Project description:The changes of mRNA expression levels were analyzed in striatum, salivary glands , and peripheral leukocyte of the juvenile rat stimulated by tickling for 4 weeks as a positive stimulation. Keywords: Stress response Twenty-one-day-old male Wistar ratsM-cM-^@M-^@after weaning were used. All rats were separately maintained in the standard cages which were shielded with screens not to see each other during the study. The rats were then subjected to the following three conditions; tickling, light touch, and no stimulus. The stimulation was given to the rats once every weekday (Monday to Friday) for 4 weeks. Tickling stimulation was used as a mimic of the rough-and-tumble play of rats which induced a positive emotional response [50 kHz ultrasonic vocalization (USV)]. Light touch was a discriminative stimulation to tickling, and neither the rat with light touch nor with no stimulus showed such a response in general. Gene expression changes in striatum, salivary glands, and peripheral leukocyte of the tickled rats were comprehensively analyzed comparing with light touched and no stimulated rats using microarray techniques. Biological replicates: 4 animals each condition. One replicate per array.

Micronome profiling in 3 pairs of salivary adenoid cystic carcinoma(SACC) and the corresponding nomal salivary gland tissues

Project description:To screen miRNAs for potential NDRG2 regulation, we performed micronome profiling in 3 pairs of SACC samples and the corresponding normal salivary glands. The sequencing analysis generated approximately 1,000,000 clean reads per sample. All reads were mapped to annotated miRNAs in the miRBase database (version 22), whereas approximately 45% of the clean reads were mapped to mature miRNAs in the database. After applying a stringent filtering criterion to compare the results from SACC tumor tissue and the adjacent normal salivary glands (log2 fold change >1, FDR<0.05), we identified 176 dysregulated miRNAs.

RNA Seq Profiling of Murine Salivary Glands in the Presence or Absence of Active MCMV Infection

Project description:Recent work indicates that salivary glands are able to constitutively recruit CD8+ T cells and retain them as tissue resident memory T cells (TRM), independently of local infection, inflammation or antigen. To understand the mechanisms supporting T cell recruitment to the salivary gland, we compared T cell migration to the salivary gland in mice infected or not with murine cytomegalovirus (MCMV), a herpesvirus that infects the salivary gland and promotes the accumulation of salivary gland TRM. We found that acute MCMV infection increased rapid T cell recruitment to the salivary gland, but that equal numbers of activated CD8+ 44 T cells eventually accumulated in both infected and uninfected glands. T cell recruitment to uninfected salivary glands depended on chemokines and the integrin α4. Several chemokines were expressed in the salivary glands of both infected and uninfected mice and many of these could promote the migration of MCMV-specific T cells in vitro. MCMV infection increased expression of chemokines that interact with the receptors CXCR3 and CCR5, but neither receptor was needed for T cell recruitment to the salivary gland during MCMV infection. Unexpectedly however, the chemokine receptor CXCR3 was critical for T cell accumulation in uninfected salivary glands. Together, these data suggest that CXCR3 and the integrin α4 mediate T cell recruitment to uninfected salivary glands, but that redundant mechanisms mediate T cell recruitment after MCMV infection.

Project description:Drosophila translationally controled tumor protein (dTCTP) is important to repair double stranded DNA breaks in cell nucleus. However, besides damaged DNA loci, dTCTP is also located in interbands region of polytene chromsomes of salivary gland tissues. To address whether dTCTP is involved in transcriptional regulation process, we performed microarrays in salivary gland tissues from dTCTP mutants and w[1118] wildtype control. Salivary glands tissues were dissected and collected from 3rd instar larvae or each genotype for RNA extraction and hybridization on Affymetrix microarrays.
