Project description:We provide gene expression data of wild-type (WT) and various knock-out (KO) bone marrow derived macrophages (BMDMs) from mice (C57BL/6) in different conditions of infections and treatments. We included BMDMs from WT (female and male), Ifnar-/-, IFNg-/-, iNOs-/-, Nlrx1-/- (female and male), Nox2-/- and Prx5-/- mice. BMDMs were either infected with a human protozoan parasite Leishmania guyanensis with or without its endosymbiant double-stranded RNA (dsRNA) virus, Leishmania RNA Virus 1 (LgyLRV1+ and LgyLRV1-, respectively). Alternatively BMDMs were treated with a TLR3 agonist poly I:C, a TLR2 agonist FSL1, with H2O2 or tBHQ.

SVZ Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 19 days (control), treated for 14 days with PLX3397 (600 ppm) diet (microglial elimination), or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 5 days (5d recovery). Mice were sacrificed and the subventricular zone was micro-dissected for RNA isolation. 28d Alt Repop +/- LPS Study: Two-month-old male C57BL/6 (Jax 000664) were maintained on control diet for 35 days (control) or treated for 14 days with PLX3397 (600 ppm) diet and then placed on control diet for 28 days (28d Recovery). At 6 or 24 hours (hr) prior to sacrifice, lipopolysaccharide (LPS; 0.33 mg/kg body weight) was administered via intraperitoneal injection. Some mice did not receive an LPS injection and serve as controls (0 hr). Mice were sacrificed and myeloid cells were extracted from whole hemispheres. RNA was extracted from single-cell suspensions generated from fluorescence-activated cell sorting (FACS) gating for CD11b+CD45int cells.

Project description:Reticuloendothelial macrophages engulf ~0.2 trillion senescent erythrocytes daily in a process called erythrophagocytosis (EP). This critical mechanism preserves systemic heme-iron homeostasis by regulating red blood cell (RBC) catabolism and iron recycling. Although extensive work has demonstrated the various effects on macrophage metabolic reprogramming by stimulation with proinflammatory cytokines, little is known about the impact of EP on the macrophage metabolome and proteome. Thus, we performed mass spectrometry-based metabolomics and proteomics analyses of bone marrow-derived macrophages (BMDMs) before and after EP of IgG-coated RBCs. Further, metabolomics was performed on BMDMs incubated with free IgG to ensure that changes to macrophage metabolism were due to opsonized RBCs and not to free IgG binding. Uniformly labeled tracing experiments were conducted on BMDMs in the presence and absence of IgG-coated RBCs to assess the flux of glucose through the pentose phosphate pathway (PPP). In this study, we demonstrate that EP significantly alters amino acid and fatty acid metabolism, the Krebs cycle, OXPHOS, and arachidonate-linoleate metabolism. Increases in levels of amino acids, lipids and oxylipins, heme products, and RBC-derived proteins are noted in BMDMs following EP. Tracing experiments with U-13C6 glucose indicated a slower flux through glycolysis and enhanced PPP activation. Notably, we show that it is fueled by glucose derived from the macrophages themselves or from the extracellular media prior to EP, but not from opsonized RBCs. The PPP-derived NADPH can then fuel the oxidative burst, leading to the generation of reactive oxygen species necessary to promote digestion of phagocytosed RBC proteins via radical attack. Results were confirmed by redox proteomics experiments, demonstrating the oxidation of Cys152 and Cys94 of glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and hemoglobin-β, respectively. Significant increases in early Krebs cycle and C5-branched dibasic acid metabolites (α-ketoglutarate and 2-hydroxyglutarate, respectively) indicate that EP promotes the dysregulation of mitochondrial metabolism. Lastly, EP stimulated aminolevulinic acid (ALA) synthase and arginase activity as indicated by significant accumulations of ALA and ornithine after IgG-mediated RBC ingestion. Importantly, EP-mediated metabolic reprogramming of BMDMs does not occur following exposure to IgG alone. In conclusion, we show that EP reprograms macrophage metabolism and modifies macrophage polarization.

Project description:IL-10 or IL-6 stimulation of control 129xC57BL/6 murine bone marrow derived macrophages in the presence of LPS. We used microarrays to detail the global programme of gene expression changes in response to IL-6 or IL-10 stimulation in the presence of lipopolysaccharide. BMDMs were isolated from control, IL-6-/-, and IL-10-/- mice on a 129XBL/6 mixed background mice and differentiated in the presence of CSF-1 for 6-7 days. Cells were scraped and plated in 6 well plates at 2x10e6/well. Cells were washed with complete DMEM and rested for 1-2 hr before stimulation with combinations of IL-10 (10 ng/ml), IL-6 (2 ng/ml) or LPS (100 ng/ml) for 45 min or 180 mins. However, the full picture of how mRNA translational factors maintain protein synthesis accuracy and co-translational protein folding are far from being fully understood.To address this question,we evaluated the role of Elp3 on protein in the IL4 induced alternative bone marrow derived macrophages (BMDMs). The BMDMs from widetype and Elp3 myeloid specific knockout mice were treated by IL-4 for 24h or not. Then the aggregates were isolated from these BMDMs and performed by MS.

Project description:Leishmania are medically relevant protozoan parasites that cause leishmaniasis, a neglected tropical disease that can differently manifest depending on the species and the immune status of the host. Previously, it has been suggested that the presence of an endosymbiont double stranded RNA virus, Leishmania RNA Virus 1 (LRV1), within the parasite may leed to an exarcerbation of the disease outcome and represents an important factor for treatment failure and relapse.
