Clinical and molecular differentiation between malignant rhabdoid tumor of normal tissue: A two-case report

Project description:Preoperative B-scan ultrasonography and Computed Tomography (CT) examination in 2 cases suggested that nephroblastoma . Treated with radical nephrectomy after general anesthesia. After operation,MRTK diagnosis by pathological examination. Regular chemotherapy 7 courses(Child 1) and 12 courses(Child 2),followed up for 2 years(Child 1) and 3 years and 1 month(Child 2), without symptoms. RNA-seq results showed 2203 differential genes (DEGs) in the kidney tissue of children with MRTK compared to normal tissue. GO analysis suggested that most DEGs participate in protein binding. KEGG results showed that the DEGs were mainly involved in the PI3K-Akt signaling pathway and microRNA-related proteins.

Tandem mass tag-based proteomic analysis in saliva of horses with acute abdominal disease

Project description:The objective of the study was to analyze the changes in proteomics that occur in saliva in acute abdominal disease (AAD) by proteomics in order to identify new proteins that could allow gaining knowledge about the physiopathological changes that can occur in this fluid and identify possible new biomarkers of acute abdominal disease. Samples from two groups of horses were analyzed by TMT proteomic analysis: - Diseased horses (n=7; 4 stallions and 3 mares; mean age = 8 years (range 1-18); Andalusians (n=5) and crossbred (n=2)). Presence of AAD was diagnosed based on history, physical examination (abdominal auscultation, rectal examination and nasogastric intubation) and additional diagnostic tests incl. complete blood count (CBC), serum biochemistry profile, abdominal ultrasound and or abdominocentesis. Final diagnoses were impaction of ascending colon (n=3), right dorsal colon displacement (n=2) and spasmodic colon (n=2). - Healthy horses (n=6, 3 geldings, 2 stallions and 1 mare; mean age = 12 years (range 4-15); Andalusians (n=4) and crossbred (n=2)). Horses were found healthy based on history, clinical examination, CBC and serum biochemistry profile. These procedures were approved by the ethical committee of the University of Murcia (CEEA 288/2017).

Expression data from the CTCR-OV01 study

Project description:All patients with suspected ovarian cancer (Raised CA 125 and a complex pelvic mass in a perimenopausal woman) were radiologically staged using CT scan and a chest x-ray. Patients with evidence of intra-abdominal metastasis and/or malignant pleural effusion were approached for entry to the study. Tissue biopsy was obtained either under radiological control (core needle biopsy) or via laparoscopic surgery (punch biopsy). Patients with histologicaly confirmed epithelial ovarian cancer were randomized to receive either three cycles of carboplatin (AUC 7) or paclitaxel (175 mg/m2). Patients with suspected ovarian cancer had a biopsy obtained before and after 3 cycles of either Carboplatin or Paclitaxel. Samples were immediately frozen for RNA extraction.

The Use of High Resolution Chest Computed Tomography in Alpha-1 Antitrypsin Deficiency (QUANTUM-1)

Project description:<p>AAT deficiency is a genetic disorder associated with emphysema. Spirometry, the lung function test that measures how well the lungs exhale air, is used to diagnose and track the progression of emphysema. Some studies have suggested that forced expiratory volume in 1 second (FEV1) measurements, a type of spirometry test, may lack accuracy in detecting disease progression in cases of severe AAT deficiency. Another method, high resolution chest CT scans, may be more accurate at measuring the progression of emphysema. The purpose of this study is to determine if high resolution CT scans are better at detecting the progression of emphysema than lung function tests. Results from this study may lead to the development of a more accurate way to assess lung tissue loss and may improve the understanding of lung destruction in AAT deficiency.</p> <p>This study will last 4 years and will enroll people with AAT deficiency who have nearly normal lung function test results. Study visits, each lasting about 4 hours, will occur at baseline and months 6, 12, 18, 24, and 36. At each visit, participants will undergo lung function tests, a CT scan, blood collection, and a physical exam. Female participants will have urine collected for a pregnancy test. All participants will also complete questionnaires to assess health status and lung function. Study researchers will call participants every 2 months to collect information on lung disease symptoms and medication changes.</p>
