RNA-seq of dorsal root ganglia (DRG) neurons of dry skin (AEW) mice

ABSTRACT: Dry skin evokes itch (pruritus), but underlying mechanisms are still elusive. To examine alterations of itch-related genes, dry skin condition was induced by AEW (Acetone/Ether/Water) treatment. In this study, RNA-seq was performed with dorsal root ganglia neurons obtained from AEW-treated ICR male mice (12-week-old) or control mice.

Project description:In this study, we performed a label-free relative quantitation to identify the peptide candidates correlated with itch sensation in murine models. We tested two different itch models-the AEW model (known commonly as the dry skin model) and the MC903 model. Performed peptide extraction from the dorsal horn (DH) and dorsal root ganglia regions of these two models and compared their levels with that of control mice.

RNA-seq of mouse tissue samples from animal model：Effects of MMP9 on gene expression.

Project description:In the present study, RNA-seq technique were performed To explore biological significance of MMP9 in the neuro-cutaneous signaling network of chronic itch. We used MMP-9-IN-1, a specific inhibitor of MMP9, or its vehicle to treat ACD model mice, and conducted high-throughput sequencing on ear tissue samples of mice. It was found that inhibition of MMP9 expression affected the expression of a variety of itching factors and skin inflammatory factors related to the skin pathology of chronic dermatitis. These changes may indicate that MMP9 is closely related to inflammation and pruritus. This finding might help us to better understand the immune-neuro-modulatory mechanism in itch circuits.

Project description:To test the hypothesis that defects in the termination of inflammatory signaling led to skin inflammation that results in the “Itchy” phenotype, we isolated RNA from the lesional skin of Itch-/- mice and from the skin of wild-type mice and performed genome-wide mRNA expression profiling by RNA sequencing. We ranked genes based on the fold change in their expression (increased or decreased) in Itch-/- skin relative to that in wild-type skin. Molecular analysis in imiquimod (IMQ)-induced psoriasis model

Project description:The mechanisms underlying pruritus of imiquimod (IMQ)-induced psoriasis remain poorly understood. In this study, we investigated whether there are certain key signaling molecules downstream of the recently identified peptides mediating itch in the spinal cord. We investigated the changes in the trascriptome by performing RNAseq of cervical spinal cords in control and IMQ-induced psoriasis. Our study represents the detailed analysis of transcriptomes of spinal cord in chronic itch model with biologic replicates, generated by RNA-seq technology.

Keratin-dependent TSLP expression suggests a link between skin blistering and atopic disease

Project description:Atopic dermatitis (AD) is a pruritic and inflammatory disorder characterized by elevated levels of thymic stromal lymphopoietin (TSLP). Pruritus is prevalent in epidermolysis bullosa (EB). Currently, epidermal barrier disruption is known to trigger TSLP upregulation, however, mechanisms controlling TSLP expression remain incompletely understood. Tslp levels were highly upregulated in the epidermis and in the serum of keratin-deficient mice. Cultured keratinocytes either lacking keratins or expressing the dominant K14p.Arg131Pro show highly increased TSLP. Re-expression of wild-type K14 normalized TSLP levels. We demonstrate that keratins regulate Tslp expression in parts through MEK1/2 activation. The finding that 8 out of 17 EBS patients show elevated TSLP serum levels supports a major role of keratins in TSLP regulation. Our data identify a novel, keratin-dependent and cell-intrinsic regulation of Tslp. Elevated TSLP levels likely explain the high prevalence of pruritus in EBS and additional forms of EB, suggesting TSLP as novel biomarker for pruritus in EB. MEK1/2, in addition to calcineurin inhibitors, might be suitable drugs to treat itch in EB. Investigation was carried out using Keratin deficient keratinocytes isolated from typeI keratin mice.
