Project description:To investigate ferroptosis-dependent changes in DNA methylation, a genome wide DNA methylation profiling of ferroptotic (RSL3-treated) multiple myeloma cells (MM1S & MM1R) was performed using the 850K MethylationEPIC BeadChip Array. The ferroptotic DNA methylation signature was compared to myeloma cells pre-treated with the ferroptosis inhibitor ferrostatin-1 (FRSL3), which served as a negative control. In total, three biological replicates per treatment per cell line were included.

Project description:Alterations of metabolic and biological processes occur in ferroptotic cells. We analyzed transcriptional responses of murine embryonic fibroblasts (MEFs) exposed to a ferroptosis inducer erastin. We found that a set of genes related to protection against oxidative stress was induced upon ferroptosis and Bach1 promoted ferroptosis by repressing a set of these genes involved in synthesis of glutathione or metabolism of intracellular labile iron. Our findings suggests that ferroptosis is programmed at transcriptional level and Bach1 works as a controller in setting a threshold of ferroptosis.

Project description:The adaptor protein ASC contributes to innate immunity through the assembly of caspase-1-activating inflammasome complexes. We demonstrate that ASC plays an inflammasome-independent cell-intrinsic role in adaptive immune cells. Asc-/- mice displayed defective antigen presentation by dendritic cells and lymphocyte migration due to impaired Rac-mediated actin polymerization. Genome-wide analysis showed that ASC, but not Nlrp3 or caspase-1, controls mRNA stability and expression of DOCK2, a guanine nucleotide exchange factor that mediates Rac-dependent signaling in immune cells. DOCK2-deficient dendritic cells showed similar defective antigen uptake as Asc-/- cells. Ectopic expression of DOCK2 in ASC-deficient cells restored Rac-mediated actin polymerization, antigen uptake and chemotaxis. Thus, ASC shapes adaptive immunity independently of inflammasomes by modulating DOCK2-dependent Rac activation and F-actin polymerization in dendritic cells and lymphocytes. Three replicates of naïve WT and three replicates of Asc-/- bone marrow derived dendritic cells were analyzed on the Affymetrix HT MG-430 PM plate array.

Project description:Dendritic cells (DC) play a crucial role in initiating, shaping and controlling the immune response. They are the most important antigen presenting cells in the immune system. DC can be divided in immature and mature DC. DCs reside in an immature state in most tissues or they circulate in the blood, where they recognize and phagocytose pathogens and other antigens. Direct contact with many pathogens leads to the maturation of DCs. The property to stimulate T cells is reserved for mature DC (O`Doherty, U. 1994). This functional maturation results in an up-regulation of the surface molecule MHC class II, adhesion molecules (CD54 and CD58) and co-stimulatory receptors (CD80 and CD86) as well as decreased antigen uptake capacity (CD206). (Banchereau, J. and Steinman, R.M. 1998). Immunosuppressive drugs have revolutionized organ transplantation and improved the therapeutic management of autoimmune diseases. These drugs interfere on lymphocytes but they also act at the earliest stage of immune response. They are targeting key functions of dendritic cells (Hackstein H. and Thomson A.W., 2004 review). Sanglifehrin A (SFA) is a new immunosuppressive drug. It is a representative of a class of macrolides produced by the actinomycetes strain Streptomyces A92-308110 that bind to cyclophilin A (CypA), the binding protein of cyclosporine A (CsA). Most studies about immunosuppressive effects of SFA took place on T and B lymphocytes. Unlike CsA, the cyclophilin-SFA-complex shows no effect on the calcium-dependent protein phosphatase calcineurin (Zenke, G. 2001). Unlike CsA and FK506, SFA shows no inhibition of IL-2 expression (Zenke, G. 2001; Zhang, L.H. 2001), nor does it suppress IL-2 receptor transcription (Zhang, L.H. 2001) in T lymphocytes. It could be shown that SFA reduces the Interleukin 12 production in dendritic cells (Steinschulte, C. 2003). IL-12p70 plays an important role in the pathogenesis of inflammation and autoimmune diseases. DCs generated in the presence of SFA show reduced macropinocytosis and lectin-mediated endocytosis. In contrast, CD89 und CD32 were increased by SFA. The effect of SFA on the expression of Ag uptake receptors and Ag capture by DCs makes SFA unique among other immunophilin-binding immunosuppressive drugs (Woltman, A. 2004). Project description:Despite significant advances in the development and refinement of immunotherapies administered to combat cancer over the past decades, a number of barriers continue to limit their efficacy. One significant clinical barrier is the inability to mount initial immune responses towards the tumor. As dendritic cells are central initiators of immune responses in the body, the elucidation of mechanisms that can be therapeutically leveraged to enhance their functions to drive anti-tumor immune responses is urgently needed. Here, we report that the dietary sugar L-fucose can be used to enhance the immunostimulatory activity of dendritic cells (DCs). L-fucose polarizes immature myeloid cells towards specific DC subsets, specifically cDC1 and moDC subsets. In vitro, L-fucose treatment enhances antigen uptake and processing of DCs. Furthermore, our data suggests that L-fucose-treated DCs increase stimulation of T cell populations. Consistent with our functional assays, single-cell RNA sequencing of intratumoral DCs from breast tumor-bearing mice confirmed transcriptional regulation and antigen processing as pathways that are significantly altered by dietary L-fucose. Together, this study provides the first evidence of the ability of L-fucose to bolster DC functionality and provides rational to further investigate how L-fucose can be used to leverage DC function in order to enhance current immunotherapy.
