Subventricular zone/white matter microglia reconstitute the empty adult microglial niche in a dynamic wave


ABSTRACT: 28d Regular Repop Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 35 days (control) or treated for 7 days with PLX3397 (600 ppm) diet, then to stimulate repopulation PLX3397 placed with control diet for 28 days (28d Recovery). Mice were sacrificed and myeloid cells were extracted from whole hemispheres. RNA was extracted from single-cell suspensions generated from fluorescence-activated cell sorting (FACS) gating for CD11b+CD45int cells. SVZ Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 19 days (control), treated for 14 days with PLX3397 (600 ppm) diet (microglial elimination), or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 5 days (5d recovery). Mice were sacrificed and the subventricular zone was micro-dissected for RNA isolation. 28d Alt Repop +/- LPS Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 35 days (control) or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 28 days (28d Recovery). At 6 or 24 hours (hr) prior to sacrifice, lipopolysaccharide (LPS; 0.33 mg/kg body weight) was administered via intraperitoneal injection. Some mice did not receive an LPS injection and serve as controls (0 hr). Mice were sacrificed and myeloid cells were extracted from whole hemispheres. RNA was extracted from single-cell suspensions generated from fluorescence-activated cell sorting (FACS) gating for CD11b+CD45int cells.
