Project description:Microarray analysis was used to evaluate expression differences from a single donor human bone marrow stromal cells (hBMSCs) as a function of varied polymer-based tissue engineering scaffolds. These scaffolds include polycaprolactone (PCL) nanofibers (PCL_NF), films (PCL_SC), poly D,L-lactic acid (PDLLA) nanofibers (PDLLA_NF), films (PDLLA_SC), tissue culture polystyrene (TCPS) and TCPS with osteogenic supplements (TCPS_OS). The results revealed that scaffold structure was able to significantly affect gene expression, with nanofiber scaffolds inducing similar gene expression patterns to hBMCSs cultured with osteogenic media. A library of scaffolds prepared from polycaprolactone or poly D,L-lactic acid was sythesized and cultured with hBMSCs for 14 days with RNA extracted from cells on Day 1 and Day 14. Gene expression analysis was performed using BRB ArrayTools. SC = spun coat, BNF = big nanofiber, TCPS = tissue culture polystyrene, TCPS+OS = tissue culture polystyrene with osteogenic supplements. Project description:In this study the gene expression differences between two titanium surfaces produced at Maastricht University were investigated. These two surfaces were: flat titanium-coated polystyrene and a titanium-coated polystyrene surface imprinted with a pattern selected from an earlier screening study (Ti1018). This pattern was selected based on the osteoinductive properties observed. As a positive control cells on the flat surface were treated with dexamethasone.

Project description:Simple limbal epithelial transplantation (SLET) is a novel in vivo limbal stem cells (LSCs) amplification technique used to reconstruct the ocular surface in¬¬¬ limbal stem cell deficiency (LSCD). However, SLET is limited to two-dimensional (2D) culture systems, and the tissue pieces are easily lost. The developments in hydrogel technology have provided the possibility of three-dimensional (3D) amplification of stem cells. In this study, an injectable photocrosslinkable hydrogel was developed based on porous gelatin methacryloyl (GelMA)/silk fibroin glycidyl methacrylate (SilMA), allowing the 3D culture of LSCs with enhanced expansion efficiency and simplified serial cell culture operations to construct an ocular surface with a uniform cell distribution. The porous GelMA/SilMA hydrogels demonstrated excellent transparency and good adhesion. More importantly, this hydrogel maintained the viability and supported the adhesion of the encapsulated rabbit LSCs and promoted their migration in vitro in 3D culture environments. Furthermore, the immunofluorescence staining results showed positive expression of CK3, CK12, P63 and CK14 in differentiated cells. The above results demonstrated that this porous GelMA/SilMA hydrogel 3D culture of LSCs is beneficial for maintaining the phenotype of stem cells and promoting their differentiation into corneal epithelial cells. These findings provide valuable insights into stem cell therapies and regenerative medicine for ocular surface reconstruction.

Project description:Neural precursor cells (NPCs) tend to aggregate and develop into three-dimensional (3D) spheres, which in return help maintain the stemness of the cells. This close relationship between spherical environment and cell stemness direct us to assume that 3D spheres of astrocytes may facilitate the acquiring of stem cell-like features and generate sufficient seed cells for regeneration of neurons. Primary astrocytes (ASTs) were isolated from the cerebra of postnatal day 1 to day 2 C57BL/6 mouse. ASTs were loaded onto the agarose-coated dishes and cultured to induce the formation of AST spheres. The status of the ASTs over time in cell spheres was characterized by immunochemistry, flow-cytometry, western blotting, and RNA-sequencing. The potency of AST-derived stem cells (A-iSCs) in repairing neural injuries was evaluated in mouse models of middle cerebral artery occlusion (MCAO), spinal cord injury (SCI), and peripheral nerve injury (PNI). In vitro results confirmed that mouse astrocytes cultured on agarose surfaces spontaneously formed cell spheres and exhibited molecular features similar to stem cells, capable of further differentiating into neurons particularly and forming functional synaptic networks with synchronous burst activities. RNA-sequencing results revealed the similarity of A-iSCs to NPCs in global gene expression profiles. It was observed that the transplanted A-iSCs expressed a series of markers for neural differentiation, such as NeuN, Tuj1, and Map2, indicating the conversion of the transplanted A-iSCs into neurons in these scenarios. We also found that the injured mice injected with A-iSCs exhibited significant improvements in sensorimotor functions over 8 weeks compared with the sham and control mice. Taken together, mouse astrocytes form cell spheres on agarose surfaces and acquire stem cell-associated features, and the derived A-iSCs possess the capacity to differentiate into neurons and facilitate the regeneration of damaged nerves across the nervous system.
