Identification of transcriptional changes after QYHJ treatment in mouse model with pancreatic cancer

ABSTRACT: Purpose: The goals of this study are to characterize the transcriptional changes after QYHJ treatment. More importantly, the transcriptome differences betweeen QYHJ treatment and control mice are compared to identify RNAs that may be targets of QYHJ. Methods: High-throughput RNA sequencing (RNA-seq) was employed to capture the transcriptional changes in QYHJ treatment samples. StringTie was used to quantify gene expression and DESeq2 was utilized to compared expression changes between different sample groups.

Project description:Purpose: The goals of this study are to analyze primary myoblast cell from mdx mice skeletal muscle transcriptome profiling (RNA-seq) treated with different concentration glycine. Methods: primary myoblast cell isolated from 6-8-week-old mdx mice limb skeletal muscle and the primary myoblast treated with 0.2mM or 0.8mM glycine 24-hour. Primary myoblast mRNA profiles of 0.2mM glycine treated(3 replicates) and 0.8mM glycine treated(6 replicates) were generated by deep sequencing, using Illumina HiSeq 2500. The sequence reads that passed quality filters were analyzed at the gene level using HISAT2 and StringTie followed by DESeq2.

Project description:Islets are known to respond to changes in ambient glucose. To quantify the transcriptome-wide changes in ambient glucose, we compared transcriptome of islets exposed to low and high glucose. Isolated islets from wild type male mice. Islets from adult males were pooled, cultured overnight in RPMI containing 11 mM glucose. The next day, all islets were starved in RPMI containing 2.8 mM glucose for 2 hours before stimulation with 2.8 mM glucose or 16.8 mM glucose for 12 hours. Islets were lysed in Trizol for RNA isolation and library construction.

Project description:Purpose: Neutrophils in lung at steady states seems have different characteristics from blood neutrophils even both of them located intravascular regions. The goals of this study are to identify what make lung neutrophils different from other neutrophils from bone marrow and peripheral blood. Methods: Sorted CD11b+ Ly6G+ cells from bone marrow, blood, and lungs of C57BL/6 were investigated with RNAseq using Illumina. Results: Qualified sequence reads per sample to the mouse genome (mm10) with Bowtie2 or STAR were processed with StringTie and identified genes. Neutrophil mRNA profiles from bone marrow, blood and lung of 8 weeks old wild type C57BL/6. Conclusion : 2,032 genes were significantly different in lung neutrophils compared to blood neutrophils.
