Global proteome changes in liver tissue 6 weeks after FOLFOX treatment of colorectal cancer liver metastases.

Project description:Study: Oxaliplatin-based chemotherapy for colorectal liver metastasis is associated with sinusoidal injury of liver parenchyma and may result in increased morbidity after liver surgery. Preclinical and human liver parenchyma studies have proposed several pathways of oxaliplatin-induced injury, but the effects on protein level remains unknown. Protein expression in liver tissue from eight patients treated with FOLFOX and seven controls without treatment was analyzed by label-free liquid chromatography mass spectrometry. Conclusion: Six weeks after FOLFOX treatment less than 1% of identified proteins showed changes in expression. The changes were associated with DNA replication, cell cycle entry and innate immune response. We hypothesize that the changes are residual after recovery from FOLFOX treatment injury.

Project description:Colorectal Cancer (CRC) is one of most common cancers in the world and a main treatment in postoperative chemotherapy is oxaliplatin and fluorouracil (FOLFOX), But the effect is different among CRC patients. In this study, LC-MS/MS strategy was used to profile the plasma proteome in FOLFOX benefit and futile group. As a result, a panel of plasma proteins by machine learning from our data was verified for a possible prediction tool in postdiagnosis.

Project description:This experiment is aimed at studying the effect of two different chemotherapies (FOLFOX and FOLFIRI) on colorectal cancer cell lines (HCT116 and HT29) at different time points. Two early time points (8h and 16h) are used to evaluate the acute response, while a late time point (two weeks, LT) is used to evaluate the long-term effect. After two week exposure, cells are grown without treatments for additional two weeks, to evaluate the transcriptomic of cells which survived the chemotherapy. A pool of the same cells has been independently profiled by scGET-seq.

Project description:Interventions: Patients of stage 3a recieved UFT+l-LV( day1-28) and autologous tumor lysate-pulsed dendritic cells (day8,22) every 2 weeks.This treatment repeats 6 courses, or recieved capecitabine (day1-14) and autologous tumor lysate-pulsed dendritic cells (day8) every 2 weeks.This treatment repeats 5 courses, Patients of 3b and/or 4 received FOLFOX or FOLFOX+ bevacizumab (day1-3) and autologous tumor lysate-pulsed dendritic cells (day8) every 2 weeks. This treatment repeats 6 courses. Primary outcome(s): Safety Study Design: Single arm Non-randomized
