Myokine secretion enhancement by the addition of lavender essential oil in cultured skeletal muscle cells

Project description:In recent years, aromatherapy for sportspersons termed as sports aromatherapy has been attracting attention in the field of sports, and aromatic baths and aromatherapy treatments are being used as a new conditioning method, mainly by athletes. Among these, aroma treatment is used to improve performance and recovery from fatigue. On the other hand, myokines (muscle-derived cytokines and chemokines) secreted from the skeletal muscles after exercise, have recently been attracting attention. Many types of myokines are known to act on various organelles and through diverse beneficial metabolic actions promote health. One of them is IL-6, which has been reported to act on muscles and promote myogenesis. Previous studies on essential oil treatment effects are thought to be due to aroma or massage stimulation, and there are no reports of direct effects on muscle at the molecular level comparing essential oils with carrier oils alone. Therefore, we aimed to elucidate the molecular mechanism of myokine secretion by the addition of an essential oil (Lavender oil, LO) using an in vivo exercise model of a mouse muscle cell line (C2C12 cells), which are known as exercise cells.

Transcriptome alteration of cultured dental mesenchymal cells

Project description:Dental mesenchymal cells (dMCs) differentiate into dentin-secreting odontoblasts and are utilized in the majority of tooth regeneration studies to produce an entire tooth organ. An in vitro culture platform of dMCs is useful in generating an adequate quantity of cells for experiments and regenerative therapy. However, culturing techniques may induce functional modification in dMCs. Here, we examined the transcriptomic alterations induced by in vitro culture and investigated the biological properties of cultured dMCs. Hierarchical clustering analysis and principal components analysis reveal an overall transcriptional similarity between the first and second passages that differed significantly from freshly obtained cells. Following in vitro culture, the biological processes of cell adhesion, chromatin organization, cell differentiation and development are significantly altered. Components of signaling pathways, including p53, transforming group factor β/bone morphogenetic protein (TGFβ/BMP), Hedgehog, and Wnt, as well epigenetic regulators are involved. Furthermore, the osteogenic potential of cultured dMCs is also impaired. Our study provides valuable insights for the modification of the current culturing methods to maintain the characteristics of in vivo dMCs.

Genes differentially expressed between Bmi1-overexpressing and empty vector control neural stem/progenitor cells

Project description:Neural stem/progenitor cells were isolated from the lateral ventricle wall of 4-6 week-old CD1 mice and grown as neurospheres under low density culture conditions. Test cells were transduced with bicistronic retroviral constructs for the over-expression of Bmi1 together with eGFP, and control cells were transduced with an empty vector construct expressing eGFP only. To identify genes, which are regulated by BMI1 in neural stem/progenitor cells, the gene expression profiles of neurosphere cells over-expressing Bmi1 were compared empty vector control cells using Affymetrix Gene mouse ST1.0 arrays 3 independent Bmi1-overexpressing neurosphere cultures (test samples) were compared to 3 independent empty vector neurosphere cultures (control samples).

Transcription profiling of human metastatic melanoma biopsies - non-supervised hierarchical clustering of gene expression data

Project description:Despite the frequent detection of circulating tumor antigen-specific T cells, either spontaneously or following active immunization or adoptive transfer, immune-mediated cancer regression occurs only in the minority of patients. One theoretical rate-limiting step is whether effector T cells successfully migrate into metastatic tumor sites. Affymetrix gene expression profiling performed on a series of metastatic melanoma biopsies revealed a major segregation of samples based on the presence or absence of T cell-associated transcripts. The presence of lymphocytes correlated with the expression of defined chemokine genes. A subset of 6 chemokines (CCL2, CCL3, CCL4, CCL5, CXCL9, and CXCL10) was confirmed by protein array and/or quantitative RT-PCR to be preferentially expressed in tumors that contained T cells. Corresponding chemokine receptors were found to be upregulated on human CD8+ effector T cells, and transwell migration assays confirmed the ability of each of these chemokines to promote migration of CD8+ effector cells in vitro. Screening by chemokine protein array identified a subset of melanoma cell lines produced a similar broad array of chemokines. These melanoma cells more effectively recruited human CD8+ effector T cells when implanted as xenografts in NOD/scid mice in vivo. Chemokine blockade with specific antibodies inhibited migration of CD8+ T cells. Our results suggest that lack of critical chemokines in a subset of melanoma metastases may limit the migration of activated T cells, which in turn could limit the effectiveness of anti-tumor immunity. Experiment Overall Design: To examine associations between CD8 expression and a diverse panel of chemokines, affymetrix gene expression profiling performed on a series of metastatic melanoma biopsies. Non-supervised hierarchical clustering analysis revealed a major segregation of samples based on the presence or absence of T-cell-associated transcripts.
