Expression data from stem samples taken from the base and the first internode of Arabidopsis wildtype and wox4-1 plants

ABSTRACT: Stem samples of wildtype Columbia plants and the wox4-1 mutant (Gabi_462G01) were analyzed in order to draw a connection between general transcriptomic changes during interfascicular formation in the wildtype and WOX4-dependent gene regulation during this process. We used microarrays to identify factors downstream of WOX4 in the formation of the interfascicular cambium.

Project description:Subsequently to primary growth, most dicotyledonous plants undergo secondary growth leading to an increased diameter of growth axes. During secondary growth initiation in shoots, a cylindrical meristem, the vascular cambium, is established by the initiation of meristematic activity in interfascicular regions, a process that ultimately leads to the formation of a continuous cylinder of vascular tissue along the shoot axis. In Arabidopsis this happens in a reduced area at the very base of the stem. In this study, a transcriptional comparison between the base and the first internode has been performed in order to asses the physiological state of both sample types and search for potential key regulators of the process. Total RNA was extracted from the base and the first internode from stems of 15cm tall Arabidopsis plants (ecotype Col-0). After RNA amplification, Cy3 or Cy5 targets were produced. Three independent biological replicates were used for each type of sample. Three hybridizations were performed, representing the three independent biological replicates, being one of them a dye swap.

Project description:In-vitro induced establishment and activity of the interfascicular cambium in Arabidopsis thaliana stems under auxin treatments. We used microarrays to detail the global programme of gene expression underlying the establishment and activity of the interfascicular cambium and identified tissue-speciffic up-regulated genes during this process. Different tissue types from in-vitro auxin treated stems were selected at successive stages of cambium establishment using laser capture microdissection for RNA extraction and hybridization on Affymetrix microarrays. We aimed to obtain genes exclussively upregulated in the interfascicular region responsible for cambium establishment and activity.

Project description:The douple mutant Arabidopsis thaliana soc1 ful, in contrast with WT, produces an interfascicular cambium and a large wood cylinder is the flowering stem. We present the RNAseq data for polyA mRNA of different developmental stages of cambium and wood formation in Arabidopsis thaliana. We sequenced 7 stages; 4 in the woody mutant soc1-6 ful-7 (herbaceous, cambium initiation, wood initiation and leaf) and 3 stages in the WT Col-0 (herbaceous, cambium and leaf). The corresponding stem anatomy is also presented in the manuscript indicating the stage of cambium development and the production of secondary xylem.
