Single-cell RNAseq analysis of kidneys from RNLS knockout mice with chronic kidney disease treated with RNLS peptide encapsulated in mesonanoparticles

ABSTRACT: We compared transcriptome profiles of kidneys from RNLS KO mice treated with RNLS peptide RP81 in mesonanoparticle (RP81-MNP) or with empty MNP (control). Our study provided some molecular mechanism of how RNLS peptide protected kidney from cisplatin-induced CKD.

Project description:RNLS global knockout (KO) and wild-type (WT) mice were used in the present study to establish a long-term CKD model (up to 18 weeks post-surgery) via 2-step 5/6 Nx, and we observed the role of RNLS by over-expressing it using a cardiac-specific adeno-associated virus (AAV). 2-weeks post-surgery, we injected AAV9-cTnT-RNLS or vector to WTor KO CKD mice by tail vein and the hearts were harvested at week 18. Then RNA-sequencing (RNA-seq) analysis was performed to explore the underlying molecular mechanism.

MicroRNA-mediated attenuation of branched-chain amino acid catabolism promotes ferroptosis in chronic kidney disease [RNA-seq]

Project description:Chronic kidney disease (CKD) complicates cisplatin-based chemotherapy of cancer patients. Here we investigate microRNA (miRNA)-regulated transcriptomic activity to unveil biological processes associated with cisplatin-induced kidney injury. Implementing chimeric-eCLIP-seq approach to a mouse model for cisplatin-induced CKD, we identify direct pairs of miRNA and their target messenger RNA in the injured kidney. We find a dedicated transcriptomic program directed by a group of miRNAs that alter metabolic pathways centered on mitochondria in the injured kidneys. Specifically, cisplatin-induced miRNA, miR-429-3p suppresses the mitochondria pathway that catalyzes branched-chain amino acid (BCAA), eventually leading to lipid peroxidation-dependent cell death, called ferroptosis. Thus, the identification of miRNA-429-3p-mediated stimulation of ferroptosis suggests a therapeutic potential for BCAA pathway modulation in ameliorating CKD and cisplatin-associated nephrotoxicity.

The transcriptome profiling of the kidneys of WT and TRPM2-KO mice treated with cisplatin was compared to find differentially expressed gene which may be related to TRPM2 on cisplatin nephrotoxicity.

Project description:To clarify the effects of cisplatin (cis-diamminedichloroplatinum II, CDDP) on the gene expression profiles in renal proximal tubules, microarray analyses were carried out using total RNA samples isolated from microdissected proximal tubules and whole kidneys. The molecular events underlying acute kidney injury (AKI) in the proximal tubules of rats with cisplatin-induced nephrotoxicity were successfully clarified with 17,000 transcripts.

microRNAs expression in kidneys from normal, cisplatin induced AKI rats and those with the restoration of MSCs

Project description:The availability of mesenchymal stem cells in kidney injury repair has been demonstrated. However, the underlying molecular mechanism remains not well understood. Herein, we profiled the altered microRNAs in renal tissiues from cisplatin-induced acute kidney injury (AKI) rats administrated with or without rat bone marrow mesenchymal stem cells, normal kidneys as control. Sprague Dawley rats were administrated with PBS by caudal vein after 24h peritoneal injection of 6mg/mg cisplatin, short for g2. 1×106 MSCs were injected to rats after 24h peritoneal injection of cisplatin, short for g3. Kidneys from healthey rats were used as control, short for g1. Renal tissues were collected at day 5 after treatment. There were 44 micorRNAs changed with significance between g2 and g3, among which 36 microRNAs upregulated and 8 microRNAs downregulated.
