Distinct miRNA profiles in human amniotic tissue and its vesicular and non-vesicular secretome

Project description:The human amniotic membrane (hAM) is a valuable tissue in regenerative medicine due to its capacity to release cell-derived bioactive factors. Among these, microRNAs (miRNAs) play a critical role in modulating gene expression. This study aimed to comprehensively characterize the miRNA landscape of native viable hAM tissue and its secretome, with a focus on distinguishing between extracellular vesicles (EV)-associated and protein-bound miRNAs. We analyzed two anatomically and functionally distinct regions of the hAM, the reflected and placental amnion. After incubation of tissue biopsies ex vivo for 72 hours, vesicular and non-vesicular components in the conditioned medium were separated by size exclusion chromatography, and small RNA sequencing was performed on tissue and secretome fractions. Our analysis identified three main clusters of miRNA expression corresponding to tissue, EV-associated, and protein-bound fractions. We observed regional differences in miRNA expression between the reflected and placental amnion and identified miRNAs selectively released into EVs or protein-bound fractions. Data of gene ontology analysis suggest distinct biological roles for miRNA depending on the sample type. This dataset provides novel insights into the spatial and functional miRNA release profile of viable hAM and contributes to a better understanding of its regenerative potential.

C/EBPδ drives key endocrine signals in human fetal membranes at parturition [ChIP-seq]

Project description:Amnion-derived prostaglandin E2 (PGE2) and cortisol are key to labor onset. Identification of the common transcription factor driving the expression of both cyclooxygenase-2 (COX-2) and 11β-hydroxysteroid dehydrogenase 1 (11β-HSD1), the key enzymes in their production, may hold the key to the treatment of pre- and post-term labor. Here, we have found that the CCAAT enhancer binding protein δ (C/EBPδ)is such a transcription factor which underlies the feed-forward induction of COX-2 and 11β-HSD1 expression by their own products PGE2 and cortisol in human amnion fibroblasts so that their production would be ensured in the amnion for the onset of labor. Moreover, the abundance of C/EBPδin amnion increases along with COX-2 and 11β-HSD1 at term and further increases at parturition. Knockout of C/EBPδ in mice delays the onset of labor further supporting the concept. In conclusion, C/EBPδmay serve as a novel pharmaceutical target in the amnion for treatment of pre- and post-term labor.

Human Initiator / Effector Gene Sets That Regulate Myometrial Contractility During Term and Preterm Labor

Project description:Distinct processes govern the transition from myometrial quiescence to activation during both term and preterm labor. We sought the specific gene sets responsible for initiating term and preterm labor, along with a core set of effector genes necessary for labor independent of gestational age and the underlying trigger. The Effector Gene Set consisted of 49 genes present in both preterm and term labor but absent from non-labor samples. 122 genes were specific to preterm labor (Preterm Initiator Set) and 229 to term labor (Term Initiator Set). The Term Initiator and the Effector Sets reflected predominantly inflammatory processes. Surprisingly, the Preterm Initiator Gene Set reflected molecular and biological events almost exclusive of inflammation. Preterm and term labor differ dramatically in their unique, initiator gene profiles, suggesting alternative pathways underlie these events. Inflammatory processes are ubiquitous to the Term Initiator and the Effector Gene Sets, supporting the idea term parturition is an inflammatory process. The absence of inflammatory processes in the Preterm Initiator Set suggests inflammation is secondary to processes triggering spontaneous preterm birth, and could explain the lack of therapeutic efficacy associated with anti inflammatory/antibiotic regimens. Experiment Overall Design: Myometrial gene expression was analyzed from samples obtained at term (n=6) or preterm (n=6) with and without labor using cDNA microarrays. Patients in preterm labor all had intra amniotic inflammation. Gene sets were generated using logical operations within a functional mapping tool (MetaCoreâ¢, GeneGo, St. Joseph, MI). Relevant gene sets were validated with quantitative real-time polymerase chain reaction.

Nucleus-localized S100A9 triggers senescence of human amnion fibroblasts as a heterochromatin buster at parturition

Project description:Aging in the amnion is considered to trigger and intensify inflammation for the initiation of labor. However, the inducers of amnion cell senescence await to be identified. Here, we unexpectedly observe that S100A9, a classical cytokines, also localizes in the nucleus of human amnion fibroblasts (hAFs), and reveal that nucleus-localized S100A9 (nS100A9) abundance in hAFs increases at parturition and that nS100A9 triggers hAFs senescence. Mechanistically, nS100A9 interacts with heterochromatin proteins, preventing them from binding and maintaining condensed heterochromatin, resulting in heterochromatin erosion, LINE1 de-repression, and type I interferon response activation via the cGAS-STING pathway. Moreover, we demonstrate that dephosphorylation of S100A9 at Thr 113 is required for its nuclear translocation. Mouse studies show that intra-amniotic injection of nS100A9 induces preterm birth along with LINE1 activation, which is blocked by reverse-transcriptase inhibitor. Together, these findings highlight a novel role for S100A9 as a heterochromatin buster that triggers hAFs aging at parturition.
