Genome-wide expression analysis of mouse chromosome 2 in gonadal fat and liver tissues reveals a complex architecture of obesity

ABSTRACT: Transcript abundance in adipose and liver tissues was used to map expression quantitative trait loci (eQTL) underlying fat accretion, weight gain and liver weight. Gonadal fat and liver tissues were used from 48 individuals F2 selected from a cohort (1,200 mice) originated from a cross between M16i and MB2 mice. A complex polygenic architecture within the MB2 locus was observed which was comprised of several locally regulated eQTL (cis-eQTL) with significant correlations to obesity traits.

Genetic and genomic analysis of hyperlipidemia, obesity and diabetes using TALLYHO x C57BL6 F2 mice

Project description:Type 2 diabetes (T2D) is the most common form of diabetes in humans and is closely associated with dyslipidemia and obesity that magnifies the mortality and morbidity related to T2D. The TALLYHO/JngJ (TH) mouse is a polygenic model for T2D characterized by obesity, hyperinsulinemia, impaired glucose uptake and tolerance, hyperlipidemia, and hyperglycemia. To determine the genetic factors that contribute to these T2D related characteristics in TH mice, we interbred TH mice with C57BL/6J (B6) mice. The parental, F1, and F2 mice were phenotyped at 8, 12, 16, 20, and 24 weeks of age for 4-hour fasting plasma triglyceride, cholesterol, insulin, and glucose levels, as well as body weights. Fat pad and carcass weights were measured at 24 weeks after sacrificing the mice. The F2 mice were genotyped genome-wide for 68 markers. Of 393 genotyped F2 mice, 16 were chosen from the extremes of the triglyceride distribution (8 high and 8 low), and liver, pancreas, muscle and adipose tissue were measured for gene expression. Gene expression quantitative trait locus (eQTL) analysis aided in selection of candidates underlying hyperlipidemia, diabetes and obesity QTLs. We identified several genetic loci that affected quantitative variation in plasma lipid and glucose levels and obesity traits. 16 (8 high and 8 low) out of 393 F2 mice were chosen from the extremes of the triglyceride distribution, excluding overtly diabetic mice, and liver, pancreas, muscle and adipose tissue were measured for gene expression. In addition to data from the 64 microarrays on the 16 mice, a supplemental file with phenotypes and marker genotypes is provided for all mice as a supplementary file on the Series record (below). Mouse identification numbers are included to connect the data files.

Project description:Dietary administration of n-3 polyunsaturated fatty acids (PUFA) is often associated with altered adipose tissue (AT) morphology and/or function in obese mice. However, it is unclear whether these observations are an indirect consequence of reduced weight gain or result from direct actions of n-3 PUFA. Here we studied the AT of fat-1 transgenic mice that convert endogenous n-6 to n-3 PUFA. These mice display equivalent weight gain and fat accretion to their wild-type (WT) counterparts. We performed Affymetrix microarray in epididymal AT of male hemizygous fat-1(+/-) transgenic mice and their wild type littermates that had been fed high fat diets from 6 weeks of age (diet-93075, 55% Kcal from fat, Harlan Teklad). Mice were sacrificed after 8 weeks on the HF diet. At sacrifice epididymal adipose tissues excised for the microarray study (n=3 per group) were rapidly homogenized in QIAZOL (QIAGEN) and snap frozen in liquid nitrogen.

Profiling of liver tissue from the F2 M16xICR mouse cross.

Project description:We profiled gene expression in liver tissue from F2 progeny from a cross between the outbred M16 (selectively bred for rapid weight gain) and ICR (control) mouse strains. We developed a framework for reconstructing tissue-to-tissue coexpression networks between genes in hypothalamus, liver or liver tissues that are independent of networks constructed from single tissue analyses. The subnetworks we identify as specific to tissue-to-tissue interactions associate with multiple obesity-relevant biological functions like circadian rhythm, energy balance, stress response, or immune response. Proteomics analysis of insulin sensitive tissues in high fat diet fed mice

Project description:The popularity of high fat foods in modern society has been associated with epidemic of various metabolic diseases characterized by insulin resistance, the pathology of which involves complex interactions between multiple tissues such as liver, skeletal muscle and white adipose tissue (WAT). To uncover the mechanism by which excessive fat impairs insulin sensitivity, we conducted a multi- tissue study by using TMT-based quantitative proteomics. 3-week-old ICR mice were fed with high fat diet (HFD) for 19 weeks to induce insulin resistance. Liver, skeletal muscle and epididymal fat were collected for proteomics screening. Additionally, PRM was used for validating adipose differential proteins. By comparing tissue-specific protein profiles of HFD mice, multi-tissue regulation of glucose and lipid homeostasis and corresponding underlying mechanisms was systematically investigated and characterized. NC: normal birth weight + chow diet; NH: normal birth weight + high fat diet; LC: low birth weight + chow diet; LH: low birth weight + high fat diet.

Transcription profiling of rat anterior pituitary gland using DES-treated COPxACI F2 rats - Expression QTL mapping.

Project description:Anterior pituitary glands were isolated from 21 week old male rats treated for 12 weeks with the synthetic estrogen diethylstilbestrol (DES). 43 COPxACI F2 animals were selected based on pituitary weight phenotypic extremes: 22 highest and 21 lowest pituitary weights in our F2 population. Expression profiles were determined using Affymetrix Rat Genome 230 v. 2.0 arrays. In conjunction with genomewide genotypes available for these 43 animals, linkage analysis was done using mRNA abundance of each transcript on the array as a separate quantitative phenotype. These arrays provide a resource for expression QTL (eQTL) mapping on a genomewide level in the pituitary gland. Experiment Overall Design: 43 samples from anterior pituitary gland were analyzed. COPxACI F2 male animals were treated with DES for 12 weeks, starting at 9 weeks of age. Pituitary glands were harvested and weighed at sacrifice (21 weeks of age). The 43 samples were selected for pituitary weight phenotypic extremes: 22 highest and 21 lowest pituitary weights in our F2 population. Genomewide genotypes are available for these animals and in conjunction with the array data, genomewide expression QTL (eQTL) mapping was done.

Expression profiling of Liver tissue from C57BL/6J X C3H/HeJ)F2 and (C3H/HeJ X C57BL/6J )F2

Project description:The purpose of this experiment was to determine the expression traits in animals from F2 intercross of inbred strains C57BL/6J, C3H/HeJ. (N=309, 164 males and 145 females). Liver from 302 F2 female and male mice were generated by intercrossing F1 mice. Mice were fed chow diet containing 4% fat (Ralston-Purina Co., St. Louis, MO) until 8 weeks of age and then were placed on a high-fat "Western" diet containing 42% fat and 0.15% cholesterol (Teklad 88137, Harlan Teklad, Madison WI) for 12 weeks. At 20 weeks mice were sacrificed, after a 12-hour fast liver tissues were dissected and flash frozen in LN2 and stored at -80Â°C.
