The combination of translation initiation and termination site ribosome profiling precisely defines novel small and alternative reading frames in a model cyanobacterium

ABSTRACT: Naturally occurring peptides and small proteins mediate multiple important functions in all domains of life. However, their direct detection by mass spectrometry or the annotation of their genes in a given genome is often only fragmentary. Therefore, approaches involving ribosome profiling (Ribo-seq) appear very attractive. Compared to mass spectrometry methods, no prior knowledge of possible peptides and their behavior in the mass spectrometer is necessary. Here, using the model cyanobacterium Synechocystis sp. PCC 6803, we present the first Ribo-seq analysis of a cyanobacterium involving inhibitors that stall ribosomes at the translation initiation and termination sites (TIS- and TTS-Ribo-seq).
