A LOX-simulated enzyme peptide used to promote bone healing in diabetes

ABSTRACT: To investigate the potential osteogenic mechanism of PMLGH, we performed an mRNA expression profiling microarray. Male Sprague Dawley rats (10 weeks old, 320-350 g weight) were used to test the engineered hydrogels in vivo. For gene expression profiling microarray analysis, tibias were harvested at week 3.

Tryptophan zipper hydrogels: bioactive self-healing antimicrobial matrices

Project description:Natural materials can reversibly self-assemble into hierarchical networks with diverse structural and functional properties. Recreating these dynamic architectures using peptide-based synthetic materials has been an elusive goal, hindered by challenges in relating sequence to structure to function. Here we report on the de novo discovery of short peptides based on the “tryptophan zipper” (Trpzip) motif that self-assemble into hierarchically structured hydrogels. Trpzip hydrogels have a tunable modulus and show self-healing, stress relaxation, antimicrobial properties, and biocompatibility. The low yield point allows syringe extrusion with cytoprotection, and cell harvest with a flick of the wrist. Integrating a pendant cell adhesion motif promotes human intestinal organoid growth and differentiation, with polarity control and improved shape fidelity. Considering these unique characteristics, we anticipate Trpzip hydrogels will prove a versatile reagent for biotechnology and medicine.

Project description:By constructing specific lentiviral vector, vefg165 protein was overexpressed in HUVEC cells. Using light-curing bioprinting, HUVEC(vefg165+)-loaded hydrogels and HUVEC(vector)-loaded hydrogels were prepared, and were used to treat rat diabetic wounds. The regenerated skin tissues (five samples for each group) were analyzed with high-throughput proteomics.

Single cell RNA-seq of hiPSC-derived kidney organoids differentiated within self-assembling peptide hydrogels and the animal-derived matrix Matrigel

Project description:scRNA-seq was used to characterise hiPSC-derived kidney organoids differentiated within fully synthetic self-assembling peptide hydrogels of variable mechanical strengths and compare these to organoids differentiated within the animal-derived matrix, Matrigel. Organoids were matured in the respective matrices until day 24 of differentiation and 6 organoids per support matrix were then pooled and dissociated using the cold-active protease from Bacillus licheniformis. Cells were processed on the 10x Genomics Chromium platform using the Single-Cell 3’ v3.1 protocol. The NextSeq500 (Illumina) was used to sequence the libraries generated and initial processing of the data was carried out using the 10X Genomic Cell Ranger v3.1.0 pipeline.
