Nearly Complete Genome Sequences of Two Bluetongue Viruses Isolated during the 2020 Outbreak in the Grand Duchy of Luxembourg.

ABSTRACT: Bluetongue is one of the major diseases of ruminants listed by the World Organisation for Animal Health. Bluetongue virus serotype 8 (BTV-8) has been considered enzootic in France since 2018. Here, we report the nearly complete genome sequences of two BTV-8 isolates from the 2020 outbreak in the Grand Duchy of Luxembourg.

Nearly complete genome sequences of the first two identified Colorado potato beetle viruses.

Project description:The Colorado potato beetle is one of the most devastating potato pests in the world. However, its viral pathogens, which might have potential in pest control, have remained unexplored. With high-throughput sequencing of Colorado potato beetle samples derived from prepupal larvae which died from an unknown infection, we have identified two previously unknown RNA viruses and assembled their nearly complete genome sequences. The subsequent genetic and phylogenetic analysis demonstrated that the viruses, tentatively named Leptinotarsa iflavirus 1 and Leptinotarsa solinvi-like virus 1, are the novel representatives of the Iflaviridae and Solinviviridae families, respectively. To the best of our knowledge, these are the first sequencing-confirmed insect viruses derived from Colorado potato beetle samples. We propose that Leptinotarsa iflavirus 1 may be associated with a lethal disease in the Colorado potato beetle.

Complete genome sequences of Sathuperi and Shamonda viruses isolated in Japan.

Project description:Sathuperi virus (SATV) and Shamonda virus (SHAV) (family Peribunyaviridae, genus Orthobunyavirus, species Schmallenberg orthobunyavirus) have been suggested to cause congenital abnormalities in ruminants. In this study, we determined the complete genome sequences of SATV KSB-6/C/02 and SHAV KSB-2/C/08 strains, which were obtained from Culicoides biting midges in Japan, by next-generation sequencing and Sanger sequencing. The 3'- and 5'-untranslated region sequences of the M segment of SHAV KSB-2/C/08 strains are distinctly different from those of SATV KSB-6/C/02 and Schmallenberg viruses. This study provides the genome characterization of Japanese strains of SATV and SHAV and presented the genetic variation in the untranslated regions of Schmallenberg orthobunyavirus M segments.

The sequences comprise 13 mosquito isolates and 1 human isolate following a documented laboratory infection. This study expands the publicly available data for this neglected virus from 4 to 18 sequences.

The genetic characterization of different enterovirus strains allows for a better understanding of their molecular epidemiology and viral evolution.
