Transcriptome Analyses of LncRNA Metabolic Regulators from High Fat Diet Induced T2DM Mouse Adipose Tissue

ABSTRACT: To search long noncoding RNAs (lncRNAs) which regulating energy metabolism in high fat diet induced T2DM mouse, we performed transcriptome analyses to simultaneously profile mRNAs and lncRNAs in epididymal adipose tissue in normal and high fat diet induced mouse. Combining genome-wide screens, bioinformatics function predictions, and cell-based analyses, we developed an integrative roadmap to identify lncRNA metabolic regulators in epididymal adipose tissue under high fat diet induced T2DM mouse.

Project description:We studied the role of the cAMP responsive factor CREB in promoting insulin resistance following its activation in adipose under obese conditions; We identified genes that were upregulated in primary cultures of mouse adipocytes following exposure to forskolin; and we characterized genes that are also induced in white adipose tissue in mice maintained on a high fat diet. Experiment Overall Design: Primary cultures of mouse adipocytes,harvested from visceral adipose tissue, were exposed to forskolin (10uM) or vehicle control (DMSO) for 2 hours. White adipose tissue (epididymal fat pads) were collected from mice maintained on a 60% high fat diet for 12 weeks and from control mice on normal chow.

Proteome-wide diet-independent adipose tissue alterations link dendritic cells to 2-aminoadipoate associated insulin sensitivity.

Project description:Adipose tissue dysfunction in obese humans is associated with disrupted metabolic homeostasis, insulin resistance, and type 2 diabetes mellitus (T2DM). In a mouse model that has preserved insulin sensitivity despite increased adiposity, we used unbiased three-dimensional integration of proteome profiles, metabolic profiles, and gene regulatory networks to understand adipose tissue proteome-wide changes and their metabolic implications. Multiple-dimensional liquid chromatography tandem mass spectrometry and extended multiplexing TMT labeling (24 biological samples) was used to analyze proteomes of epididymal adipose tissues isolated from wildtype (Csf2+/+) and GM-CSF driven dendritic cell deficient (Csf2-/-) mice that were fed low fat, high fat, or high fat plus cholesterol diets for 8 weeks. The peripheral metabolic health (as measured by body weight, adiposity, plasma fasting glucose, insulin, triglycerides, phospholipids, total cholesterol levels, and glucose and insulin tolerance tests) deteriorated with diet for both genotypes, while mice lacking Csf2 were protected from insulin resistance. Regardless of diet, 30, mostly mitochondrial metabolism proteins participating in amino acid and branched chain amino acid pathways were altered, between Csf2-/- and Csf2+/+ mice. Tissue DHTKD1 levels were >4-fold upregulated and plasma 2-aminoadipoate (2-AA) levels were >2 fold reduced in Csf2-/- mice. GM-CSF driven dendritic cells play a detrimental role in insulin sensitivity via lysine metabolism involving Dhtkd1/2-AA axis.

Next Generation Sequencing Facilitates Quantitative Analysis of epididymal adipose tissue transcriptomes from mice fed either a control-diet or aWestern-diet for one or five successive generations

Project description:Purpose: Successive paternal generations of unhealthy diet induce a fat mass increase. The goal of this study is to compare epididymal adipose tissue transcriptome profiling (RNA-seq) of mice fed either a control-Diet or a High-Fat-Diet for One or Five successive generations. The aim of the study is to identify differentially expressed RNA in epididymal adipose tissue of obese males which might be involved in the exacerbation of induced by the maintenance of High Fat Diet feeding for 5 successive generations. Methods: Epididymal adipose tissue mRNA profiles of 18-months-old mice fed either a control-Diet or a Western-Diet for One or Five successive generations were generated by deep sequencing, in triplicate, using Illumina GAIIx. The sequence reads that passed quality filters were quantitavely analyzed. Reads abundance was evaluated for each gene followed by annotation versus mouse GTF by using the featureCounts function.The R package Edger was used in order to normalize the reads and to identify differentially expressed (DE) genes. Results: RNA-seq data revealed a specific enrichment in gene related to metabolic syndrome traits in male mice obtained after successive paternal generations of Western-Diet.

High-fat diet decreases expression of genes controlling lipid metabolism, mitochondrial function and skeletal system development in adipose tissue, along with increased expression of extracellular matrix remodeling- and inflammation-related genes

Project description:We identified differentially expressed genes in epididymal white adipose tissue of high fat diet(HFD)-fed mice compared to low fat diet-fed mice using microarray analysis. Microarray analysis revealed that genes related to lipolysis, fatty acid metabolism, mitochondrial energy transduction, oxidation-reduction, insulin sensitivity, and skeletal system development were downregulated in HFD-fed mice, and genes associated with extracellular matrix (ECM) components, ECM remodeling, and inflammation were upregulated. The top 10 up- or downregulated genes include Acsm3, mt-Nd6, Fam13a, Cyp2e1, Rgs1, and Gpnmb, whose roles in obesity-associated adipose tissue deterioration are poorly understood. Total RNA of epididymal white adipose tissue was obtained from low fat diet (10 kcal% fat)- and high fat diet(45 kcal% fat)-fed mice and mRNA expression was measured using microarray analysis.

Retinal proteomic study of high-fat diet-induced T2DM model: exploring the interventional targets of nonproliferative diabetic retinopathy

Project description:Objective: This study aims to establish a T2DM rat model consistent with the natural history of the disease, and apply TMT proteomics technology to analyze the retina to reveal the pathogenic mechanism of NPDR and search for new targets for NPDR intervention. Methods: Six-week-old SD male rats were randomly divided into type 2 diabetes group (T2DM group) and normal group (NOR group). T2DM group rats were fed with high-fat diet containing 60% fat energy, while NOR group rats were fed with normal chow diet. After 6 weeks, oral glucose tolerance tests were conducted on the two groups of rats. Following confirmation of insulin resistance, the T2DM group rats were intraperitoneally injected with 2% STZ (30mg/kg), and blood glucose levels were monitored 72 hours later. The rats with random blood glucose levels higher than 16.7 mmol/L were fed with the high-fat diet for another 6 weeks, and then retinas were collected from the two groups of rats for TMT proteomic analysis. Results: After 72 hours and 6 weeks after STZ injection, the T2DM group rats showed typical symptoms of diabetes such as hyperglycemia, weight loss, increased food intake and water consumption, suggesting the establishment of a rat model of T2DM. The bioinformatics analysis results of proteomics reveal the close relationship between differentially expressed proteins, fatty acid metabolism, and angiogenesis. Conclusion: In a T2DM rat model consistent with the natural history of the disease, this study used TMT proteomics technology to deeply analyze the molecular mechanism of NPDR and revealed the key role of fatty acid metabolism in the pathogenesis of NPDR. In addition, Fabp3, Tinagl1, Col4a3, and Snrpd1, may subserve candidate targets for NPDR intervention.

Time-course microarrays reveal early activation of the immune transcriptome and adipokine dysregulation leads to fibrosis in visceral adipose depots during diet-induced obesity

Project description:Time-course analysis of adipocyte gene expression profiles response to high fat diet. The hypothesis tested in the present study was that in diet-induced obesity, early activation of TLR-mediated inflammatory signaling cascades by CD antigen genes, leads to increased expression of pro-inflammatory cytokines and chemokines, resulting in chronic low-grade inflammation. Early changes in collagen genes may trigger the accumulation of ECM components, promoting fibrosis in the later stages of diet-induced obesity. New therapeutic approaches targeting visceral adipose tissue genes altered early by HFD feeding may help ameliorate the deleterious effects of a diet-induced obesity.
