RNA-sequence on prostates of wild type (C57BL/6J) and ARNoC/Y mice that underwent orchidectomy and testosterone (or vehicle) replacement for 4 days

ABSTRACT: We generated a mouse model (ARNoC/Y) with a mutation in the FQNLF motif located on the aminoterminal domain (NTD) that disrupts the N/C interactions of the androgen receptor (AR). These mice exhibit a normal male phenotype. To be able to compare WT males and ARNoC/Y mice more directly, we performed a castration experiment with testosterone (T) supplementation for 4 days. Since slightly differences in prostate weight were observed, we performed RNA-seq on these prostate to evaluate the biological relevance.

Project description:We generated a mouse model (ARLmon/Y) with disrupted dimerization of the ligand-binding domain 25 (LBD) of the androgen receptor (AR). These mice exhibit an external female phenotype with undescended testes. Histological analysis of the testes of ARLmon/Y mice revealed some residual spermatogenesis. Hormone measurements in serum of the ARLmon/Y mice showed significantly high levels of androgens. To study if there is any remaining AR activity, we orchidectomized WT and ARLmon/Y mice and gave them supraphysiological testosterone replacement for 4 days. RNA-seq was performed on kindeys.

Perioperative Testosterone Replacement Therapy in Sarcopenic Male Colorectal Cancer Patients

Project description:With increasing age and the additional impact from the bowel cancer and the chemotherapy and/ or radiotherapy it has been described that testosterone (a male hormone produced naturally in the body) levels are reduced. Testosterone has an impact on numerous body functions including the muscle mass and quality. Previous studies have identified that muscle mass is reduced as a result of ageing but also because of the deleterious effect of cancer and chemotherapy and/or radiotherapy. There is growing evidence from published studies that patients with better muscle mass and quality, do better after surgery. Mr Jenkins and his team are therefore looking at ways, the investigators can try and prevent or reduce this muscle loss and therefore improve patient outcomes. The aim of this study is to assess whether using testosterone replacement therapy in the form of a topically applied gel daily for a total of 12 weeks, is feasible and acceptable by the patients who are diagnosed with colorectal cancer and are waiting to undergo surgery. The investigators will also collect information related to the testosterone replacement therapy such as questionnaires on the quality of life, fatigue and muscle mass, and blood biomarker changes in the blood.

Testosterone Replacement versus Placebo Bone and Bone Marrow in Castrated Mice

Project description:Metastatic prostate cancer is commonly localized in bone and treated with androgen ablation, which is initially highly effective, but eventually results in resistant cancer cells. The decreased dependence on androgen is associated with activation of the androgen receptor (AR) via other growth factor receptor pathways. The current study analyzed global gene expression in bone from castrated and sham operated C57BL/6 mice to identify factors that may be responsible for androgen signaling. Androgen suppression by castration increased the expression of 159 genes with insulin-like growth factor binding protein 5 (IGFBP5) being the most consistently increased. IGFBP5 protein was detected by immune histochemistry in osteoblasts, bone marrow stromal cells and endothelial cells. In vitro treatment of marrow stromal cells with charcoal-stripped serum increased IGFBP5 mRNA expression approximately 25-fold and was reversed by androgen supplementation. Endogenous IGFBP5 was incorporated into extracellular matrix. In addition, treatment of extracellular matrices with exogenous IGFBP5 and IGF-1 or IGF-2 enhanced the growth of immortalized human prostate cells. These results suggest that androgen ablative therapy may increase IGFBP5 in the marrow microenvironment, thereby providing support for the development of therapy resistance prostate cancer cells. Strength training with suppressed testosterone level

Project description:Human subjects were randomized for treatment with a GnRH-analogue, Goserelin, which suppresses endogenous testosterone or placebo for 12 weeks. Strength training was performed during the last 8 weeks. The suppression of testosterone resulted in an attenuation of the normal muscle adaptation to strength training (increased muscle mass and strength). To identify molecular signals involved in the response to testosterone levels, biopsies were obtained 4 hours after the last training session and gene expression compared with Affymetrix 3' microarrays. This timepoint should capture goserelin effect on both constitutive expression, training induced changes as well as acute exercise induced (4 hours) differences in mRNA levels.
