ABSTRACT: Chondrosarcoma (CHS) is a malignant bone tumour resistant to adjuvant treatment, with high-grade patients suffering an unfavorable prognosis. Heparin-binding EGF-like growth factor (HBEGF) is a member of the epidermal growth factor family, which plays a crucial role in various biological processes, including cell proliferation, differentiation, migration, and angiogenesis. HBEGF has been implicated in the development and progression of numerous cancers. We here aimed to investigate the role of HBRGF in CHS.

A logic-gated synthetic circuit coupled with microenvironmental reprogramming overcomes immune exclusion in antigen-poor chondrosarcoma

Project description:Chondrosarcoma (CHS) remains refractory to immunotherapy due to a paucity of targetable-antigen and a suppressive tumor microenvironment (TME). We developed a modular strategy integrating tumor-intrinsic genetic logic with rational TME reprogramming to overcome these barriers. Using a 57,715-element massively parallel reporter assay (MPRA), we identified CHS-selective synthetic promoters and engineered a Boolean AND-gate circuit that ensures high-fidelity expression of a surface T-cell engager (STE) exclusively within malignant cells. To further resolve the immunosuppressive TME, we identified macrophage migration inhibitory factor (MIF) as a blockade target. Single-cell RNA sequencing in human-PBMC-reconstituted CHS-xenograft models revealed that the circuit initiates T-cell activation, while MIF blockade prevents exhaustion by promoting proliferative T-cells and M1-polarized macrophages. Local administration induced a systemic abscopal effect, regressing distant, untreated tumors. Collectively, these findings provide a programmable blueprint for converting immunologically cold, antigen-poor malignancies into systemically immune-responsive states.

Targeting Fibroblast growth factor-23 (FGF23) responsive pathways in the control of renal phosphate and vitamin D metabolism

Project description:FGF23 is a bone-derived hormone that mediates renal phosphate reabsorption and 1,25(OH)2 vitamin D metabolism via its required co-receptor alpha-Klotho (KL). The functional pathways guiding this hormone’s activity in kidney have not been studied extensively, and whether using other factors with overlapping signaling profiles to produce FGF23-like responses is unclear. To map FGF23-related genes, gene array and single-cell RNA sequencing were utilized on wild type mouse kidneys. After identifying Heparin-binding EGF-like growth factor (HBEGF) as an up-regulated gene in response to FGF23 delivery, KL-null and phosphate-deficient diet fed mouse models and in vitro experiments were utilized to further test HBEGF bioactivity in kidney. Gene array demonstrated that HBEGF was significantly up-regulated following FGF23 delivery to wild type (WT) mice. Next, mice injected with HBEGF had phenotypes consistent with partial FGF23-mimetic activity including robust induction of EGR1, and increased CYP24A1 mRNAs. Single cell RNA sequencing showed overlapping HBEGF and EGFR expression in the proximal tubule (PT), and KL expression in PT and distal tubule (DT) segments. In KL-null mice devoid of canonical FGF23 signaling, HBEGF injections significantly increased EGR1 and CYP24A, and correction of basally-elevated CYP27B1 was observed. In addition, mice placed on a phosphate deficient diet to suppress FGF23 had endogenously increased CYP27B1 mRNA, which was rescued in mice receiving HBEGF. In HEK293 renal epithelial cells, HBEGF and FGF23 increased CYP24A1 mRNA. Targeting pathways known to be downstream of FGF23 in kidney may help to control renal phosphate handling in diseases of altered FGF23 bioactivity.

Multi-omics Analysis of Metabolic Characteristics and Validation of HBEGF-Mediated Regulatory Mechanisms of Metabolism during Catch-Up Growth in Small-for-Gestational-Age Infants

Project description:<p>Small-for-gestational-age (SGA) is a globally recognized public health concern. Infants born SGA may experience metabolic disturbances. This study elucidated the key regulatory factors and mechanisms underlying catch-up growth (CUG) and metabolic homeostasis in SGA infants. High-throughput targeted metabolomics were employed to compare the serum profiles of SGA and appropriate-for-gestational-age (AGA) neonates, identifying key metabolic pathways and associated regulatory signaling pathways enriched with differentially expressed metabolites. Liver tissue samples were collected from neonatal SGA and AGA rats. Subsequently, mRNA sequencing to identify and analyze differentially expressed genes. Heparin-binding epidermal growth factor-like growth factor (HBEGF)— a gene critically involved in metabolic processes—was significantly upregulated in SGA rat liver tissues. HBEGF silencing can suppress CUG in SGA rats and influence carbohydrate and lipid metabolism. Furthermore, metabolomic profiling was performed to investigate the role of HBEGF in metabolic regulation during CUG in individuals with SGA. Finally, the impact of HBEGF on the AKT/GSK-3β (protein kinase B/glycogen synthase kinase 3 beta) signaling pathway was explored in both in vivo and in vitro settings. This study confirms metabolic differences between SGA and AGA individuals are evident at early-life. HBEGF may be crucial in CUG progression in SGA by modulating metabolic processes via AKT/GSK-3β signaling pathway.</p>

BHLHE40 modulates gene transcription in breast cancer cells exposed to hypoxia and low glucose

Project description:Knockdown of BHLHE40 expression significantly reduced primary tumor growth and spontaneous lung metastasis in an orthotopic xenograft model of human breast cancer. Gene expression analysis implicated a role of BHLHE40 in hypoxia-induced exosomic secretion of Heparin Binding Epidermal Growth Factor HBEGF, which promotes cell survival and invasion. BHLHE40 induces HBEGF transcription by blocking DNA binding of HDAC1 and HDAC2.

Glutamine metabolism controls amphiregulin-facilitated chemoresistance to cisplatin in human chondrosarcoma

Project description:Chondrosarcoma is the second most common type of bone cancer. At present, the most effective clinical course of action is surgical resection. Cisplatin is the chemotherapeutic medication most widely used for the treatment of chondrosarcoma; however, its effectiveness is severely hampered by drug resistance. In the current study, we compared cisplatin-resistant chondrosarcoma SW1353 cells with their parental cells via RNA sequencing. Our analysis revealed that glutamine metabolism is highly activated in resistant cells but glucose metabolism is not. Amphiregulin (AR), a ligand of the epidermal growth factor receptor, enhances glutamine metabolism and supports cisplatin resistance in human chondrosarcoma by promoting NADPH production and inhibiting ROS accumulation. The MEK, ERK, and NrF2 signaling pathways were shown to regulate AR-mediated SLC1A5 and GLS expression as well as glutamine metabolism in cisplatin-resistant chondrosarcoma. The knockdown of AR expression in cisplatin-resistant chondrosarcoma cells was shown to reduce the expression of SLC1A5 and GLS in vivo. These results indicate that AR and glutamine metabolism are worth pursuing as therapeutic targets in dealing with cisplatin-resistant human chondrosarcoma.
