Project description:Humic acid (HA), not only promote the growth of crop roots, they can be combined with nitrogen (N) to increase fertilizer use efficiency and yield. However, the effects of HA urea fertilizer (HA-N) on root growth and yield of sweet potato has not been widely investigated. Xushu 28 was used as the experimental crop to investigate the effects of HA-N on root morphology, active oxygen metabolism and yield under field conditions. Results showed that nitrogen application alone was not beneficial for root growth and storage root formation during the early growth stage. HA-N significantly increased the dry weight of the root system, promoted differentiation from adventitious root to storage root, and increased the overall root activity, total root length, root diameter, root surface area, as well as root volume. HA-N thus increased the activity of superoxide dismutase (SOD), peroxidase (POD), and Catalase (CAT) as well as increasing the soluble protein content of roots and decreasing the malondialdehyde (MDA) content. HA-N significantly increased both the number of storage roots per plant increased by 14.01%, and the average fresh weight per storage root increased by 13.7%, while the yield was also obviously increased by 29.56%. In this study, HA-N increased yield through a synergistic increase of biological yield and harvest index.
Project description:The enhancement of plant tolerance toward abiotic stresses is increasingly being supported by the application of biostimulants. Salinity represents a serious problem in the Mediterranean region. To verify the effects deriving from the application of biostimulants, trials on Romaine lettuce plants under salt exposure were performed, in greenhouse. Plants were subjected to three NaCl solutions with 0.8, 1.3, and 1.8 dS/m of electrical conductivity. The volume of the solution was 200 mL/plant and delivered every 3 days. Biostimulant treatments started after crop establishment and were: control (water) and two doses (0.1 or 0.2 mL/plant) of the commercial biostimulant Retrosal® (Valagro S.p.A), containing calcium, zinc, and specific active ingredients. Four Retrosal® treatments were applied, every 7 days, directly to the substrate. Non-destructive analyses were conducted to assess the effects on leaf photosynthetic efficiency. At harvest, plants fresh weight (FW) and dry weight were determined, as well as the concentration of chlorophylls, carotenoids, total sugars, nitrate, proline, and abscisic acid (ABA). The biostimulant tested increased significantly the FW of lettuce (+65% in the highest dose) compared to controls. Results indicate that treatments positively affected the chlorophyll content measured in vivo (+45% in the highest dose) and that a general positive effect was observable on net photosynthesis rate. Retrosal® seems to improve the gas exchanges under our experimental conditions. The total sugars levels were not affected by treatments. Biostimulant allowed maintaining nitrate concentration similar to the untreated and unstressed controls. The increasing levels of water salinity caused a raise in proline concentration in control plants (+85%); biostimulant treatments at 0.2 mL/plant dose kept lower the proline levels. All plants treated with the biostimulant showed lower value of ABA (-34%) compared to controls. Results revealed that Retrosal® is able to stimulate plant growth independently from the salinity exposure. However, treated plants reached faster the commercial maturity stage. The fresh biomass of control at the end of experiment, after 30 days, ranged from 15 to 42 g/head, while in biostimulant treated plants ranged from 45 to 94 g/head. The product applied at maximum dose seems to be the most effective in our experimental conditions.

The data were analysed to assess the potential of transcriptomics to detect differences in gene expression as a result of these environmental conditions. Differences were found for both factors, both in PCA and in ANOVA analysis.
Project description:Drought is a common environmental stress with great negative impacts on plant growth, development and geographical distribution as well as agriculture and food production. Sweet potato is characterized by starchy, fresh and pigmented tuber, and is regarded as the seventh most important food crop. However, there has been no comprehensive study of the drought tolerance mechanism of different sweet potato cultivars to date. Here, we studied the mechanism for drought response of seven sweet potato drought-tolerant cultivars using the drought coefficients, physiological indicators and transcriptome sequencing. The seven sweet potato cultivars were classified into four groups of drought tolerance performance. A large number of new genes and transcripts were identified, with an average of about 8000 new genes per sample. Alternative splicing events in sweet potato, which were dominated by first exon and last exon alternative splicing, were not conserved among different cultivars and not significantly affected by drought stress. Furthermore, different drought-tolerance mechanisms were revealed through differentially expressed gene analysis and functional annotation. Two drought-sensitive cultivars, Shangshu-9 and Xushu-22, mainly resisted drought stress by up-regulating plant signal transduction. The other drought-sensitive cultivar Jishu-26 responded to drought stress by down-regulating isoquinoline alkaloid biosynthesis and nitrogen/carbohydrate metabolism. In addition, the drought-tolerant cultivar Chaoshu-1 and drought-preferred cultivar Z15-1 only shared 9% of differentially expressed genes, as well as many opposite metabolic pathways in response to drought. They mainly regulated flavonoid and carbohydrate biosynthesis/metabolism in response to drought, while Z15-1 increased photosynthesis and carbon fixation capacity. The other drought-tolerant cultivar Xushu-18 responded to drought stress by regulating the isoquinoline alkaloid biosynthesis and nitrogen/carbohydrate metabolism. The extremely drought-tolerant cultivar Xuzi-8 was almost unaffected by drought stress and responded to drought environment only by regulating the cell wall. These findings provide important information for the selection of sweet potatoes for specific purposes.
Project description:The influence of ultrasound (US) pretreatments combined with infrared (IRD) and hot-air (HAD) drying on drying kinetics, mathematical modeling, bioactive compounds (antioxidant activities, Vitamin C, phenolics, and flavonoid contents), qualitative properties (β-carotene, total carotenoids, color indexes, textural profile), enzyme inactivation, and exergetic analysis of sweet potatoes. The US pretreatment at 40 kHz combined with IRD and HAD (70 °C) significantly lessened the drying time and water contents. Besides, it did not affect the sweet potato's bioactive components and other quality-related attributes. The samples' activation energy (Ea) ranged from 17.60 to 29.86 kJ/mol for both dryers, with R2 (0.999-0.9809). Control samples had the highest specific energy consumption (SEC) due to the extended drying period, whereas ultrasound (40 kHz) treated samples had the lowest SEC during HAD and IRD at 80 °C. The thermodynamic parameters indicated that increasing the drying temperature lowers the enthalpy and Gibbs free energy, while entropy resulted in negative values. HAD had better textural qualities (hardness and resilience). The US pretreatments followed by HAD or IRD may lead to an energy-efficient method with acceptable quality maintenance.
Project description:Nothofagus alessandrii (ruil) is an endangered relict species, endemic to the Mediterranean area of Chile, and one of the most threatened trees in the country. Its natural distribution area has been greatly reduced by the effect of human activities; the remaining fragments are mostly intervened and highly deteriorated as a habitat and refuge for the associated biodiversity. In order to produce healthy and resistant nursery plants for recovery and restoration of N. alessandrii forests, this study evaluates the early effects of mycorrhizal fungal inoculum (MFI) combined with fertilization on the cultivation of seedlings. The experiment was established under a completely randomized design with a factorial arrangement of the mycorrhizal factors (M0 = without mycorrhizal, M1 = Thelephora sp. and M2 = Hebeloma sp.) and fertilization (F1 = standard fertilization and F2 = intensive fertilization), with three replicates of each combination, for each type of plant (P1 = plants from one season and P2 = plants from two seasons). Each experimental unit corresponded to a group of 20 plants, with 720 plants in the test. The results indicate that application of fertilizer and MFI significantly affects some growth and photosynthesis parameters of ruil plants in one and two seasons. The morphological parameters obtained in the study show shoot height values ranging between 67 and 91 cm for P1 and between 96 and 111 cm for P2; while, for shoot diameter, values ranged between 7.91 and 8.24 mm for P1 and between 10.91 and 11.49 mm for P2. Although formation of fully developed mycorrhizal roots was not observed during the assay period, we conclude that inoculation of mycorrhizal fungi combined with fertilization could be an efficient strategy to produce a quality plant, in addition to maintaining a high photosynthetic capacity and, therefore, a higher percentage of survival in the field.
Project description:DNA modification techniques are increasingly applied to improve the agronomic performance of crops worldwide. Before cultivation and marketing, the environmental risks of such modified varieties must be assessed. This includes an understanding of their effects on soil microorganisms and associated ecosystem services. This study analyzed the impact of a cisgenic modification of the potato variety Desirée to enhance resistance against the late blight-causing fungus Phytophthora infestans (Oomycetes) on the abundance and diversity of rhizosphere inhabiting microbial communities. Two experimental field sites in Ireland and the Netherlands were selected, and for 2 subsequent years, the cisgenic version of Desirée was compared in the presence and absence of fungicides to its non-engineered late blight-sensitive counterpart and a conventionally bred late blight-resistant variety. At the flowering stage, total DNA was extracted from the potato rhizosphere and subjected to PCR for quantifying and sequencing bacterial 16S rRNA genes, fungal internal transcribed spacer (ITS) sequences, and nir genes encoding for bacterial nitrite reductases. Both bacterial and fungal communities responded to field conditions, potato varieties, year of cultivation, and bacteria sporadically also to fungicide treatments. At the Dutch site, without annual replication, fungicides stimulated nirK abundance for all potatoes, but with significance only for cisgenic Desirée. In all other cases, neither the abundance nor the diversity of any microbial marker differed between both Desirée versions. Overall, the study demonstrates environmental variation but also similar patterns of soil microbial diversity in potato rhizospheres and indicates that the cisgenic modification had no tangible impact on soil microbial communities.
Project description:Sweet potato provides rich nutrients and bioactive substances for the human diet. In this study, the volatile organic compounds of five pigmented-fleshed sweet potato cultivars were determined, the characteristic aroma compounds were screened, and a correlation analysis was carried out with the aroma precursors. In total, 66 volatile organic compounds were identified. Terpenoids and aldehydes were the main volatile compounds, accounting for 59% and 17%, respectively. Fifteen compounds, including seven aldehydes, six terpenes, one furan, and phenol, were identified as key aromatic compounds for sweet potato using relative odor activity values (ROAVs) and contributed to flower, sweet, and fat flavors. The OR sample exhibited a significant presence of trans-β-Ionone, while the Y sample showed high levels of benzaldehyde. Starch, soluble sugars, 20 amino acids, and 25 fatty acids were detected as volatile compounds precursors. Among them, total starch (57.2%), phenylalanine (126.82 ± 0.02 g/g), and fatty acids (6.45 μg/mg) were all most abundant in Y, and LY contained the most soluble sugar (14.65%). The results of the correlation analysis revealed the significant correlations were identified between seven carotenoids and trans-β-Ionone, soluble sugar and nerol, two fatty acids and hexanal, phenylalanine and 10 fatty acids with benzaldehyde, respectively. In general, terpenoids and aldehydes were identified as the main key aromatic compounds in sweet potatoes, and carotenoids had more influence on the aroma of OR than other cultivars. Soluble sugars, amino acids, and fatty acids probably serve as important precursors for some key aroma compounds in sweet potatoes. These findings provide valuable insights for the formation of sweet potato aroma.
Project description:BackgroundUltra-processed foods (UPF) are becoming extensively available in the food environments. UPF are industrial formulations that are designed to maximize palatability and consumption through a combination of calorie-dense ingredients and chemical additives. UPFs are also aggressively marketed, which may make them more attractive than unprocessed/minimally processed foods (UMPF). Since consumers' purchase decisions are guided by food-evoked emotions, we aimed to provide evidence that UPF visual cues trigger higher emotional responses and approach motivation than UMPF visual cues, with potential impacts on individuals' intention to consume the UPF over the UMPF.MethodsParticipants (n = 174; 144 women; mean age = 20.7 years; standard deviation = 4.35) performed two tasks. In the first task, 16 pictures of foods (8 UPF and 8 UMPF), and 74 pictures from other affective categories, were presented. After viewing each picture, the participants rated it along two basic dimensions of emotion through the Self-Assessment Manikin scale: pleasantness and arousal. In the second task, the participants viewed the same food pictures, and they rated their intention to consume the foods depicted in the pictures. Each picture was plotted in terms of its mean pleasantness and arousal ratings in a Cartesian plane, which resulted in an affective space.ResultsPictures of UPF and UMPF were positioned in the upper arm of the boomerang-shaped affective space that represents approach motivation. Pictures containing UPF triggered higher approach motivation and intention to consume than pictures containing UMPF. We also found a stronger association between emotional responses and intention to consume UPF relative to UMPF.ConclusionThese results shed new light on the role of ultra-processed foods evoked emotions that contribute to less healthy and sustainable food environments.
Project description:To improve our understanding of how coal mining areas can be re-vegetated and ecosystem function restored, we examined the potential effects of five water (W) regimes (40, 50, 60, 70 and 80% of field capacity), five nitrogen (N) (0, 24, 60, 96 and 120 mg kg‒1 soil) and five phosphorus (P) fertilizer doses (0, 36, 90, 144 and 180 mg kg‒1 soil), which control the growth and development of Elaeagnus angustifolia under adverse environmental conditions. To optimize the W-N-P application rate, three factors and five levels of central composite design along with an optimization technique named response surface methodology were utilized. Here we provide data on root-shoot biomass ratio, leaf dry matter content, stomatal conductance, chlorophyll (Chl) a, Chl b, membrane stability index and soluble protein content of E. angustifolia. The data described in this article are available in Mendeley Data, DOI: 10.17632/2vfbrdxyf2.2[1]. These data could be used to evaluate the improvement in growth performance of E. angustifolia subjected to various regimes of W, N and P. This dataset showed that E. angustifolia grew optimally in coal-mine spoils when irrigated at 66% of field capacity and supplemented with 74.0 mg N and 36.0 mg P kg‒1 soil. This could considerably help the success of revegetation in coal-mined degraded arid areas where W is scarce.
