Effects of different biomass materials as a salt-isolation layer on water and salt migration in coastal saline soil.

ABSTRACT: The aim of this study was to find a material suited for the prevention of evaporative water loss and salt accumulation in coastal saline soils. One-dimensional vertical water infiltration and phreatic evaporation experiments were conducted using a silty loam saline soil. A 3-cm-thick layer of corn straw, biochar, and peat was buried at the soil depth of 20 cm, and a 6-cm-thick layer of peat was also buried at the same soil depth for comparison. The presence of the biochar layer increased the upper soil water content, but its ability to inhibit salt accumulation was poor, leading to a high salt concentration in the surface soil. The 3-cm-thick straw and 6-cm-thick peat layers were most effective to inhibit salt accumulation, which reduced the upper soil salt concentration by 96% and 93%, respectively. However, the straw layer strongly inhibited phreatic evaporation and resulted in low water content in the upper soil layer. Compared with the straw layer, the peat layer increased the upper soil water content. Thus, burying a 6-cm-thick peat layer in the coastal saline soil is the optimal strategy to retain water in the upper soil layer and intercept salt in the deeper soil layer.

Project description:Soil salinization limits crop growth and yield in agro-ecosystems worldwide by reducing soil health and altering the structure of microbial communities. Salt-tolerant plant growth-promoting rhizobacteria (PGPR) alleviate plant salinity stress. Wild soybean (Glycine soja Sieb. and Zucc.) is unique in agricultural ecosystems owing to its ability to grow in saline-alkali soils and fix atmospheric nitrogen via symbiotic interactions with diverse soil microbes. However, this rhizosphere microbiome and the nodule endosymbionts have not been investigated to identify PGPR. In this study, we investigated the structural and functional rhizosphere microbial communities in saline-alkali soil from the Yellow River Delta and coastal soil in China, as well as wild soybean root nodule endosymbionts. To reveal the composition of the microbial ecosystem, we performed 16S rRNA and nifH gene amplicon sequencing on root nodules and root zones under different environmental conditions. In addition, we used culture-independent methods to examine the root bacterial microbiome of wild soybean. For functional characterization of individual members of the microbiome and their impact on plant growth, we inoculated isolates from the root microbiome with wild soybean and observed nodulation. Sinorhizobium/Ensifer accounted for 97% of the root nodule microbiome, with other enriched members belonging to the phyla Actinobacteria, Bacteroidetes, Chloroflexi, Acidobacteria, and Gemmatimonadetes; the genera Sphingomonas, Microbacterium, Arthrobacter, Nocardioides, Streptomyces, Flavobacterium, Flavisolibacter, and Pseudomonas; and the family Enterobacteriaceae. Compared to saline-alkali soil from the Yellow River Delta, coastal soil was highly enriched for soybean nodules and displayed significant differences in the abundance and diversity of β-proteobacteria, δ-proteobacteria, Actinobacteria, and Bacteroidetes. Overall, the wild soybean root nodule microbiome was dominated by nutrient-providing Sinorhizobium/Ensifer and was enriched for bacterial genera that may provide salt resistance. Thus, this reductionist experimental approach provides an avenue for future systematic and functional studies of the plant root microbiome.

Project description:Poor soil quality is affected by salinity, which limits land productivity and sustainable agricultural development in coastal China. Hence, it is essential to choose suitable and efficient approaches to revitalize coastal saline soil quality and improve agricultural productivity. Biochar and polyacrylamide (PAM) have been widely applied as soil amendments to enhance soil structure, but the interactive effects of biochar and PAM on rice growth are unclear. The experiment described in this study was conducted over five consecutive growing seasons (from 2016 to 2020) with biochar (at 0, 32, and 79 t/hm2) and PAM (at 0, 0.6, and 1.6 t/hm2) applications to study the effects of amendments on soil properties, rice photosynthesis, and rice yield in coastal saline land. The soil property results showed that wheat straw biochar and PAM lowered soil total salt and bulk density, but increased the soil organic matter (SOM), mean weight diameter of water-stable aggregates (MWD), and macroaggregate (> 0.25 mm) content. The application of either biochar or PAM increased the rice net photosynthetic rate, transpiration rate, and stomatal conductance. The combined application of 32 t/hm2 biochar + 0.6 t/hm2 PAM increased the net photosynthetic rate by 26.0% and the transpiration rate by 24.8% relative to the control. The application of 32 t/hm2 biochar and 1.6 t/hm2 PAM significantly increased the rice grain yield. The path analysis model showed that spikelets per panicle and canopy gross photosynthesis had strong and significant positive effects on grain yield, whereas soil total salt had a negative effect on grain yield. The combined application of 32 t/hm2 biochar + 0.6 t/hm2 PAM was identified as the most effective for rice growth. Biochar and PAM amendments at an optimal level may enhance soil properties by reducing salinity. These findings indicate that biochar and PAM have the potential to remediate coastal saline soil quality and the environment, which would simultaneously increase the sustainable use of coastal land resources and food production to preserve the ecological environment.

Project description:Soil salinization is one of the major land degradation processes that decreases soil fertility and crop production worldwide. In this study, a long-term coastal saline soil remediation experiment was conducted with three salt-tolerant plant species: Lycium chinense Mill. (LCM), Tamarix chinensis Lour. (TCL), and Gossypium hirsutum Linn. (GHL). The three plant species successfully remediated the saline soil but showed different efficacies. The archaeal, bacterial, and fungal communities in barren soil and in four rhizocompartments (distal-rhizosphere soil, proximal-rhizosphere soil, rhizoplane, and endosphere) of the three plant species were assessed. All three plant species significantly decreased the richness of the archaeal communities but increased that of the bacterial and fungal communities in both the rhizosphere and rhizoplane compared with those in the barren soil. The archaeal and bacterial community structures were strongly influenced by the rhizocompartment, while specific fungal communities were recruited by different plant species. The microbial taxa whose abundance either increased or decreased significantly during remediation were identified. Soil electrical conductivity (EC) was identified as the main factor driving the variation in microbial community composition between the remediated and barren soil, and total nitrogen (TN), total carbon (TC), and available potassium (AK) were the main factors driving the differences among plant species. This report provides new insights into the responses of the root zone microbial communities of different salt-tolerant plant species during phytoremediation.IMPORTANCE Despite knowing that phytoremediation by salt-tolerant plants is an effective technology for ameliorating saline soils and that microorganisms contribute significantly to plant stress tolerance and soil fertility, we still lack a comprehensive understanding of how microbes respond to the growth of salt-tolerant plants and the subsequent decline in soil salinity. The results of this study revealed different response patterns among bacterial, archaeal, and fungal communities and indicated that the decline in archaeal abundance might be a sign of successful remediation of coastal saline soils. The recruitment of specific fungal communities by different plant species indicated the importance of fungi in plant species-specific remediation functions. We also identified the taxa that may play key roles during remediation, and these taxa could potentially be used as indicators of phytoremediation. Overall, these findings highlight the importance of microbes in the phytoremediation of saline soil and suggest that the mechanisms involved are plant species specific.
