Identification of transcriptional changes after treatment with different Traditional Chinese Medicine (TCM) in pancreatic cancer mouse model

ABSTRACT: Purpose: The goals of this study are to characterize the transcriptional changes of pancreatic cancer mouse model after TCM treatment. More importantly, the transcriptome differences betweeen different TCMs are compared to identify RNAs that may be targets of different TCMs. Methods: High-throughput RNA sequencing (RNA-seq) was employed to capture the transcriptional changes in QYHJ treatment samples. StringTie was used to quantify gene expression and DESeq2 was utilized to compared expression changes between different sample groups.

The Gene Expression Analysis of Peripheral Blood Monocytes from Psoriasis Vulgaris Patients with Different Traditional Chinese Medicine (TCM) Syndromes

Project description:Psoriasis is chronic skin disease and an important health concern. Traditional Chinese Medicine (TCM) has shown great promise in the treatment of psoriasis. However, the correlation between TCM Syndromes and genomics of psoriasis has not been evaluated. Here, we analyzed gene expression profiling of monocytes from psoriasis vulgaris patients with different TCM syndrome types to reveal the molecular basis of different psoriasis syndromes. Of the 62 cases of psoriasis vulgaris recruited, 16, 23, and 23 cases were of blood-heat syndrome, blood stasis syndrome, and blood-dryness syndrome, respectively; 10 healthy controls were recruited as controls. Affymertix's Gene Chip ®clariom D gene chip was used to detect the gene expression profile of peripheral blood monocytes collected from recruited individuals. Compared with the healthy control group, 1570 genes were up-regulated and 977 genes were down-regulated in the psoriasis vulgaris patients group; 798 genes and 108 genes were up- and down-regulated in the blood-heat syndrome group respectively; 319 and 433 genes were up- and down-regulated in the blood-dryness syndrome group, respectively; and 502 and 179 genes were up-and down-regulated in the blood-stasis syndrome group. Our analyses indicated not only common differential genes and pathways between psoriasis syndrome groups and healthy controls, but also syndrome-specific genes and pathways. The results of this study link the three syndromes at the gene level and will be useful for clarifying the molecular basis of TCM syndromes of psoriasis. Gene expression profiles of MCF7 cells treated with traditional Chinese medicine components

Project description:At present, the DNA microarray application to study traditional Chinese medicines (TCMs) has received more and more attention. Furthermore, kinds of TCMs were too large, and ingredients in TCMs were also extremely abundant, which afforded an extraordinary source for drug development. To pursue a suitable approach on the research of TCM components, we produced gene expression profiles of 102 TCM ingredients treating MCF7 cells. All selected molecules were main ingredients in Chinese herb and TCM formula. In addition, the gene expression profiles data can be analyzed in combination with public database connectivity map (CMAP) and other bioinformatics methods, which could identify the underlying pharmacological mechanisms and molecular targets/pathway of numerous components. This study indicated that the gene expression profiles data of TCM components was a very convenient and useful approach for researchers engaged in study of TCMs.

Differences in serum proteins in traditional Chinese medicine constitutional population: analysis and verification

Project description:Using high-throughput antibody microarray, through cross-sectional sample detection and verification of samples that had undergone physical changes over time, it was found that people with balanced constitution and dampness constitution in Chinese medicine showed differences in serum protein expression. The differences were expressed as the level changes of 19 proteins such as Dectin-2, Siglec-7, AIF and TACI. The research results provided the basis for the scientific expression of traditional Chinese medicine (TCM) constitution.

Radiation effects on a 3D cell culture of primary human keratinocytes and the protective effect of N-acetyl-cysteine and the Traditional Chinese Medicine (TCM) Qingre-Liyan decoction

Project description:We studied the radiation effect on cell culture model of oral mucositis. The musositis model (tissues) used is an organotypic model which consisted of 3-dimensional (3-D) cell cultures of primary human oral keratinocytes. The tissues were purchased from MatTek corporation (Ashland, MA) and were grown on top of microporous membrane and supplemented with MatTek serum free media. These tissues were irradiated with 12 Gy. Six hours after the irradiation the tissues were cut in half. Half of the tissue was used to extract total RNA (RNeasy Plus Mini kit, Qiagen, Germantown, MD) and the other half was used to evaluate the histology and the apoptosis. We had the following groups: Non-Irradiated Tissue, Irradiated Tissue with 12 Gy; Irradiated Tissue with 12 Gy pretreated with N-acetylcysteine (ACS); Irradiated Tissue with 12 Gy pretreated with the Traditional Chinese Medicine (TCM) Qingre-Liyan decoction; and Irradiated Tissue with 12 Gy pretreated with 50:50 w/w ACS:TCM (AT) Group 1: Human Prirmary 3-D Cultures of Human Keratinocytes were irradiated with 12 Gy; Group 2: Human Prirmary 3-D Cultures of Human Keratinocytes were pretereated with N-acetylcysteine (ACS) and then irradiated with 12 Gy; Human Prirmary 3-D Cultures of Human Keratinocytes were pretreated with the Traditional Chinese Medicine (TCM), Qingre-Liyan Decostion and then irradiated with 12 Gy; Human Prirmary 3-D Cultures of Human Keratinocytes were pretreated with a 50:50 w/w mixture of ACS:TCM (AT) and then irradiated with 12 Gy. Each sample was done in triplicate.

Characteristics of Gut Microbiota in Patients With Colon Cancer of Different Traditional Chinese Medicine Syndromes

Project description:Guided by the idea of combining disease differentiation with syndrome differentiation in traditional Chinese medicine, this study used the form of questionnaire to summarize the changes of TCM syndromes, TCM constitution types in colorectal cancer patients before and after surgery. The Next-generation sequencing technology was used to explore the changes and differences of intestinal microorganisms before and after surgery. The correlation between changes of TCM syndromes, TCM constitution types, intestinal microbial evolution and tumor biological characteristics in colorectal cancer patients before and after surgery was analyzed. To explore the rationality of TCM in the prevention and treatment of colorectal cancer recurrence and metastasis in order to guide the individualized treatment and dietary regulation of TCM.
