Project description:Diffuse intimal thickening (DIT) is a preclinical stage of atherogenesis in humans that does not exist in commonly used mouse models of atherosclerosis. Vascular smooth muscle cells (SMCs) are the predominant cell type that occupies the intimal and medial layers of human coronary arteries. The intimal layer of DIT is particularly prone to developing atherosclerosis later in life. Differences in the molecular features between the intimal and medial layers of DIT regions may represent the earliest changes that promote atherogenesis in humans, and can be studied by spatial molecular phenotyping.

Project description:In order to find the difference between human lung tissue-derived fibroblasts and human vascular adventitial fibroblasts for enhancing tumor formation ablity of human lung adenocarcinoma cell line A549, we found that human vascular adventitial fibroblasts enhance A549 tumor formation in vivo compared to human lung tissue-derived fibroblasts. CD10 expression in human adventitial cells

Project description:The tunica adventitia ensheaths arteries and veins and contains presumptive mesenchymal stem cells (MSCs) involved in vascular remodeling. We show here that a subset of human adventitial cells expresses the CD10/CALLA cell surface metalloprotease. Sorted CD10+ adventitial cells developed promptly in culture into MSCs, exhibiting higher proliferation, clonogenic and osteogenic potentials than CD10- counterparts. CD10+ adventitial cells increased expression of the cell cycle protein CCND2 via ERK1/2 and prompted NELL1 expression via NF-κB. CD10 expression was upregulated in CD10+ adventitial cells through neural secreted protein sonic hedgehog-mediated Gli1 signaling. These results suggest that CD10 expression, which marks rapidly dividing cells in other normal and malignant cell lineages, plays a role in MSC homeostasis in human tissues.

Single-Cell RNA-Seq Reveals Adventitial Fibroblast Alterations during Mouse Atherosclerosis

Project description:Background: Atherosclerotic cardiovascular disease (ASCVD) remains the leading cause of mortality in the western world despite the success of lipid lowering therapies, highlighting the need for novel lipid-independent therapeutic strategies. Genome-wide association studies (GWAS) have identified numerous genes associated with ASCVD that appear to predominantly function in the vessel wall, suggesting both that vascular cells mediate ASCVD, and that the genes and pathways essential for this vascular cell function may be novel therapeutic targets for the treatment of ASCVD. Furthermore, some of these implicated genes appear to function in the adventitial layer of the vasculature, suggesting these cells are able to potentiate ASCVD. Methods: To investigate the contribution of adventitial cells in atherosclerosis, we conducted single-cell RNA sequencing (scRNA-seq) of the aortic adventitia during atherogenesis in male Ldlr-/- mice via pools of three mice, two samples per condition. We cross-referenced the scRNA-seq data with human ASCVD GWAS to identify regulators of adventitial responses in ASCVD. These regulators were then validated in vitro in human adventitial fibroblasts. Results: We identified four adventitial fibroblast populations, all of which displayed shifts in population size and gene expression over the course of atherogenesis. SERPINH1, an ASCVD-linked GWAS gene, was differentially expressed in adventitial fibroblasts during atherogenesis. Knockdown of SERPINH1 in vitro reduced fibroblast migration and altered subcluster marker gene expression. Conclusions: These findings reveal dynamic changes in adventitial fibroblasts during atherosclerosis and suggest that reduced SERPINH1 expression disrupts adventitial fibroblast function, contributing to ASCVD progression.

Effects of high cholesterol diet on intimal transcriptome [Intima_mRNA]

Project description:Purpose: To investigate the effects of high cholesterol diet on the intimal transcriptome related to atherosclerosis. Methods: 3-week-old LDLR-/- male mice were fed a high cholesterol or low cholesterol diet for 9 weeks. Intimal RNA was extracted by injecting Trizol into the aorta and collecting the flow through for further processing. Results: Mice fed a high cholesterol revealed upregulation in atherosclerosis and immune/inflammatory related genes.
