Targeting phosphodiesterase 8A (PDE8A) and adipogenesis to promote hair regeneration

ABSTRACT: To maximally capture adipocytes and hair follicle keratinocytes, which are largely lost through enzymatic digestion in scRNA-seq, we performed single-nuclei RNA sequencing (Sn-RNAseq) of the developing mouse skin samples. The aim of this analysis was to identify key cell-cell interactions between adipocytes and hair follicle keratinocytes. Briefly, dorsal skin biopsies were collected from the developing C57BL/6 pups (at postnatal day 2) and 10x Genomics Sn-RNAseq was applied to identify distinct cell populations. After low quality nuclei and outliers were discarded, ~16481 nuclei were used for downstream bioinformatic analyses, including Unsupervised clustering, marker gene expression, and Cell-Chat signaling network analyses. Overall, our results reveal that adipocytes secrete growth factor to support hair follicle growth.
