Study of Glutamine as Prophylaxis for Irinotecan Induced Diarrhea

ABSTRACT: Irinotecan (7-ethyl-10- {4(-1-piperidino)-1-piperidino} carbonyloxy camptothecin) is a semisynthetic camptothecin derivative introduced in the 1980’s. Irinotecan is a prodrug metabolized by carboxylesterases to an active metabolite 7-ethyl-10-hydroxy-camptothecin (SN38). SN38 exerts its cytotoxic effect by forming stable complexes with topoisomerase I and DNA. These complexes collide with replication forks and cause breaks in DNA. Studies substantiated irinotecan’s activity in 5FU resistant colorectal cancer and led to its approval for treatment of 5FU resistant colorectal cancer in the United States, Canada and Europe.Colorectal cancer studies demonstrated that single agent irinotecan’s dose limiting toxicity was diarrhea occurring 5 to 6 days after its administration. High dose loperamide at first occurrence of diarrhea has decreased the incidence of diarrhea but the incidence of grade 3/4 diarrhea remains high at 28 to 40%.

CD133+CD24lo defines a 5-Fluorouracil-resistant colon cancer stem cell-like phenotype

Project description:Metastatic human colon carcinoma cell lines LS411N and SW620 were cultured in the presence of increased concentration of 5-FU. The selected stable cell lines (LS411N-5FU-R and SW620-5FU-R) are CD133+ that are resistant to 5-FU. However, FACS-sorted CD133+ cells from LS411N and SW620 are not resistant to 5-FU, suggesting that only a subset of CD133+ cells are 5-FU-resistant colon cancer stem cells. A global gene expression profiling was performed to identify differentiated expressed genes between LS411N-CD133+ cells and LS411N-5FU-R, and between SW620-CD133+ and SW620-5FU-R cells. These differentially expressed genes are potentially responsible for the colon cancer stem cell phenotypes and chemoresistance.

Reversible 5-formyluracil DNA-histone crosslinks: formation and biological effects

Project description:5-Formyluracil (5fU) is a vital DNA modification that widely presents in cells and organisms. A unique feature of 5fU is the presence of a potentially reactive aldehyde group in its structure, which can realize addition and condensation reactions. However, the biological function details of 5fU are still largely elusive; especially the relationship with protein is still unclear. Herein, we have first showed that 5fU bases have strong affinity with proteins and can yield reversible DNA-protein conjugates with nuclear proteins even under certain spatial space. Besides, 5fU can influence physical properties of nucleosome even chromatin by increasing nucleosome occupancy and stability. What’s more, we firstly confirmed the existence of 5fU-histone crosslinking in thyroid carcinoma tissue by the CHIP of histone H4 combined with LC-MS. These results suggesting the DNA−protein crosslinks (DPC) formation between 5fU and nucleosome core particles may play a potential role in vivo, such as influencing DNA transactions, altering chromatin structure, and stabilizing the nucleosome. In addition, the Schiff base resulted by 5fU may provide us a new direction for capture even quantification of this modification nucleobase based on this condensation reaction with proteins for further epigenetic studies.

Verticillin A is a selective HMTase inhibitor that inhibit H3K9 methylation to overcome chemoresistance of human colon carcinoma

Project description:We have determined that verticillin A is a histone methyltransfease inhibitor that selectively inhibits human SUV39H1, SUV39H2, G9a and GLP to inhibit H3K9 methylation in human colon cancer cells. The objective here is to identify verticillin A target genes in human colon cancer cells. 5-FU-resistant human colon carcinoma cell lines LS411N-5FU-R was established from the metastatic human colon carcinoma cell line LS411N. LS411N-5FU-R cells were cultured in the absence or presence of verticillin A (50 nM) for 3 days and used for RNA lsolation. DNA microarray analysis was done in replicates in the Georgia Regents University Cancer Center Genomic Core Facility.

Project description:We have established a neuroblastoma model resistant in vivo to the topoisomerase I inhibitor irinotecan (CPT-11, Camptothecin-11, CAMPTOSAR®) in order to study resistance to these agents acquired in a therapeutic setting. Common mechanisms of resistance were not involved in this model and we thus explored the gene expression signature associated with resistance by comparing the transcriptomes of sensitive, resistant, and reverted tumours using high-throughput DNA microarrays.

Transcriptomic analysis of sturgeon embryos in response to chemically induced DNA damage

Project description:In this study we show that the exposure to genotoxicants (10 nM camptothecin and 20 µM Olaparib) affects embryo development in a stage-dependent manner with early stages being the most sensitive to DNA damage. The transcriptomic and proteomic patterns changed significantly after exposure to genotoxicants compared to the control. Camptothecin and Olaparib downregulated metabolic pathways and developmental processes. Additionally, the exposure to genotoxicants led to upregulation of cell proliferation, neural precursor cell proliferation, generation of neurons, taxis and locomotion. Protein patterns were altered as well by DNA damage. The results of this study indicate that there is a strong correlation between phenotype formation and changes in transcriptomic and proteomic profiles.

(2-Benzimidazolyl)acetonitrile derivative for the highly selective la-beling 5-formyluracil and its application in genome-wide mapping in tissues

Project description:Natural chemical modifications to 5-formyluracil (5fU) has been identified as an important oxidative nucleobase modification, is present in the genomes of many cell types. However, its distribution in genome is poorly understood. Herein, we present a selec-tive labeling and pull-down strategy termed fU-Seq for genome-wide 5fU profiling in mouse hippocampus and human thyroid carcinoma tissues. The mainly essence is using an azido modified (2-benzimidazolyl)acetonitrile derivative that selectively labels 5fU. Surprisingly, 5fU is highly enriched at intergenic and intron, and 5fU-rich regions are positively correlated with H3K27me3 and negatively correlated with H3K27ac, indicating that 5fU is associated with transcription. Collectively, the sequencing of 5fU allow comprehensive analysis of 5fU in genome and provide effective technology for mapping and paves the way for addressing biological and chemical questions regarding this nucleobase modification and for revealing its potential roles in epigenetics.
