Ionic liquids as effective additives to enhance the solubility and permeation for puerarin and ferulic acid.

ABSTRACT: Ionic liquids, especially the cholinium-amino acid-based ionic liquids (CHAAILs), have recently been found to be effective ingredients in formulation of transdermal drug delivery system. In this work, we synthesized six CHAAILs, and investigated their ability to enhance the solubility and permeation of two active pharmaceutic ingredients (APIs), i.e. ferulic acid and puerarin. The solubility measurements showed that a low amount of CHAAILs can significantly increase the solubility of APIs. Moreover, the effective enhancement of permeation of APIs across a polyethersulfone (PES) membrane was achieved at low concentration (4 mg ml^-1) of CHAAILs. It is more worthwhile that the presence of CHAAIL brings much less cytotoxicity as compared to traditional types of ionic liquids. Therefore, CHAAILs can be considered as great potential candidates of green and effective additives in transdermal drug delivery systems.

Choline-Amino Acid Ionic Liquids as Green Functional Excipients to Enhance Drug Solubility.

Project description:The development of effective forms to incorporate poorly soluble drugs into delivery systems remains a problem. Thus, it is important to find alternatives such as finding excipients that increase drug solubility. Ionic liquids (ILs), particularly choline-based ILs, have been studied as solubility enhancers in drug delivery systems. Nonetheless, to acknowledge this property as a functionality, it needs to be proven at non-toxic concentrations. Hence, herein two choline-amino acid ILs were studied as functional excipients by evaluating their influence on the solubility of the poorly water-soluble ferulic acid and rutin, while considering their safety. The solubility of the drugs was always higher in the presence of the ILs than in water. Ionic liquids did not affect the radical scavenging activity of the drugs or the cell viability. Moreover, stable oil-in-water (O/W) emulsions were prepared containing each drug and the ILs, allowing a significantly higher drug loading. Globally, our results suggest that choline-based ILs may act as green functional excipients, since at non-toxic concentrations they considerably improve drug solubility/loading, without influencing the antioxidant activity of the drugs, the cell viability, or the stability of the formulations.

Solubility Studies of Cyclosporine Using Ionic Liquids.

Project description:Six ionic liquids (ILs) were selected based on their chemical and physical properties to study the solubility of cyclosporine A. Of these, cyclosporine exhibited higher room temperature solubility in 1-ethyl-3-methylimidazolium acetate ([C2mim][OAc]) than in acetone, an effective molecular solvent used to solubilize and purify cyclosporine. The solubility of cyclosporine in the ILs dramatically increased at higher temperatures, a critical factor that cannot be varied in a wide range with low boiling molecular solvents. The differences in solubility were explored for cyclosporine purification. Cyclosporine was purified up to ∼93% with n-butylammonium acetate ([C4NH3][OAc]) and could be further purified to 95% using an IL/organic solvent biphasic system. After purification, cyclosporine was recovered as an amorphous solid using the ILs.

Ionogels Derived from Fluorinated Ionic Liquids to Enhance Aqueous Drug Solubility for Local Drug Administration.

Project description:Gelatin is a popular biopolymer for biomedical applications due to its harmless impact with a negligible inflammatory response in the host organism. Gelatin interacts with soluble molecules in aqueous media as ionic counterparts such as ionic liquids (ILs) to be used as cosolvents to generate the so-called Ionogels. The perfluorinated IL (FIL), 1-ethyl-3-methylpyridinium perfluorobutanesulfonate, has been selected as co-hydrosolvent for fish gelatin due to its low cytotoxicity and hydrophobicity aprotic polar structure to improve the drug aqueous solubility. A series of FIL/water emulsions with different FIL content and their corresponding shark gelatin/FIL Ionogel has been designed to enhance the drug solubility whilst retaining the mechanical structure and their nanostructure was probed by simultaneous SAXS/WAXS, FTIR and Raman spectroscopy, DSC and rheological experiments. Likewise, the FIL assisted the solubility of the antitumoural Doxorubicin whilst retaining the performing mechanical properties of the drug delivery system network for the drug storage as well as the local administration by a syringe. In addition, the different controlled release mechanisms of two different antitumoral such as Doxorubicin and Mithramycin from two different Ionogels formulations were compared to previous gelatin hydrogels which proved the key structure correlation required to attain specific therapeutic dosages.

Solubility and solvation of monosaccharides in ionic liquids.

Project description:Herein, solubility experimental data for six monosaccharides, viz.d-(+)-glucose, d-(+)-mannose, d-(-)-fructose, d-(+)-galactose, d-(+)-xylose and l-(+)-arabinose, in four ionic liquids (ILs), at temperatures ranging from 288.2 to 348.2 K, were obtained aimed at gathering a better understanding of their solvation ability and molecular-level mechanisms which rule the dissolution process. To ascertain the chemical features that enhance the solubility of monosaccharides, ILs composed of dialkylimidazolium or tetraalkylphosphonium cations combined with the dicyanamide, dimethylphosphate or chloride anions were investigated. It was found that the ranking of the solubility of monosaccharides depends on the IL; yet, d-(+)-xylose is always the most soluble while d-(-)-fructose is the least soluble monosaccharide. The results obtained show that both the IL cation and the anion play a major role in the solubility of monosaccharides. Finally, from the determination of the respective thermodynamic properties of solution, it was found that enthalpic contributions are dominant in the solubilization process. However, the observed differences in the solubilities of monosaccharides in 1-butyl-3-methylimidazolium dicyanamide are ruled by a change in the entropy of solution.

Fluorine-functionalized ionic liquids with high oxygen solubility.

Project description:Eight fluorine-functionalized ionic liquids were synthesized and the oxygen solubility was compared to commercial ionic liquids without the extra fluorinated chain. The concentration of dissolved oxygen increased with the fluorine content of the alkyl chain, which can be attached either to the cation or the anion. This approach maintains the freedom to design an ionic liquid for a specific application, while at the same time the oxygen solubility is increased.

Project description:The aim of the present study was to determine the antioxidant activity of the aqueous extracts from Lycopodium clavatum, Cetraria islandica and Dipsacus fullonum obtained using aqueous solutions of ionic liquids by the ultrasound-assisted extraction (IL-UAE) method. Triethanolammonium salts [TEAH]+[AA]- of four amino acids of different hydrophobicity - isoleucine - Ile, methionine - Met, threonine - Thr and arginine - Arg, were chosen as ionic liquids, because they are based on natural, bio-renewable raw materials, such as amino acids and contain a pharmaceutically and cosmetically acceptable counterion of triethanolamine. Triethanolammonium salts were synthesized, identified by spectroscopic methods (NMR and FT-IR) and characterized by thermal methods (DSC and TGA). The 2.5% w/v aqueous solutions of triethanolammonium amino acid salts were used as the solvents in combination with ultrasound assisted extraction (UAE). The estimation of antioxidant properties was carried out using the DPPH, FRAP and CUPRAC assays. Total polyphenol content was measured using the reagent Folin-Ciocalteu. The results showed that the use of [TEAH]+[Thr]- or [TEAH]+[Met]- aqueous solutions increased the antioxidant activity of extracts in comparison to that achieved for extracts with pure water. The use of [TEAH]+[Thr]- as an additive for ultrasound-assisted extraction was characterized by obtaining plant extracts with the highest antioxidant potential, even 2.4-fold. The use of the AAIL-UAE method allowed obtaining higher amounts of polyphenols compared to pure water extracts, even 5.5-fold. The used method allowed the extraction of thermosensitive natural compounds, shortened the extraction time and lowered energy consumption.

Synthesis and investigation of phosphorus-free ionic liquids as multifunctional lubricating additives.

Project description:Ionic liquids (ILs) have been extensively studied as lubricants or lubricant additives for two decades by virtue of their unique physical and chemical properties. Here, two phosphorus-free multifunctional protic ILs were synthesized by reacting oleic acid and a dimer acid with an alkyl aromatic amine. Both were completely miscible in various base fluids like mineral oil, polyether, synthetic ester, and polyalpha olefin. Furthermore, no corrosion towards copper strips was found for these additives due to the absence of phosphorus, halogen, and sulfur in their molecular structures. Tribological tests found that they could significantly improve the tribological performance of base oil in a wide range of test conditions. Additionally, due to the presence of an alkyl diphenylamine group, they could considerably enhance the oxidative stability of the base oil. Overall, the facile preparation approach, good solubility, low corrosion, and excellent tribological behavior and antioxidation property make them suitable as multifunctional additives in various lubricants.

Solubility model of metal complex in ionic liquids from first principle calculations.

Project description:A predictive model based on first principles calculations has been proposed to study the solid-liquid equilibria comprising of metal complexes and ionic liquids. The model is based on first principle COSMO calculation followed by post statistical thermodynamical treatment of self-consistent properties of solute and solvent molecules. The metal complex and ionic liquid have been treated as a simple binary mixture. The ionic liquid has been treated here as a single intact molecule. The experimentally observed dual-solute relationship between the ionic liquid and redox active species in presence of a third organic solvent has been established using our model in this work. Within the model, the dual-solute relationship appeared as a simple Gibbs-Duhem relationship between these two species at ambient condition. The dual-solute relationship between the metal complex (V(acac)3, Cr(acac)3 and Mn(acac)3) and ionic liquid ([Tea][BF4]) has been validated by calculating the Gibbs-Duhem relationship, x solute vs. x solvent(IL) and 1/γ solute vs. x solvent(IL) plots. The present model has been applied to a set of ionic liquids, metal complexes and organic solvent (acetonitrile) for which experimental study has been done. The solvation mechanism of the metal complexes in those ionic liquids was obtained using the model. According to our findings, the ionic liquid containing imidazolium cation and [NTf2]- anion is appeared as a suitable solvent for the non-aqueous redox flow cell. We have compared our results with the already reported experimental results where they were available for the non-aqueous solvents.
