Transcriptome profiles of LAM cell models exposed to oleic acid


ABSTRACT: RNA-seq profiling was performed on Tsc2-deficient mouse embryonic fibroblasts (MEFs) and TSC2-deficient 621-101 human angiomyolipoma cells cultured under basal conditions or treated with oleic acid (30 microM) in DMEM supplemented with 1% FBS for 72 hours. Total RNA was extracted using the RNeasy kit (Qiagen) and RNA integrity was assessed with the Agilent 2100 Bioanalyzer (Agilent Technologies). RNA libraries were prepared following standard Illumina TruSeq protocols and sequenced. Base calling was performed using Illumina Casava 2.20 software. Raw sequencing reads were processed to remove adapter sequences and low-quality or low-complexity regions, followed by alignment to the mouse (mm10) or human (hg19) reference genomes using STAR (v2.7.10a). The resulting SAM/BAM files were processed with SAMtools (v1.18), and read quantification was performed using BEDTools multicov (v2.31.0). Gene expression levels were calculated as transcripts per million (TPM). The submitted dataset includes TPM gene expression matrices.
