The identification of 3,3', 5,5',-tetraiodothyroformic acid within the rat liver.

ABSTRACT: Homogenized rat livers were extracted with organic solvents and the extracts converted into more readily volatile derivatives (trimethylsilylmethyl esters and permethyl esters). Combined g.l.c.-mass-spectrometric analysis identified the presence of tetraiodothyroformate. It is postulated that mitochondrial beta-oxidation of tetraiodothyropyruvate results in the formation of this apparently undescribed metabolite.

High Oxygen Barrier Polyester from 3,3'-Bifuran-5,5'-dicarboxylic Acid.

Project description:An exceptional oxygen barrier polyester prepared from a new biomass-derived monomer, 3,3'-bifuran-5,5'-dicarboxylic acid, is reported. When exposed to air, the furan-based polyester cross-links and gains O2 permeability 2 orders of magnitude lower than initially, resulting in performance comparable to the best polymers in this class, such as ethylene-vinyl alcohol copolymers. The cross-links hinder the crystallization of amorphous samples, also rendering them insoluble. The process was observable via UV-vis measurements, which showed a gradual increase of absorbance between wavelengths of 320 and 520 nm in free-standing films. The structural trigger bringing about these changes appears subtle: the polyester containing 5,5'-disubstituted 3,3'-bifuran moieties cross-linked, whereas the polyester with 5,5'-disubstituted 2,2'-bifuran moieties was inert. The 3,3'-bifuran-based polyester is effectively a semicrystalline thermoplastic, which is slowly converted into a cross-linked material with intriguing material properties once sufficiently exposed to ambient air.

Project description:The mol-ecular structure of 5,5'-bis(-hydroxy-methyl)-3,3'-biisoxazole, C8H8N2O4 (1), is composed of two trans planar isoxazole rings [r.m.s deviation = 0.006 (1) Å], each connected with a methyl hydroxyl group. Similarly, the structure of 4,4',5,5'-tetrakis-(hydroxy-methyl)-3,3'-biisoxazole, C10H12N2O6 (2), is composed of two planar isoxazole rings [r.m.s. deviation = 0.002 (1) Å], but with four hydroxymethyl groups as substituents. Both mol-ecules sit on a center of inversion, thus Z' = 0.5. The crystal structures are stabilized by networks of O-H⋯N [for (1)] and O-H⋯O hydrogen-bonding inter-actions [for (2)], giving rise to corrugated supra-molecular planes. The isoxazole rings are packed in a slip-stacked fashion, with centroid-to-centroid distances of 4.0652 (1) Å for (1) (along the b-axis direction) and of 4.5379 (Å) for (2) (along the a-axis direction).

3,3',5,5'-Tetra-nitro-biphen-yl.

Project description:The title compound, C(12)H(6)N(4)O(8), is a biphenyl system that was synthesized as a building block for a new series of anti-malarial compounds. The aromatic rings are oriented at a dihedral angle of 45.5?(2)°, and inter-molecular short O?O contacts form a chain along the b axis. The strength of the inter-actions involved in this chain cause one of the rings to be slightly distorted, with the torsion angle between the nitro groups being 23.4?(2)°, whereas, in the other ring, both nitro systems are parallel, forming an angle of 9.6?(2)° with the plane of the aromatic ring to which they are bound. Furthermore, the three ring C atoms around the ring-ring linkage belong to a plane inclined by 4.5?(1)° in relation to the plane containing the other three C atoms, i.e. (NO(2)-)C-C-C(NO(2)). This distortion of the ring causes uncommonly short intermolecular O?O [3.038?(2)?Å] and O?C [3.000?(4) and 3.214?(1)?Å] contacts.

Project description:The title compound, C(8)H(2)Br(4)S(2), was prepared by bromination of 2,2'-bithio-phene with bromine. The mol-ecule is located on a crystallographic twofold rotation axis, thereby imposing equal geometry of the two thio-phene rings. Each five-membered ring is planar [maximum deviation 0.011 (9) Å] and the dihedral angle between the planes through the rings is 47.2 (4)°. The mol-ecules are arranged to minimize intramolecular contacts between the 3-3' and 5-5'-bromine atoms.

Identification of lipidomic markers of chronic 3,3',4,4',5-pentachlorobiphenyl (PCB 126) exposure in the male rat liver.

Project description:Exposure to PCB 126, an environmentally relevant aryl hydrocarbon receptor agonist, is an environmental factor causing hepatic steatosis in rodent models; however, the lipidome of PCB 126-exposed rats has not been investigated in-depth. The objective of the present study was therefore to characterize dose-dependent changes in the lipid profile in the liver of male Sprague-Dawley rats exposed to PCB 126. Rats were exposed for three month to intraperitoneal injections of 0.01, 0.05 and 0.2μmol/kg bw PCB 126 in corn oil. Control animals were exposed in parallel and received corn oil alone. Lipids were extracted from whole liver homogenate and levels of polar lipids and fatty acids incorporated into triglycerides (FATAGs) were determined with tandem mass spectrometry using electrospray ionization. PCB 126 exposure increased the hepatic content of polar lipids and FATAGs. Protein adjusted levels of several polar lipid classes, in particular phosphatidylserine levels, decreased, whereas FATAGs levels typically increased with increasing PCB 126 dose. Sensitive, dose-dependent endpoints of PCB 126 exposure included an increase in levels of adrenic acid incorporated into triglycerides and changes in levels of certain ether-linked phospholipid and 1-alkyl/1-alkenyldiacylglycerol species, as determined using partial least square discriminant analysis (PLS-DA) and ANOVA. These changes in the composition of polar lipids and fatty acid in the liver of PCB 126 exposed rats identified several novel markers of PCB 126-mediated fatty liver disease that need to be validated in further studies.

In the crystal structure, the anions and water mol-ecules are linked into sheets parallel to the bc plane by inter-molecular O-H⋯O hydrogen bonds and sustained by C-H⋯O contacts. The linking of the anions and water mol-ecules with the cations by N-H⋯O hydrogen bonds creates a three-dimensional extended network. The crystal structure is further stabilized by very weak C-H⋯π inter-actions.

Project description:In the title compound, C(10)H(12)N(6), the two 5,6-dimethyl-1,2,4-triazine halves of the mol-ecule are related by a centre of symmetry. The two triazine rings are coplanar to within a maximum deviation of 0.013 (2) Å from the mean plane of the ring atoms. In the crystal, mol-ecules form layers parallel to the (100) crystallographic plane. Adjacent layers are held together via a C-H⋯π inter-action involving mol-ecules related by an a-glide plane.

One tert-butyl group is disordered equally over two positions.
