Project description:BackgroundBioactive peptides can prevent damage associated with oxidative stress in humans when consumed regularly. Recently, peptides have attracted immense interest because of their beneficial functional properties, safety and little or no side effects when used at high concentration. Most antioxidant peptides are small in size, less than 1 kDa, and contains a high proportion of hydrophobic amino acid. Particularly, tyrosine, leucine, alanine, isoleucine, valine, lysine, phenyalanine, cysteine, methionine and histidine in peptide chain exhibited high antioxidant activity. Mungbean meal protein (MMP) is highly abundant in hydrophobic amino acids. It indicated that MMP might be a good source of antioxidants. Therefore, the objectives were to optimize the conditions used to generate mungbean meal protein hydrolysate (MMPH) with antioxidant activity from bromelain and to investigate the antioxidant activities of different molecular weight (MW) peptide fraction.MethodsResponse Surface Methodology (RSM) was used for screening of the optimal conditions to produce MMPH. After that MMPH was fractionated using ultrafiltration membranes with different MW distributions. Crude-MMPH and four fractions were investigated for five antioxidant activities: 2,2,1-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl, superoxide, ferric reducing antioxidant power (FRAP) and metal ion chelation activity.ResultsThe optimal condition to produce the MMPH was 15% (w/w) of bromelain and hydrolysis time for 12 h which showed the greatest DPPH and ABTS radical scavenging activity. After mungbean protein from optimal condition was separated based on different molecular weight, the DPPH radical scavenging activity was the highest for the F4 (less than 1 kDa) peptide fraction. Metal ion chelating activity was generally weak, except for the F4 that had a value of 43.94% at a protein concentration of 5 mg/mL. The F4 also exhibited high hydroxyl and superoxide activities (54 and 65.1%), but moderate activity for ferric reducing antioxidant power (0.102 mmole Fe2+/g protein) compared to other peptide fractions and crude-MMPH. Molecular weight and amino acid were the main factors that determined the antioxidant activities of these peptide fractions. Results indicated that F4 had strong antioxidant potentials.DiscussionThe lowest MW fraction (less than 1 kDa) contributed to the highest DPPH, superoxide, hydroxyl and metal chelation activity because influence of low MW and high content of hydrophobic amino acid in peptide chain. Results from this study indicated that MMPH peptides donate protons to free radicals because they had significantly high DPPH value compared to superoxide, hydroxyl and FRAP, which reactions were electron donation. Moreover, MMPH peptides had the ability to inhibit transition metal ions because of highly abundant glutamic acid and aspartic acid in peptide chain.

Project description:In this study, various techniques, including X-ray diffraction (XRD), high-resolution transmission electron microscopy (HRTEM), scanning electron microscopy (SEM), energy-dispersive X-ray spectrometry (EDS) mapping, X-ray photoelectron spectroscopy (XPS), and water-contact-angle goniometry (WCAG), were used to characterize the crystalline structure and morphological properties of terbium-doped cerium magnesium aluminate (Ce0.67Tb0.33MgAl11O19 or CMAT) in powder form. The results demonstrated that CMAT was successfully synthesized with a particle size of less than 5 µm and a fully evident distribution of elements, as revealed by the SEM images. This was further confirmed by the XRD and HRTEM images. XPS analysis confirmed the presence of all necessary components in CMAT. Additionally, WCAG results showed that the contact angle of CMAT was more hydrophilic with a value of 8.4°. To evaluate its performance, CMAT particles were dispersed in a Polyethersulfone (PES) solution and used to modify a PES ultrafiltration membrane through a phase-inversion method. The resulting membranes were characterized by SEM, atomic force microscopy (AFM), thermogravimetric analysis (TGA), WCAG, and permeability performance and fouling experiments. The addition of CMAT to the PES membranes did not have a significant effect on the structure of the SEM images of the top layer and cross-section of surface properties. However, increasing the concentration of CMAT improved the membrane surface roughness in AFM, and the modified membranes had the ability to resist fouling. The addition of CMAT did not lead to significant energy loss, indicating that the heat flux loss observed can indeed be explained by the amount of C-OH on the PES membrane's surface. The contact angle of the membranes became more hydrophilic with increasing concentration of CMAT from PES G0 to PES G7. The PES origin membrane showed a higher permeation than the membranes mixed with CMAT, and the modified membranes with CMAT displayed significant fouling resistance.

Project description:Chemical reaction of main polymer and additive with oxidative cleaning agents plays an important role in aging of polymeric membrane for water and wastewater treatment. As a green and powerful oxidant, hydrogen peroxide (H2O2) can achieve good cleaning efficacy under alkaline condition, but its influence on membrane aging was poorly understood. In this study, degradation of polyethersulfone (PES) membrane due to H2O2 exposure under alkaline condition (pH 9 and 11) was holistically investigated by humic acid (HA) filtration experiments and multiple membrane characterization techniques, with sodium hypochlorite (NaClO) aging examined as a comparison. Membrane permeability and HA retention rate was hardly changed by H2O2 aging at an exposure dose of 500 g·h/L, whereas NaClO aging led to substantial increase of membrane permeability and significant decrease of retention ability. Meanwhile, H2O2 aging slightly increased fouling propensity with HA filtration, while NaClO aging resulted in more serious fouling. ATR-FTIR and XPS analysis revealed much less degradation of PES and hydrophilic additive by H2O2 than that by NaClO, and membrane morphology and surface properties were characterized to explain the variation of filtration performance. Overall, compared with cleaning with NaClO, membrane degradation can be minimized by cleaning with H2O2.

Project description:Self-cleaning, antimicrobial ultrafiltration membranes are urgently needed to alleviate the low flux problems caused by membrane fouling in water treatment processes. In this study, in situ generated nano-TiO2 MXene lamellar materials were synthesized and then 2D membranes were fabricated using vacuum filtration. The presence of nano TiO2 particles as an interlayer support layer widened the interlayer channels, and also improved the membrane permeability. The TiO2/MXene composite on the surface also showed an excellent photocatalytic property, resulting in enhanced self-cleaning properties and improved long-term membrane operational stability. The best overall performance of the TiO2/MXene membrane at 0.24 mg cm-2 loading was optimal, with 87.9% retention and 211.5 L m-2 h-1 bar-1 flux at a filtration of 1.0 g L-1 bovine serum albumin solution. Noticeably, the TiO2/MXene membranes showed a very high flux recovery under UV irradiation with a flux recovery ratio (FRR) of 80% as compared to the non-photocatalytic MXene membranes. Moreover, the TiO2/MXene membranes demonstrated over 95% resistance against E. coli. And the XDLVO theory also showed that the loading of TiO2/MXene slowed down the fouling of the membrane surface by protein-based contaminants.

Project description:Freshwater biofilms assemble from a pool of rare water column genotypes. Random density fluctuations and temporal species turnover of functionally equivalent potential colonizers result in compositional variability of newly formed biofilm communities. We hypothesized that stronger environmental filtering as induced by enhanced substrate levels might reduce the impact of a locally variable pool of colonizers and instead select for more universal habitat specialists. Our model were heterotrophic biofilms that form on membranes during gravity-driven ultrafiltration of lake water. In four separate experiments, biomass of the cyanobacterium Microcystis was added to the feed water of one set of treatments (BM) and the resulting biofilm communities were compared to unamended controls (CTRL). Biomass addition led to a significant shift of community assembly processes: Replicate BM biofilms were more similar to each other than by chance in 3 of 4 experiments, whereas the opposite was the case for CTRL communities. Moreover, BM communities were more stochastically assembled across experiments from a common 'regional' pool of biofilm colonizers, whereas the composition of CTRL communities was mainly determined by experiment-specific 'local' genotypes. Interestingly, community assembly processes were also related to both, physiology (aerobic vs. anaerobic lifestyle) and the phylogenetic affiliation of biofilm bacteria.

Project description:In this study, novel composites were produced by blending partially cyclized polyacrylonitrile (cPAN) and poly(amide-imide) (PAI) in N-methylpyrrolidone in order to fabricate asymmetric membranes via phase inversion method. The compatibility of PAI and cPAN through possible intermolecular interaction was examined by quantum chemical calculations. The composite membranes were characterized by FTIR, SEM, contact angle measurements, etc. A considerable reduction in the contact angles of water and ethylene glycol (EG) was observed after adding cPAN to the PAI membrane, which is evidence of improved membrane hydrophilicity. Membrane transport properties were investigated in ultrafiltration tests by measuring the pure water flux, rejection of proteins, and flux recovery ratio (FRR). The best properties were found for the membrane containing 5 wt% cPAN; an increase in BSA rejection and a remarkable increase in FRR were observed, which can be explained by the hydrophilization of the membrane surface provided by the presence of cPAN.

Project description:Gravity-driven membrane (GDM) ultrafiltration systems require little maintenance: they operate without electricity at ultra-low pressure in dead-end mode and without control of the biofilm formation. These systems are already in use for water purification in some regions of the world where adequate treatment and distribution of drinking water is not readily available. However, many water bodies worldwide exhibit harmful blooms of cyanobacteria that severely lower the water quality due to the production of toxic microcystins (MCs). We studied the performance of a GDM system during an artificial Microcystis aeruginosa bloom in lake water and its simulated collapse (i.e., the massive release of microcystins) over a period of 21 days. Presence of live or destroyed cyanobacterial cells in the feed water decreased the permeate flux in the Microcystis treatments considerably. At the same time, the microbial biofilms on the filter membranes could successfully reduce the amount of microcystins in the filtrate below the critical threshold concentration of 1 µg L(-1) MC for human consumption in three out of four replicates after 15 days. We found pronounced differences in the composition of bacterial communities of the biofilms on the filter membranes. Bacterial genera that could be related to microcystin degradation substantially enriched in the biofilms amended with microcystin-containing cyanobacteria. In addition to bacteria previously characterized as microcystin degraders, members of other bacterial clades potentially involved in MC degradation could be identified.

Project description:We developed novel Ti-Zr-Be-Co bulk metallic glasses through Co addition based on a ternary Ti45Zr20Be35 alloy. By altering the alloying routes and alloying contents, the influence of Co alloying on glass-forming ability, thermal stability, thermoplastic formability, crystallization behavior, and corrosion resistance has been investigated systematically. It was found that the best alloying route for enhancing the glass-forming ability, thermoplastic formability, compressive plasticity, and corrosion resistance is to replace Be by Co. Ti45Zr20Be23Co12 possesses the largest critical diameter of 15 mm for glass formation. Ti45Zr20Be27Co8 possesses the highest thermoplastic formability which is comparable to that of Vitreloy alloys. Ti45Zr20Be25Co10 exhibits the largest room temperature plasticity of 15.7% together with a high specific strength of 3.90 × 105 Nm/kg. The addition of Co also strongly affects the crystallization behavior of the base alloy, resulting in a more complex crystallization process. The corrosion resistance of Ti-Zr-Be alloy in 1 mol/L HCl solution can also be enhanced by Co alloying. The related mechanisms have been explained in detail, which provide guidance for the composition design of Ti-based metallic glasses with improved properties.

Project description:Coking nanofiltration (NF) concentrates, as typical wastewater with high salinity and refractory organics, have become one of the greatest challenges for "near-zero emission" processes. In our study, an advanced oxidation technology based on ferrous iron/persulfate (Fe(ii)/PS) and polyferric sulfate (PFS) coagulation coupled with ultrafiltration (UF) was used to treat NF concentrates and mitigate membrane fouling. Based on batch experiments, the optimal parameters of Fe(ii)/PS were obtained, during which we discovered that the slow reaction stage of total organic carbon (TOC) removal followed first-order degradation kinetics. Under the optimal reaction conditions, Fe(ii)/PS could efficiently mineralize 69% of organics in coking NF concentrates. In order to eliminate the iron floc generated in the Fe(ii)/PS step, a small amount of PFS (0.05 mM) was added to coagulate the iron floc, which could further improve the effluent quality so that the turbidity, iron content and TOC were significantly reduced by 79.18%, 98% and 21.79% respectively. Gas chromatography coupled with time-of-flight mass spectrometry (GC × GC-TOFMS) and fluorescence excitation-emission matrix spectrometry (EEM) were performed to characterize the removal of phenols, PAHs, quinolines and humic acids in NF concentrates which were responsible for UF membrane fouling. Moreover, scanning electronic microscopy (SEM) and atomic force microscopy (AFM) were conducted to study the surface of the UF membrane after treatment of NF concentrates. The result exhibited that the organic pollutants deposited on the UF membrane surface were reduced by Fe(ii)/PS-PFS pretreatment, and UF membrane flux was thus enhanced. Our results show the potential of the approach of applying Fe(ii)/PS-PFS-UF in NF concentrate treatment.
