Tire-wear particle leachate toxicity to Americamysis bahia: analysis of sublethal and molecular effects

ABSTRACT: Tire-wear particles (TWPs) are considered among the largest contributors of microplastics to the environment. They are subject to break-down due to environmental weathering, which allows for potentially toxic chemicals to be leached from and sorbed onto the particles. In this study, leachate generated from “weathered” and “un-weathered” TWPs were used for sublethal toxicity tests with Americamysis bahia.

Developmental toxicity of tire wear particle (TWP) leachates produced in the presence or absence of simulated natural light

Project description:Tire wear particles (TWP) are small micro- or nano-particles resulting from the friction of tire tread against roads. These microplastics have been found in waterways, usually through rain and wind, posing a potential risk to exposed aquatic life. The goal of this project was to assess the toxicity of chemical leachates from TWP and characterize how they may impact the health and development of aquatic life, like Danio rerio, zebrafish. Furthermore, we also determined if solar-simulated or dark conditions intensified the effects of TWP, considering processes like photodegradation and photochemical oxidation occur in sunlight. Zebrafish were exposed from 0-4 days post fertilization (dpf) to a filtered TWP solution at varying concentrations, and each concentration was leached in either solar-simulated or dark conditions. Mortality and hatching rates were quantified throughout the exposures, and physical anomalies were recorded at 4 dpf using microscopy. Ethyoxyresorufin-O-deethylase (EROD) activity, an indicator of Phase I biotransformation, was performed using fluorescent microscopt. RNA sequencing was performed for 4 dpf larvae. TWP exposures, regardless of leaching light status, increased mortality, yolk sac edema, pericardial edema, spinal curvature, and craniofacial malformations, while decreasing hatching and swim bladder inflation. Regardless of lighting during leaching, EROD activity increased in the liver and brain and decreased in the gut CYP1A. RNA sequencing revealed some conserved processes in response to TWP, regardless of light status during leaching, including those involved in neurobiological function. However, divergent responses were also observed for processes such as amino acid metabolism, steroid biosynthesis, and fatty acid metabolism. Overall, these results suggest a disruption of embryonic development in zebrafish when exposed to TWP, and that sunlight during chemical leaching can alter the molecular responses to TWP.

Gene expression profiling in wear-particle induced and infectious endoprosthesis loosening

Project description:The aim of the study was to identify markers for the early diagnosis of endoprosthesis loosening, for the differentiation between wear-particle induced and septic loosening, as well as to gather new insights into the pathogenesis. 824 genes were differentially expressed with a fold change greater than 2. Among these were Chitinase 1, CD52, Calpain 3, Apolipoprotein, CD18, Lysyl oxidase, Cathepsin D, E-Cadherin, VE-Cadherin, Nidogen, Angiopoietin 1 and Thrombospondin 2, and the differential expression levels were validated by RT-PCR. The chitinase activity was significantly higher in the blood from patients with wear-particle induced prosthesis loosening (p=0.001). However, using chitinase activity as a marker for early diagnosis, it has a specificity of 83% and a sensitivity of only 52%, due to a high variability both in the disease and in the control group. Transcriptomic Analysis of Zebrafish Larvae Exposed to Tire Wear Particle Leachates

Project description:For transcriptomic analysis, total RNA was extracted from pooled zebrafish larvae (80 per sample, three biological replicates per group) preserved in RNAlater™. High-quality RNA (RIN > 7.0), isolated using the RNeasy Mini Kit, was quantified and assessed for integrity. Ribosomal RNA was depleted prior to cDNA library construction using the TruSeq Stranded Total RNA Library Prep Gold Kit. The resulting libraries were quantified, size-validated, and sequenced using the Illumina NovaSeqX platform with 2×100 bp paired-end reads.

Quantification of tire tread wear particle in road dust through pyrolytic technique.

Project description:Road dust cotains tire wear particles (TWPs) and a large amount of mineral particles (MPs). Given that tire tread in vehicles is mainly comprised of natural rubber (NR), isoprene and dipentene could be the main pyrogenic products stemmed from the thermolysis of NR. This offers a great chance to quantify the exact mass of TWP in road dust. As such, this study focused on the influence of MPs on the trends in thermolytic behaviors of NR using the resistive furnace (furnance) and Curie point pyrolyzers. This study confirmed that a reliable correlation in line with the formation of isoprene and dipentene could not be realized using the furnace type of a pyrolyzer. This means that employing the furnace type of a pyrolyzer in quantitification of TWPs could not be a viable and approproiate option due to the diverted thermolytic trends of NR due to differences in the heat transfer and adsoprtion of the pyrogenic products triggered by MPs. In the Curie point type of a pyrolyzer, the production rates of isoprene and dipentene were linearly responded to the mass of NR. The ferromagnetic substance in MPs could lead to the thermolytic trend change of NR. Thus, adopting the Curie point type of a pyrolyzer could be a viable option for quantification of TWPs in road dust when the effects of ferromagnetic substance are well neutralized.

Uptake, Metabolism<b>,</b> and Accumulation of Tire Wear Particle-Derived Compounds in Lettuce.

Project description:Tire wear particle (TWP)-derived compounds may be of high concern to consumers when released in the root zone of edible plants. We exposed lettuce plants to the TWP-derived compounds diphenylguanidine (DPG), hexamethoxymethylmelamine (HMMM), benzothiazole (BTZ), N-phenyl-N'-(1,3-dimethylbutyl)-p-phenylenediamine (6PPD), and its quinone transformation product (6PPD-q) at concentrations of 1 mg L-1 in hydroponic solutions over 14 days to analyze if they are taken up and metabolized by the plants. Assuming that TWP may be a long-term source of TWP-derived compounds to plants, we further investigated the effect of leaching from TWP on the concentration of leachate compounds in lettuce leaves by adding constantly leaching TWP to the hydroponic solutions. Concentrations in leaves, roots, and nutrient solution were quantified by triple quadrupole mass spectrometry, and metabolites in the leaves were identified by Orbitrap high resolution mass spectrometry. This study demonstrates that TWP-derived compounds are readily taken up by lettuce with measured maximum leaf concentrations between ∼0.75 (6PPD) and 20 μg g-1 (HMMM). Although these compounds were metabolized in the plant, we identified several transformation products, most of which proved to be more stable in the lettuce leaves than the parent compounds. Furthermore, continuous leaching from TWP led to a resupply and replenishment of the metabolized compounds in the lettuce leaves. The stability of metabolized TWP-derived compounds with largely unknown toxicities is particularly concerning and is an important new aspect for the impact assessment of TWP in the environment.

Copper-enriched automotive brake wear particles perturb human alveolar cellular homeostasis

Project description:Background Airborne fine particulate matter with diameter < 2.5 μm (PM2.5), can reach the alveolar regions of the lungs, and is associated with over 4 million premature deaths per year worldwide. However, the source-specific consequences of PM2.5 exposure remain poorly understood. A major, but unregulated source is car brake wear, which exhaust emission reduction measures have not diminished.AQ1 Methods We used an interdisciplinary approach to investigate the consequences of brake-wear PM2.5 exposure upon lung alveolar cellular homeostasis using diesel exhaust PM as a comparator. This involved RNA-Seq to analyse global transcriptomic changes, metabolic analyses to investigate glycolytic reprogramming, mass spectrometry to determine PM composition, and reporter assays to provide mechanistic insight into differential effects. Results We identified brake- wear PM from copper-enriched non-asbestos organic, and ceramic brake pads as inducing the greatest oxidative stress, inflammation, and pseudohypoxic HIF activation (a pathway implicated in diseases associated with air pollution exposure, including cancer, and pulmonary fibrosis), as well as perturbation of metabolism, and metal homeostasis compared with brake wear PM from low- or semi-metallic pads, and also, importantly, diesel exhaust PM. Compositional and metal chelator analyses identified that differential effects were driven by copper. Conclusions We demonstrate here that brake-wear PM may perturb cellular homeostasis more than diesel exhaust PM. Our findings demonstrate the potential differences in effects, not only for non-exhaust vs exhaust PM, but also amongst different sources of non-exhaust PM. This has implications for our understanding of the potential health effects of road vehicle-associated PM. More broadly, our findings illustrate the importance of PM composition on potential health effects, highlighting the need for targeted legislation to protect public health.
