ABSTRACT: The experiment was to construct a library of targeted insertion mutants, using the "targetron" method, and compare the fitness of mutants in a murine UTI model by measuring the relative quantity of mutants before and after experimental UTI. Since the targetron disrupts the gene function, enriched mutants after this selection suggests the gene is deleterious for this process and vice versa. Mutations were in genes predicted to enable the bacteria to import sugars. The targetron sequence is provided in a supplementary file.
