (PA) or eicosapentaenoic acid (EPA) for 24 h increased glucose oxidation and oleic acid uptake. EPA pretreatment increased the glucose and fatty acid uptake and reduced leucine fractional oxidation in myotubes from donors with obesity. In conclusion, these results suggest that myotubes from individuals with obesity showed increased fatty acid, glucose, and amino acid uptake compared to cells from lean donors. Furthermore, myotubes from individuals with obesity had reduced fatty acid oxidative capacity, increased glucose oxidation, and a higher glycolytic reserve capacity compared to cells from lean donors. Fatty acid pretreatment enhances glucose metabolism, and EPA reduces oleic acid and leucine fractional oxidation in myotubes from donor with obesity, suggesting increased metabolic flexibility after EPA treatment

Project description:Metabolically healthy skeletal muscle is characterized by the ability to switch easily between glucose and fat oxidation, whereas loss of this ability seems to be related to insulin resistance. The aim of this study was to investigate whether different fatty acids (FAs) and the LXR ligand T0901317 affected metabolic switching in human skeletal muscle cells (myotubes). Pretreatment of myotubes with eicosapentaenoic acid (EPA) increased suppressibility, the ability of glucose to suppress FA oxidation, and metabolic flexibility, the ability to increase FA oxidation when changing from “fed” to “fasted” state. Adaptability, the capacity to increase FA oxidation with increasing FA availability, was increased after pretreatment with EPA, linoleic acid (LA) and palmitic acid (PA). T0901317 counteracted the effect of EPA on suppressibility and adaptability, but did not affect these parameters alone. EPA itself accumulated less, however, EPA, LA, OA and T0901317 increased the number of lipid droplets (LDs) in myotubes, whereas LD size and mitochondria amount were independent of pretreatment. Microarray analysis showed that EPA regulated more genes than the other FAs. Some pathways involved in carbohydrate metabolism were induced only by EPA. The present study suggests a possible favorable effect of EPA on skeletal muscle metabolic switching and glucose utilization. Keywords: Analysis of target gene regulation by using microarrays. Project description:Satellite cells (SCs), the stem cell population of skeletal muscle, are crucial for growth and regeneration, and their dysfunction is increasingly recognized as a contributing factor to Duchenne muscular dystrophy (DMD). DMD is a severe, X-linked disorder caused by DMD gene mutations, leading to loss of dystrophin expression in muscle tissue and a progressive muscle degeneration. Here, we provide RNA-seq data from wild-type (WT), dystrophin-deficient (DMD), and heterozygous (HET) pig SCs, representing a translational model for human DMD. Muscle biopsies were collected from fetal and 3-day-old animals, enzymatically digested, and subjected to magnetically activated cell sorting for SC purification. Purified cells were cultured under normal growth conditions in proliferation (PROL) and after induction to differentiate into multinucleated myotubes (DIFF), and total RNA was extracted for 3′ mRNA-seq. Libraries were prepared using NexteraXT and sequenced on a NextSeq550Dx. After quality control and deduplication, reads were aligned to the Sus scrofa reference genome, and gene counts were generated with STAR. Parts of this data have been previously published in a larger study and are now made fully accessible as an independent dataset. By capturing stage- and genotype-specific transcriptional signatures of SCs, this dataset offers new insights into the molecular defects associated with dystrophin-deficiency and serves as a reference resource for future studies investigating DMD pathogenesis.

Project description:We identified the target genes of FTO ("fat mass and obesity associated") in primary cultures of human skeletal muscle cells using adenoviral vectors expressing FTO or GFP and oligonucleotide microarrays. Human myotubes were prepared from 4 different skeletal muscle biopsies. After differentiation, myotubes were infected for 48 hours with recombinant adenovirus expressing either GFP or FTO. Each FTO-infected myotubes culture was compared to GFP-infected myotubes culture. GFP-infected myotubes were regarded as the control. Four biological replicates were processed.

Project description:Background. Skeletal muscle is a major target organ for ethanol induced perturbations with resultant sarcopenia in alcohol-related liver disease (ALD). Targeted studies show that in skeletal muscle, ethanol causes mitochondrial oxidative dysfunction and impaired mTORC1 signaling with consequent decreased protein synthesis and increased autophagy. However, complex interactions and pathway intersections involved in cellular adaptive and maladaptive responses are not well understood. Unlike hypotheses driven focused experiments, an integrated multiomics-experimental validation approach helps identify the global landscape of complex, interacting perturbations in the skeletal muscle. Methods. We used an integrated multiomics approach in a comprehensive array of models, including murine and human induced pluripotent stem cell-derived myotubes (hiPSCm), skeletal muscle from a mouse model of ALD (mALD), and human patients with alcohol-related cirrhosis (CIR) and controls (CON), to identify novel regulatory mechanisms of sarcopenia in ALD. We generated 13 untargeted datasets, including chromosomal conformation, RNA sequencing, proteomics, phosphoproteomics, acetylomics, and metabolomics and conducted a comprehensive analysis using upset plots and feature extraction-based approaches. The most altered molecular interactions were experimentally validated in different models using immunoblots, redox measurements, imaging methods, and measures of senescence associated molecular phenotype (SAMP). Redox ratio was reversed using mitochondrial targeted Lactobacillus brevis NADH oxidase (MitoLbNOX). Results. Enrichments in mitochondrial oxidative function, protein synthesis, and senescence pathways were identified. Multiomics analyses were consistent with the known effects of hypoxia inducible factor 1 (HIF1) that inhibits mitochondrial oxidative dysfunction and promotes skeletal muscle senescence. Pertubrations in genes in the nicotinamide dinucleotide-related pathways including sirtuin signaling, known regulators of senescence, was also noted with ethanol treatment. Experimentally, HIF1 protein was stabilized in myotubes and muscle tissue without cellular hypoxia. Mitochondrial electron transport chain protein expression was less in murine myotubes and hiPSCm with lower redox ratio, less expression of sirtuins, and increased acetylation of mitochondrial proteins. SAMP noted in multiple models in response to ethanol exposure and reversing redox ratio with MitoLbNOX prevented HIF1a stabilization and SAMP in murine myotubes. Conclusions. Our integrated multiomics analyses combined with complementary experimental validation, identify HIF1a stabilization with accelerated skeletal muscle post-mitotic senescence as novel mechanisms of sarcopenia in ALD, highlighting the complex interactions that result in mitochondrial dysfunction with persistent and progressive sarcopenia in ALD.

Project description:Liver X receptors (LXRs) are important regulators of cholesterol, lipid and glucose metabolism and have been extensively studied in liver, macrophages and adipose tissue. However, their role in skeletal muscle is not yet fully elucidated and the functional role of each of the LXRalpha (NR1H3) and LXRbeta (NR1H2) subtypes in skeletal muscle is at present unknown. To study the importance of each of the receptor subtypes, myotube cultures derived from wild type (WT), LXRalpha and LXRbeta knockout (KO) mice were established. The present study shows that treatment with the unselective LXR agonist T0901317 increased mRNA levels of LXR target genes such as sterol regulatory element-binding transcription factor 1 (SREBF1), fatty acid synthase (FASN), stearoyl-CoA desaturase 1 (SCD1) and ATP-binding cassette transporter A1 (ABCA1) in myotubes established from WT and LXRalpha KO mice. However, only minor changes in expression level were observed for these genes after treatment with T0901317 in myotubes from LXRbeta KO mice. Gene expression analysis using Affymetrix NuGO Genechip arrays showed that few other genes than the classical, well known LXR target genes were regulated by LXR in skeletal muscle. Furthermore, functional studies using radiolabeled substrates showed that treatment with T0901317 increased lipogenesis and apoA1 dependent cholesterol efflux, in myotubes from WT and LXRalpha KO mice, but not LXRbeta KO mice. The data suggest that the lipogenic effects of LXRs, as well as the LXR-stimulated cholesterol efflux, are mainly mediated by LXRbeta in skeletal muscle.
