Molecular Scissors for Tailor-Made Modification of Siloxane Scaffolds.

ABSTRACT: The controlled design of functional oligosiloxanes is an important topic in current research. A consecutive Si-O-Si bond cleavage/formation using siloxanes that are substituted with 1,2-diaminobenzene derivatives acting as molecular scissors is presented. The method allows to cut at certain positions of a siloxane scaffold forming a cyclic diaminosilane or -siloxane intermediate and then to introduce new functional siloxy units. The procedure could be extended to a direct one-step cleavage of chlorooligosiloxanes. Both siloxane formation and cleavage proceed with good to excellent yields, high regioselectivity, and great variability of the siloxy units. Control of the selectivity is achieved by the choice of the amino substituent. Insight into the mechanism was provided by low temperature NMR studies and the isolation of a lithiated intermediate.

Exhaustive Baeyer-Villiger oxidation: a tailor-made post-polymerization modification to access challenging poly(vinyl acetate) copolymers.

Project description:The discovery of exhaustive (nearly quantitative) post-polymerization modifications (PPM) relies heavily on the efficiency of their corresponding small-molecule protocols. However, the direct translation of existing small-molecule protocols into PPM methods has never been guaranteed due to the intrinsic differences between small-molecule substrates and polymers. Herein, we introduce the direct optimization on polymers (DOP) as a complementary approach to developing exhaustive PPM reactions. As proof of the DOP concept, we present an exhaustive Baeyer-Villiger (BV) post-modification which cannot be accessed by conventional approaches. This user-friendly methodology provides general access to synthetically challenging copolymers of vinyl acetate and more activated monomers (MAMs) including both statistical and narrow-dispersed block copolymers. Furthermore, a scalable one-pot copolymerization/exhaustive BV post-modification procedure was developed to produce such materials showing improved performance over regular PVAc.

Site-Selective Acylations with Tailor-Made Catalysts.

Project description:The acylation of alcohols catalyzed by N,N-dimethylamino pyridine (DMAP) is, despite its widespread use, sometimes confronted with substrate-specific problems: For example, target compounds with multiple hydroxy groups may show insufficient selectivity for one hydroxyl, and the resulting product mixtures are hardly separable. Here we describe a concept that aims at tailor-made catalysts for the site-specific acylation. To this end, we introduce a catalyst library where each entry is constructed by connecting a variable and readily tuned peptide scaffold with a catalytically active unit based on DMAP. For selected examples, we demonstrate how library screening leads to the identification of optimized catalysts, and the substrates of interest can be converted with a markedly enhanced site-selectivity compared with only DMAP. Furthermore, substrate-optimized catalysts of this type can be used to selectively convert "their" substrate in the presence of structurally similar compounds, an important requisite for reactions with mixtures of substances.

Tailor-made thermoplastic elastomers: customisable materials <i>via</i> modulation of molecular weight distributions.

Project description:The ability to change polymer properties has in the past largely been a factor of modulating the molecular weight, molecular weight distribution breadth, crosslinking, or branching. The use of controlled MWD shape has recently emerged as a promising avenue towards modifying polymer properties. Taking advantage of molecular weight distribution shape, we report a simple and efficient approach for tuning material properties in polystyrene-block-polyisoprene-block-polystyrene (SIS) thermoplastic elastomers (TPEs). We find that the skew of the MWD function governs tensile properties and can be used as a handle to predictably vary polymer toughness while reducing energy dissipation.

Zinc Finger Nucleases: Tailor-made for Gene Therapy.

Project description:Genome editing with the use of zinc finger nucleases has been successfully applied to variety of a eukaryotic cells. Furthermore, the proof of concept for this approach has been extended to diverse animal models from Drosophila to mice. Engineered zinc finger nucleases are able to target specifically and manipulate disease-causing genes through site-specific double strand DNA breaks followed by non-homologous end joining or homologous recombination mechanisms. Consequently, this technology has considerable flexibility that can result in either a gain or loss of function of the targeted gene. In addition to this flexibility, gene therapy by zinc finger nucleases may enable persistent long term gene modification without continuous transfection- a potential advantage over RNA interference or direct gene inhibitors. With systemic viral delivery systems, this gene-editing approach corrected the mutant factor IX in models of mouse hemophilia. Moreover, phase I clinical trials have been initiated with zinc finger nucleases in patients with glioblastoma and HIV. Thus, this emerging field has significant promise as a therapeutic strategy for human genetic diseases, infectious diseases and oncology. In this article, we will review recent advances and potential risks in zinc finger nuclease gene therapy.

Tailor-made temperature-dependent thermal conductivity via interparticle constriction.

Project description:Managing heat is a major challenge to meet future demands for a sustainable use of our energy resources. This requires materials, which can be custom-designed to exhibit specific temperature-dependent thermal transport properties to become integrated into thermal switches, transistors, or diodes. Common crystalline and amorphous materials are not suitable, owing to their gradual changes of the temperature-dependent thermal conductivity. We show how a second-order phase transition fully controls the temperature-dependent thermal transport properties of polymer materials. We demonstrate four major concepts based on a colloidal superstructure: (i) control of transition temperature, (ii) width of phase transition regime, (iii) multistep transitions, and (iv) step height of the transition. Most importantly, this unique control over thermal conductivity is only governed by the interparticle constriction, the particle composition, and its mesostructure. Our concept is therefore also applicable to a wide variety of other particulate materials.

Ultrafast and Reproducible Synthesis of Tailor-Made Octacalcium Phosphate.

Project description:Octacalcium phosphate (OCP) has excellent bone formation ability and a good resorption rate as compared to the commercial bone substitutes [e.g., Bio-Oss (Geistlich Pharma AG) and MBCP+ (Biomatlante)], as well as synthesized biomaterials (hydroxyapatite and tricalcium phosphate). The synthesis approach to obtain phase-pure OCP possesses a great challenge due to its complex reaction mechanism and narrow synthesis window. Thus, the current study aimed to overcome the synthesis challenges and to define the precise reaction conditions required for controllable and reproducible synthesis of OCP. Using the principles of the coprecipitation method, novel synthesis protocols were developed ensuring ultrafast synthesis of OCP within minutes. XRD analysis confirmed that phase-pure OCP was obtained. These processing schemes enabled the synthesis of tailor-made OCP with specific surface areas ranging from 16 to 91 m2/g. The synthesized OCPs exhibited plate-like morphology. The interaction of the synthesized OCPs with MC3T3-E1 cells was found to be nontoxic, confirming their cytocompatibility. The synthesis approach developed in this study indicates that challenges such as the reaction volume, stirring rate, and flow rate need to be addressed in the future to upscale the technology.

Enhanced Bioactivity of Tailor-Made Glycolipid Enriched Manuka Honey.

Project description:Glycolipids can be synthetized in deep eutectic solvents (DESs) as they possess low water content allowing a reversed lipase activity and thus enables ester formation. Based on this principle, honey can also serve as a media for glycolipid synthesis. Indeed, this supersaturated sugar solution is comparable in terms of physicochemical properties to the sugar-based DESs. Honey-based products being commercially available for therapeutic applications, it appears interesting to enhance its bioactivity. In the current work, we investigate if enriching medical grade honey with in situ enzymatically-synthetized glycolipids can improve the antimicrobial property of the mixture. The tested mixtures are composed of Manuka honey that is enriched with octanoate, decanoate, laurate, and myristate sugar esters, respectively dubbed GOH, GDH, GLH, and GMH. To characterize the bioactivity of those mixtures, first a qualitative screening using an agar well diffusion assay has been performed with methicillin-resistant Staphylococcus aureus, Bacillus subtilis, Candida bombicola, Escherichia coli, and Pseudomonas putida which confirmed considerably enhanced susceptibility of these micro-organisms to the different glycolipid enriched honey mixtures. Then, a designed biosensor E. coli strain that displays a stress reporter system consisting of three stress-specific inducible, red, green, and blue fluorescent proteins which respectively translate to physiological stress, genotoxicity, and cytotoxicity was used. Bioactivity was, therefore, characterized, and a six-fold enhancement of the physiological stress that was caused by GOH compared to regular Manuka honey at a 1.6% (v/v) concentration was observed. An antibacterial agar well diffusion assay with E. coli was performed as well and demonstrated an improved inhibitory potential with GOH upon 20% (v/v) concentration.

Depolymerisation of Kraft Lignin by Tailor-Made Alkaliphilic Fungal Laccases.

Project description:Lignins released in the black liquors of kraft pulp mills are an underutilised source of aromatics. Due to their phenol oxidase activity, laccases from ligninolytic fungi are suitable biocatalysts to depolymerise kraft lignins, which are characterised by their elevated phenolic content. However, the alkaline conditions necessary to solubilise kraft lignins make it difficult to use fungal laccases whose activity is inherently acidic. We recently developed through enzyme-directed evolution high-redox potential laccases active and stable at pH 10. Here, the ability of these tailor-made alkaliphilic fungal laccases to oxidise, demethylate, and depolymerise eucalyptus kraft lignin at pH 10 is evidenced by the increment in the content of phenolic hydroxyl and carbonyl groups, the methanol released, and the appearance of lower molecular weight moieties after laccase treatment. Nonetheless, in a second assay carried out with higher enzyme and lignin concentrations, these changes were accompanied by a strong increase in the molecular weight and content of β-O-4 and β-5 linkages of the main lignin fraction, indicating that repolymerisation of the oxidised products prevails in one-pot reactions. To prevent it, we finally conducted the enzymatic reaction in a bench-scale reactor coupled to a membrane separation system and were able to prove the depolymerisation of kraft lignin by high-redox alkaliphilic laccase.

Tailor-made RNAi knockdown against triplet repeat disease-causing alleles.

Project description:Nucleotide variations, including SNPs, in the coding regions of disease genes are important targets for RNAi treatment, which is a promising medical treatment for intractable diseases such as triplet repeat diseases. However, the identification of such nucleotide variations and the design of siRNAs conferring disease allele-specific RNAi are quite difficult. In this study we developed a pull-down method to rapidly identify coding SNP (cSNP) haplotypes of triple repeat, disease-causing alleles, and we demonstrated disease allele-specific RNAi that targeted cSNP sites in mutant Huntingtin alleles, each of which possessed a different cSNP haplotype. Therefore, the methods presented here allow for allele-specific RNAi knockdown against disease-causing alleles by using siRNAs specific to disease-linked cSNP haplotypes, and advanced progress toward tailor-made RNAi treatments for triplet repeat diseases.
