Role of mitofusins in oocyte development

ABSTRACT: We performed RNA sequencing (RNAseq) analysis to determine the effect of single (Mfn1-/- or Mfn2-/-) and double (Mfn1-/- and Mfn2-/-; Mfn1&2-/-) knockouts of mitofusins on oocyte gene expression profile. We found that in comparison with the wild-type (WT) control, 161 (Mfn1-/-), 538 (Mfn2-/-) and 474 (Mfn1&2-/-) genes were differentially expressed in knockout oocytes at the germinal-vesicle stage (FDR<0.05 and log fold change > 1 or < -1). Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis of genes exclusively down-regulated in each group indicated an over-representation in Mfn1-/- oocytes of Rap1 signaling pathway and PI3K-Akt signaling pathways. In comparison, these pathways were found to be up-regulated across Mfn2-/-, Mfn1&2-/- and WT oocytes. Our findings provide new insights into the roles of mitofusins in oocytes, shedding light on the potential mechanism of infertility associated with Mfn1 deficiency.
