Sedation of children prior to the surgery using nebulization of two doses of a same drug called dexmedetomidine to reduce parental separation anxiety.

ABSTRACT: Intervention1: Group B - 39 patients, 3 microgram/kg dexmedetomidine mixed in normal saline to make a total volume of 3 ml.: Group B will receive nebulisation with dexmedetomidine at a dose 3mcg/kg diluted with 0.9% normal saline to a total volume of 3 ml with a nebuliser face mask and a continuous flow of 100% oxygen at 6 L/min for 10 min once before induction of anaesthesia in sitting position. Control Intervention1: Group A - 39 patients, 2 microgram/kg of dexmedetomidine mixed in normal saline to a total volume of 3 ml.: Group A will receive nebulisation with dexmedetomidine at a dose 3mcg/kg diluted with 0.9% normal saline to a total volume of 3 ml with a nebuliser face mask and a continuous flow of 100% oxygen at 6 L/min for 10 min once before induction of anaesthesia in sitting position. Plasma Proteome Dynamics in the Pig Model of Trauma

Project description:The effect of IV-administered Vitamin C on the plasma proteome in the pig trauma model was investigated with label free proteomics. Plasma samples were drawn from 9 male Sinclair swine at baseline, post-trauma, and 0.25, 2, and 4 hours post resuscitation with 500 mL of hydroxyethyl starch. Swine were randomized to receive either saline, low-dose VitC (50 mg/kg), or high-dose VitC (200 mg/kg) during volume resuscitation (N = 3/treatment). High performance LC-MS/MS proteomics was then used to quantitatively measure plasma protein levels over time.

Brief Hydrocortisone Infusion Differentially Influences Blood Leukocyte and Monocyte Gene Expression Before and After Endotoxin Administration in Humans (supported by NIH grant GM34695)

Project description:Fourteen human subjects (18-40 years of age) were studied. The control group (saline + LPS [n = 7]) received a 12 hour infusion of saline and the experimental group (cortisol + LPS [n = 7]) received a 12 hour infusion of cortisol (3 µg/kg/min). After 6 hours of saline or cortisol infusion, intravenous lipopolysaccharide (2 ng/kg) was administered to all subjects. Blood samples were collected at -24 hours, 0 hours, 6 hours and 24 hours relative to LPS administration at 0 hours. Leukocytes and monocytes were isolated from these blood samples, total RNA extracted, and microarray analyses performed using Affymetrix Focus GeneChips®.

RNA sequencing to detect the differentially expressed genes in AngII infused mice after Ethoxysanguinarine, Baicalin, Gastrodin or Valsartan treatment

Project description:The goals of this study is to identify the differential expressed genes in cardiac tissue of C57BL/6 mice with or without Angiotensin II (AngII) treatment, and compare the differential expressed genes in the cardiac tissue of Ang II infused C57BL/6 mice after Ethoxysanguinarine (ETH), Baicalin (BAI), Gastrodin (GAS) or valsartan (VAL) treatment. Briefly, the mice (n=30) were randomly divided into 6 groups: control, AngII, AngII + ETH, AngII + BAI, AngII + GAS and AngII + VAL groups (n=5 for each group). Mice in Control and AngII groups were infused with saline and 500 ng/kg/min of AngII respectively, and orally administrated with saline; the mice in AngII + ETH, AngII + BAI and AngII + GAS groups were infused with AngII (500 ng/kg/min) and orally administrated with 5 mg/kg /day of ETH, BAI or GAS daily for total 4 weeks. The mice in AngII + VAL group were infused with AngII (500 ng/kg/min) and orally administrated with 10.4mg/kg /day of VAL. Then the cardiac tissues were used to identify differentially expressed genes among different groups.
