Gastrointestinal parasites in captive olive baboons in a UK safari park.

ABSTRACT: From the safety inside vehicles, Knowsley Safari offers visitors a close-up encounter with captive olive baboons. As exiting vehicles may be contaminated with baboon stool, a comprehensive coprological inspection was conducted to address public health concerns. Baboon stools were obtained from vehicles, and sleeping areas, inclusive of video analysis of baboon–vehicle interactions. A purposely selected 4-day sampling period enabled comparative inspections of 2662 vehicles, with a total of 669 baboon stools examined (371 from vehicles and 298 from sleeping areas). As informed by our pilot study, front-line diagnostic methods were: QUIK-CHEK rapid diagnostic test (RDT) (Giardia and Cryptosporidium), Kato–Katz coproscopy (Trichuris) and charcoal culture (Strongyloides). Some 13.9% of vehicles were contaminated with baboon stool. Prevalence of giardiasis was 37.4% while cryptosporidiosis was <0.01%, however, an absence of faecal cysts by quality control coproscopy, alongside lower than the expected levels of Giardia-specific DNA, judged RDT results as misleading, grossly overestimating prevalence. Prevalence of trichuriasis was 48.0% and strongyloidiasis was 13.7%, a first report of Strongyloides fuelleborni in UK. We advise regular blanket administration(s) of anthelminthics to the colony, exploring pour-on formulations, thereafter, smaller-scale indicator surveys would be adequate.

Project description:BackgroundThe hamadryas baboon (Papio hamadryas) is a highly social primate that lives in complex multilevel societies exhibiting a wide range of group behaviors akin to humans. In contrast to the widely studied human microbiome, there is a paucity of information on the host-associated microbiomes of nonhuman primates (NHPs). Here, our goal was to understand the microbial composition throughout different body sites of cohabiting baboons.ResultsWe analyzed 170 oral, oropharyngeal, cervical, uterine, vaginal, nasal and rectal samples from 16 hamadryas baboons via 16S rRNA gene sequencing. Additionally, raw Miseq sequencing data from 1041 comparable publicly available samples from the human oral cavity, gut and vagina were reanalyzed using the same pipeline. We compared the baboon and human microbiome of the oral cavity, gut and vagina, showing that the baboon microbiome is distinct from the human. Baboon cohabitants share similar microbial profiles in their cervix, uterus, vagina, and gut. The oral cavity, gut and vagina shared more bacterial amplicon sequence variants (ASVs) in group living baboons than in humans. The shared ASVs had significantly positive correlations between most body sites, suggesting a potential bacterial exchange throughout the body. No significant differences in gut microbiome composition were detected within the maternity line and between maternity lines, suggesting that the offspring gut microbiota is shaped primarily through bacterial exchange among cohabitants. Finally, Lactobacillus was not so predominant in baboon vagina as in the human vagina but was the most abundant genus in the baboon gut.ConclusionsThis study is the first to provide comprehensive analyses of the baboon microbiota across different body sites. We contrast this to human body sites and find substantially different microbiomes. This group of cohabitating baboons generally showed higher microbial diversity and remarkable similarities between body sites than were observed in humans. These data and findings from one group of baboons can form the basis of future microbiome studies in baboons and be used as a reference in research where the microbiome is expected to impact human modeling with baboons.

Project description:BackgroundZoological gardens (Zoo) provide abode to various threatened animals or trafficked animals seized by the authorities, and injured and orphaned animals. Captive animals are more susceptible to infection as they are under significant stress due to diet and space which further dwindle their resistance to parasitic infections.ObjectivesThis study was conducted to determine the prevalence and burden of gastrointestinal parasites in captive mammals housed at Central Zoo.MethodsFresh faecal samples from three orders of mammals including carnivora (n = 24), rodentia (n = 28) and artiodactyla (n = 35) were examined by direct smear, faecal floatation and sedimentation techniques, and the McMaster technique was applied to quantify parasite eggs per gram (EPG)/oocysts per gram (OPG) of a faecal sample.ResultsOne or more parasite taxa were detected in 19.54% of the examined samples and five types of GIPs including one protozoon (Eimeria spp.) and four helminths (Strongyloides spp., Haemonchus spp. and Trichostrongylus spp. and hookworm) were recorded. The protozoan prevalence (6.89 %) was lower than helminths (12.64%). The Eimeria spp. was the most prevalent parasite (6.89%) with the highest OPG (427.77 ± 25.45SD) in spotted deer (Axis axis), and the highest prevalence was noticed among artiodactyla (34.28%) followed by carnivora (12.5%) and rodentia (7.14%). Artiodactyla had both single infection (25.71%) and double (8.57%) infection. The percentage of single infection (16.09%) was found to be higher than double infection (3.44%) among the captive mammals. The wild boar (Sus scrofa) had the highest EPG of 383.33 ± 76.37SD (Strongyloides spp.), while the spotted deer had the lowest EPG of 216.66 ± 76.37SD (hookworm).ConclusionsDespite careful management practices, the parasitic infection may be attributed to the narrow enclosure, group housing and environmental contamination. The present finding provides baseline information on the parasitic infection in captive mammals, and can be used by zoo managers for the better life of captive animals.
