Phase II study of PM60184 in patients with advanced colon-rectal cancer after standard treatment.

ABSTRACT: Primary objectives: To evaluate the efficacy of PM060184 in terms ofprogression-free survival at 12 weeks (PFS3) in patients withadvanced colorectal carcinoma (CRC) after standard therapy. Primary endpoints: PFS3, defined as the percentage of patients remaining aliveand progression-free at Week 12 (Month 3) after the firsttreatment dose.

Project description:Adipose, liver, and skeletal muscle tissue from 6-month and 12-month CC-RIX cohorts underwent transcriptomic profiling. CC-RIX cohorts were exposed to sensitizer (HFHS diet) and a widely used pharmaceutical intervention (metformin). Chow (control) or high fat high sugar (HFHS) diet started at 6 weeks of age.. At six months of age, a subset of mice from each chow group and of each sex were euthanized for tissue collection. At 7 months of age, half of the remaining mice in each diet treatment group will receive metformin treatment. In a subset of the 12 month cohort animals, metformin (5 mg/mL) was delivered ad libitum in the drinking water (Chang, Moro et al. 2018). Terminal phenotyping and tissue collection was done at 12 months of age.

IQGAP2 knockout model of hepatocellular carcinoma

Project description:Whole body knockout mice lacking IQ-motif containing GTPase-activating protein 2 (IQGAP2) develop spontaneous hepatocellular carcinoma (HCC) at the age of 12 months and older (Schmidt et al., 2008). Hepatic transcript expression profiles were obtained for IQGAP2 knockout and wild-type control mice of two age groups, 6- and 24-month-old. Liver samples from 24-month-old IQGAP2 knockout mice were HCC tumors, livers from all other groups were tumor-free. Results provide insights into the potential role of IQGAP2 as a liver-specific tumor suppressor. Transcript profiles of four groups of mouse livers (N = 3 in each group) were compared using Affymetrix GeneChip® Mouse Genome 430 2.0 Array. The groups included livers from 6- and 24-month-old wild-type (WT) mice and 6- and 24-month-old (KO) Iqgap2-/- mice.

Project description:1 day-, 14-day-, 8-week-, 8-month- and 12-month-old mice ovaries were collected for single-cell RNA sequencing. The developmental stage-specific somatic cells were defined and analyzed with the dynamics of cell-cell communication networks to reveal their association with ovarian development and senescence. These results provide strong support for further research on the supporting role and regulation mechanism of somatic cells to follicles.
